Association between the CYBA and NOX4 genes of NADPH oxidase and its relationship with metabolic syndrome in non-alcoholic fatty liver disease in Brazilian population

Background: Oxidative stress has been implicated in the progression of severe forms of non-alcoholic fatty liver disease(NAFLD). NADPH oxidase produces reactive oxygen species. In the present study, we investigated for the first time two single nucleotide polymorphisms(SNPs) in the regulatory region of genes encoding NADPH oxidase 4(NOX4) and p22 phox(CYBA) in ***: A total of 207 biopsy-proven NAFLD patients [simple steatosis(n = 27); nonalcoholic steatohepatitis(NASH)(n = 180)] were evaluated. Genomic DNA was extracted from peripheral blood cells, and polymorphisms in CYBA(unregistered) and NOX4(rs3017887) were determined by direct sequencing of ***: Associations of CYBA-675 T/A with high-density lipoprotein(HDL)(TT vs TA vs AA; P erides(TGL)(TT vs XA; P ere observed only in NASH patients. For polymorphisms in the NOX4 gene, NOX4(rs3017887) CA + AA genotypes was significant associated with alanine aminotransferase(ALT)(CA + AA vs CC; P = 0.02). However, there was no association of SNPs in the CYBA and NOX4 genes encoding the NADPH oxidase system proteins and the presence of NASH. Regarding the clinical results, it was observed that the most advanced degrees of fibrosis occurred in patients diagnosed with type 2 diabetes mellitus(66.9% vs 37.5%, P e who were more obese(32.2 vs 29.0 kg/m2,P erum glucose and insulin levels increased significantly in the presence of ***: There were associations between the presence of the allele A in the NOX4 SNP and a higher concentration of ALT in the NAFLD population; between the presence of the AA genotype in the polymorphism of the CYBA-675 T/A CYBA gene and a higher level of TGL and lower HDL in NASH patients. The presence of metabolic syndrome was associated with advanced degrees of fibrosis in NAFLD patients.

Injuries to the spinal cord result in permanent disabilities that limit daily life *** main reasons for these poor outcomes are the limited regenerative capacity of central neurons and the inhibitory milieu that is established upon traumatic *** decades of research,there is still no efficient treatment for spinal cord *** strategies are tested in preclinical studies that focus on ameliorating the functional outcomes after spinal cord *** these,molecular compounds are currently being used for neurological recovery,with promising *** molecules target the axon collapsed growth cone,the inhibitory microenvironment,the survival of neurons and glial cells,and the re-establishment of lost *** this review we focused on molecules that are being used,either in preclinical or clinical studies,to treat spinal cord injuries,such as drugs,growth and neurotrophic factors,enzymes,and *** mechanisms of action of these molecules are discussed,considering traumatic spinal cord injury in rodents and humans.

AIM To determine the prevalence of epstein-barr virus(eb V)-associated gastric carcinomas in the North Region of Portugal and to study its clinicopathological characteristics. MeTHODS We have performed a retrospective study including a total of 179 consecutive patients with gastric cancer(GC) submitted to gastrectomy during 2011 at the Portuguese Oncology Institute of Porto. Clinical and pathological data was collected from individual clinical records and inserted on a database with unique codification. Tumour tissues were collected from the institutional tumour bank. eb V was detected by in situ hybridization for the detection of eb V-encoded small RNAs(ebe Rs) and eb V latent proteins(LMP1 and LMP2 A) were detected by *** The analysis showed that eb V-associated gastric carcinomas(eb Va GC) represents 8.4%(15/179) of all GC cases, with a significant differential distribution among histological types(P edullary carcinomas, 100%(1/1) of adenosquamous carcinoma, 8.7%(8/92) of tubular adenocarcinomas, 8.0%(2/25) of mixed carcinomas and 2%(1/51) in poorly cohesive carcinomas. The analysis revealed a higher predominance of eb Va GC in the upper third and middle(cardia, fundus and body) of the stomach(P = 0.041), a significant lower number of regional lymph nodes invasion(P = 0.025) and a tendency for better prognosis(P = 0.222). eb V latent protein expression revealed that all eb Va GC cases were LMP1-negative, nevertheless 6 cases(40%) expressed LPM2 A, which reveals that these cases show a distinct eb V-Latency profile(latency II-like).CONCLUSION eb Va GC represents 8.4% of all GC in the North Region of Portugal. The eb V-infected patients have specific clinic-pathological features that should be further explored to develop new strategies of management and treatment.

