Combined Use of NMR, LC-ESI-MS and Antifungal Tests for Rapid detection of Bioactive Lipopeptides Produced by Bacillus

The science and technology interact with the art in several ways. Biotechnological coupled with analytical approaches can play an important role in protecting and preserving cultural heritage for future generations. Many microorganisms influenced by environmental conditions are the main responsible for biological contamination in built heritage. Biocides based on chemical compounds have been used to mitigate this problem. Thus, it is vitally important to develop proper remediation actions based on environmentally innocuous alternative. Bacillus specie is emerging as an optimistic alternative for built heritage treatment due to their capacity to produce secondary metabolites with antagonistic activities against many fungal pathogens. Therefore, the intent of this work was to access a rapid evaluation of antifungal potential of bioactive metabolites produced by Bacillus strains and simultaneously their characterization using spectroscopic (NMR) and chromatographic techniques (LC-ESI-MS). The high antifungal activity obtained for Bacillus sp. active compounds produced in this study confirms the great potential to suppress biodeteriogenic fungi growth on historical artworks. Additionally, the proposed methodology allowed to access bioactive metabolites produced without need of the laborious total previous isolation and could be used as a viable alternative to be employed for screening and production of new green biocides.

Influence of Photoperiod over Morphometric and Hematological Parameters of Juvenile Piracanjubas （Brycon orbygnianus）

The aim of this study was to evaluate morphometric and hematological parameters of juveniles Piracanjubas treated with two distinct photoperiod. Twenty nine fish with 100 g, in a factorial design were performed in 18 h light：6 h dark （18L：6D） and 6L：lSD two different photoperiod, and morphometric and hematological parameters were evaluated at 45 d and 90 d after photoperiod treatment. Benzocaine 1% was used for euthanasia. Fish were weighed and measured, so as visceral fat, liver and gonads. Hematocrit, total count of white cells, lymphocytes and neutrophils were counted. Results showed that there was no significant relationship between bodyweight and standard length. Significant interaction （P = 0.00018） had been found between the fat index （FI） and gonado-somatic index （GSI） and the photoperiod and time. At 90 d, the fish kept in 18L：6D had a lower rate of fat than the fish kept in 6L：18D （P 〈 0.05）. The GSI was higher in the group 6L.＇18D compared with the 18L：6D （P 〈 0.005）. There was no interaction between photoperiod and period of treatment for total leukocyte count, however with 90 d, the total leukocyte count was decreased （P 〈 0.05）. Moreover, decrease of the hematocrit from 58.1% to 46.9% and an increase of lymphocytes from 31.5% to 43.5% had been found in fish kept in 18L：6D after 90 d （P 〈 0.05）. Fish kept in 6L：ISD presented an increase in segmented neutrophils and lymphocytes （P 〈 0.05）. It is concluded that photoperiod maintained at 6L： 18D for Piracanjubas permits a greater gonadal development and an amount of immune cells over the photoperiod maintained in 18L： 6D.

The Phytochemical Profile and Photoprotective Potential of Bark and Leaves of Erythrina velutina Willd.—A Medicinal Species from the Caatinga Region of Brazil

The use of medicinal plants as therapeutic and economic resources has been on the rise in recent years. In Brazil, however, increasing doubt is being cast on the quality of these products, owing to the prevalence of adulteration and fraud. Solar radiation can cause serious damage to human skin, as a result, mostly, of ultraviolet light, which is a cause of skin cancer. Photoprotective substances are capable of absorbing, reflecting, or refracting ultraviolet radiation and thus protecting skin from exposure to sunlight. The present study aimed to characterize samples and examine the phytochemical profile and photoprotective potential of bark and leaves of Erythrina velutina Willd. The samples underwent five extraction methods using 80% ethanol. The phenolic content was measured using spectrophotometry. Antioxidant activity was examined using the DPPH and the photoprotective properties of the plant extracts were assessed using the method developed by Mansur. There was a quantitative difference in some groups of metabolites, with higher levels of tannin in the bark and of flavonoids in the leaves. The latter showed greater DPPH free radical scavenging capacity than the bark, although higher levels of SPF were obtained from the bark, with no statistically significant differences between methods. The results indicate that Erythrina velutina Willd. has potential as a photoprotector.

Difficulties Faced by Physicians in the Diagnosis of Endometriosis in the Brazilian Public Health System

Endometriosis is a clinical and recurrent condition characterized by the presence of functional endometrial tissue outside the uterine cavity and myometrium. It is estimated that 6% to 10% of women of reproductive age, 50% to 60% of adolescents and adults with pelvic pain, and up to 50% of women with infertility are pregnant from the condition. However, this proportion tends to be even greater due to the difficulty in diagnosing it: because it presents different symptoms, lack of information from health professionals, and a high-cost diagnostic test. Another aggravating factor is the average time from the onset of symptoms to the medical diagnosis, which even today ranges from five to ten years;this delay has consequences for disease progression, as it prevents early treatment, which is important for improving pain levels and physical and psychological well-being. Aware of these mishaps, it is therefore important to assess the difficulties faced by doctors in the diagnosis of endometriosis in the public health system, in Brazil, the Unified Health System (SUS). With this purpose, a cross sectional was carried out, developed in the virtual environment using traffic in Google forms, with the difficulties quantified in the Likert scale. Participating physicians pointed out a medium difficulty for patients with endometriosis to access the consultation (58%) and moderate difficulty (52%) to perform an ultrasound examination;also signaled great difficulty in performing magnetic resonance imaging (77%) and laparotomy (61%) in these patients. It is hoped that knowledge about the difficulties faced by physicians in the diagnosis of endometriosis in the SUS will give these physicians a voice and corroborate, even if minimally, so that there is more research that provokes the search for these difficulties, in order to improving the quality of life of these women.

The volatile constituents of the essential oils from leaves from the weed plants Parthenium hysterophorus and Ambrosia polystachya were identified and quantified by GLC-MS and GLC. Allelopathic activities were determined by methods that evaluated the volatile effects and the direct application of these essential oils on the seed germination and seedling vigor of lettuce. We identified 27 compounds in the essential oil from P. hysterophorus, and the main constituents were germacrene-D (35.9%), trans-β-ocimene (8.5%) and β-myrcene (7.6%). In the essential oil from A. polystachya, 40 constituents were identified and the principal compounds were germacrene-D (29.3%), trans-β-ocimene (13.6%) and β-caryophyllene (9.8%). In both methods, the essential oil from A. polystachya presented a greater potential for reducing seed germination and seedling vigor in lettuce than the essential oil from P. hysterophorus, and this activity might be attributed to its higher content of monoterpenes.

Comparative Morpho-Biochemical Responses of Sunflower Lines Sensitive and Tolerant to Water Stress

Water stress is likely the most important factor that adversely affects plant growth and development. In our study two inbred lines of sunflower (Helianthus annuus L.) with contrasting behavior to water deficit, the inbred lines B59, sensitive, and B71, tolerant, were subjected to water stress and compared in terms of growth parameters, relative water content, photosynthetic pigments and osmo-compatibles contents. Mannitol treatment generated a moderate water stress which resulted in a relative water content decline in both lines. In aerial part of B59 and B71 stressed-seedlings, a dramatic reduction of osmotic potential was observed, although in B71 the osmotic potential was 1.8-fold lower than in B59. The B71 line recovered the osmotic potential to control values after 8 d of re-watering. Several morphological parameters were altered by the water stress. Shoot and root relative fresh weight decreased in both lines under water stress, although B71 showed a minor drop. However, the root was less affected than the shoot in stress condition and recovery period. Water stress was affected in a greater proportion shoot dry weight (DW) than root DW in B59 and B71 lines. Nevertheless, a significant increase in root DW/shoot DW ratio was observed in stressed seedlings. A higher reduction in the individual leaf area was observed in B71 line under stress. The basal chlorophyll content was higher in the tolerant line B71 than in the sensitive B59, and carotenoids showed a high content in B71 control and stressed seedlings. Under water stress, the aerial and root proline content increased in the tolerant B71 line. Glycinebetaine decreased significantly in the aerial part of both lines in response to water stress indicating that this compound is not the main osmo-regulator. In conclusion, B71 has suitable mechanisms to enable it to respond more effectively to moderate water stress.