Pompe disease (PD) is a rare inborn error of metabolism due to an abnormal acid alpha-glucosidase (GAA) activity that comprises glycogen breakdown mainly in the lysosomes. Since the introduction of enzyme replacement therapy (eRT), with recombinant human GAA for the early onset PD patient, a relevant field of clinical research due to the benefits regarding survival rate has been widely documented worldwide. Objective: To describe the clinical characteristics and the eRT effects in a series of Brazilian patients with infantile onset PD (IOPD) under eRT. Methods: Brazilian patients diagnosed with IOPD under eRT were recruited through their physicians participating in the International Pompe Disease Registry from 2009 to 2017. Data were collected by an online survey. Results: 10 IOPD patients were identified through the survey with a death rate of 30% and technology dependency rate reported as 80% (motor, respiratory or nutritional fields) of the patients. After the third year of eRT, motor disabilities were lost in 50% of ambulated patients. The overall characteristics were similar to international studies. Conclusion: Despite eRT benefits in cardiac involvement, motor disabilities seem to be much more compromised in IOPD patients, with high technology dependence, especially after three years of age.

A standard ultramarine pigment was used to produce phase change material composites, by adsorbing n-hexadecane paraffin around the pigment surface with the aim of obtaining a pigment providing thermal storage capacity apart from color. Vacuum impregnation method was employed optimizing the process variables to maximize the latent heat of the hexadecane/pigment composite. In addition to the process optimization, the stability of the composite having the maximum latent heat was investigated. The hexadecane/pigment composite providing the highest latent heat has a Latent heat of fusion of 44 J/g (around a 20%wt. hexadecane adsorbed in the pigment). Durability of the material was tested by thermo-diffractometric measurements. The results indicate slow reduction of the area intensity up to 6.5% after the 100 cycles.

Characterization and Biological Activities of Protein-Bound Polysaccharides Produced by Cultures of Pleurotus ostreatus

Several species of mushrooms, as Pleurotus ostreatus, have been valued as edible and medicinal resources. These mushrooms may be an important source of polysaccharides with medicinal properties as antioxidant, antitumoral, antimicrobial and immunological properties. The aim of this work was to produce and to evaluate the biological properties of protein-bound polysaccharide complexes, extra intracellular （e-PPS and I-PPS）, extracted from P. ostreatus cultures, using agricultural sunflower wastes as carbon source. Three main compounds in the e-PPS and four main compounds in the I-PPS were identified by SeC-UV-RI-HPLC. These complexes of P. ostreatus present no toxicity in Artemia salina cultures, after 24 h of incubation. Antioxidant properties of the complexes were evaluated by radical scavenging activity using DPPH method and lipid peroxidation inhibition capacity, determined by erytbsocytes hemolysis. Additionally, e-PPS and I-PPS extracts revealed capacity to mimetize superoxide dismutase and catalase enzymatic activities. The hepatoprotector effect of e-PPS extracts in Wistar rats was evaluated by AST, ALT, ALP and y-GT activities, showing capacity to reduce the liver damage induced by ethanol-administration. This hepatoprotective effect is equivalent to that observed by silymarin, a standard drug. Our results suggests that the extracts of e-PPS and I-PPS produced by P. ostreatus cultures, using agricultural sunflower wastes as main carbon source, can be used as an important source of bioactive compounds with potential medicinal value.

Single Layer Centrifugation with Androcoll-e^(TM) improved progressive motility and percentage of live spermatozoa with intact acrosome of chilled stallion semen but did not have an effect on DNA integrity

A significant number of stallions produce low quality ejaculates with high sensibility to chilling. Single Layer Centrifugation (SLC) with Andro-coll-eTM has been presented as an efficient method of selecting good quality spermatozoa. In the current study, changes in sperm quality (motility, viability, acrosome integrity and DNA damage) occurring during storage at 5?C for a maximum of 72 h, were investigated. For that, one ejaculate from 12 stallions was split in two aliquots: control and SLC-selected. Both aliquots were chilled and stored at 5?C and spermatozoa were evaluated for motility, viability and acrosome integrity at 24, 48 and 72 h post collection. DNA damage was evaluated at 48 h post collection using the comet assay. In the SLC-selected aliquots, there was a significant improvement in terms of progressive motility (0 h: P = 0.005;24 h: P 0.05). SLC with Androcoll-eTM improved semen quality prolonging sperm longevity of chilled semen (P = 0.012). This positive effect was more evident in ejaculates most sensitive to chilling that had a sharp decrease in motility in the first 24 h of refrigeration and for all ejaculates at 72 h post-chilling. Therefore, this method reveals to be a useful technique for selecting spermatozoa and maintain sperm quality during storage.

effect of oxygen and nitrogen functionalization on the physical and electronic structure of graphene

Covalent functionalization of graphene offers opportunities for tailoring its properties and is an unavoidable consequence of some graphene synthesis techniques. However, the changes induced by the functionalization are not well understood. By using atomic sources to control the extent of the oxygen and nitrogen functionalization, we studied the evolution in the structure and properties at the atomic scale. Atomic oxygen reversibly introduces epoxide groups whilst, under similar conditions, atomic nitrogen irreversibly creates diverse functionalities including substitutional, pyridinic, and pyrrolic nitrogen. Atomic oxygen leaves the Fermi energy at the Dirac point （i.e., undoped）, whilst atomic nitrogen results in a net n-doping; however, the experimental results are consistent with the dominant electronic effect for both being a transition from delocalized to localized states, and hence the loss of the signature electronic structure of graphene.