determining the Soil Erodibility for an Experimental Basin in the Semi-Arid Region Using Geoprocessing

Erosion is the natural process which has the greatest environmental impact, and is the principal trigger for desertification around the globe. The main model used to estimate soil loss by erosion is the Universal Soil Loss Equation (USLE), which unites the major factors that influence erosion into one equation. The soil erodibility factor (K) is the component of this equation that represents soil physics, and is defined as the inherent capacity of the soil to withstand disintegration of its particles and their subsequent transport. The use of geostatistics is seen as an alternative in spatializing this variable from sampled to non-sampled points. The aim of this study therefore, was to determine the soil erodibility factor for an experimental basin in the semi-arid region of Brazil, in addition to generating the soil erodibility map using geostatistics. Disturbed and undisturbed soil samples were collected from 35 points, and laboratory samples were processed to determine the granulometry, permeability and organic matter of the soil, data which are used to determine the K-factor. Kriging was performed to spatialize the study variable, when spherical, exponential and Gaussian semivariograms were tested for generation of the soil erodibility map, these being evaluated by their respective deviations resulting from cross-validation. The mean value of K for the Haplic Luvisol was 0.0328 ton·ha·h/ha·MJ·mm;for the eutrophic Red-Yellow Argisol it was 0.0258 ton·ha·h/ha·MJ·mm;and for the Fluvic Neosol, it was 0.0424 ton·ha·h/ha·MJ·mm. The experimental basin is classified as highly erodible. The semivariogram that presented the best fit for generating the soil erodibility map of the study area was Gaussian.

Standardization of Sampling for Isolation of Exosome-Like Small-Extracellular Vesicles from Peripheral Blood from Reproductive-Aged Women

Exosome-like small-extracellular vesicles (sEVs) are extracellular vesicles that act in intercellular communication and are involved in several biologic and pathologic processes. While sEVs increase the stability of their cargo molecules, there is still a need for standardization of sampling and isolation of these microvesicles. We aimed to determine the best sampling method for isolation of sEVs from peripheral blood from reproductive-aged women. Material and Methods: We included samples of plasma from our biobank collected in 2014 by venipuncture in heparin tubes and stored at -80°C. We also included blood samples collected in heparin tubes and Ethylenediamine tetraacetic acid (EDTA) tubes and stored at -80°C for one to two weeks prior processing. All blood samples were collected from the same nine reproductive-aged female volunteers. sEVs were isolated from plasma by ultracentrifugation and filtration and indirectly quantified using Pierce BCA Protein Assay kit. Transmission electron microscopy (TEM) and Nano Tracking analysis (NTA) were performed to confirm the isolation of sEVs. Results and Discussion: TEM and NTA confirmed the isolation of sEVs. Protein concentration of short-time stored heparin samples was not statistically different from long-time stored heparin samples (1847.2 ± 651.4 vs. 2363.2 ± 1025.1, p = 0.14). There was no difference between heparin and EDTA plasma samples recently collected (2363.2 ± 1025.1 vs. 2044.8 ± 653.2, p = 0.44). In conclusion, blood samples may be collected using heparin or EDTA for isolation of sEVs. Long-time stored plasma samples maintain sEVs integrity and may be used, especially in comparative studies.

Objective: To evaluate the association of socio-demographic and clinical characteristics, and risk factors with occurrence of pressure ulcers in institutionalized elderly people. Method: This cross-sectional, analytical study, in quantitative approach, performed from the database analysis linked to the project “Pressure Ulcer in institutionalized elderly people: Association of incidence with the risk factors to functional and nutritional assessment” developed in six long-stay institutions for the elderly people in João Pessoa. Data were analyzed using the Statistical Package for Social Sciences (SPSS) version 20.0. Associations were made through chi square test and Odds Ratio. Results: The clinical conditions of significant associations with the occurrence of pressure ulcers were the variables neurological disorders (p = 0.011) and visual impairment (p = 0.005). As for risk factors, the most important was fecal incontinence (p Conclusion: The analysis of the associations pointed out problems that require intervention in order to prevent health implications of the elderly people and to minimize the risk and occurrence of this injury.
