AIM： To investigate and compare the hepatogenic transdifferentiation of adipose tissue-derived stem cells （ADSC） and bone mar.ow-derived mesenchymal stem cells （BMSC） in vitro. Transdifferentiation of BMSC into hepatic cells in vivo has been described. Adipose tissue represents an accessible source of ADSC, with similar characteristics to BMSC. METHODS： BMSCs were obtained from patients undergoing total hip arthroplasty and ADSC from human adipose tissue obtained from lipectomy. Cells were grown in medium containing 15% human serum. Cultures were serum deprived for 2 d before cultivating under similar pro-hepatogenic conditions to those of liver development using a 2-step protocol with sequential addition of growth factors, cytokines and hormones. Hepatic differentiation was RT-PCR-assessed and liver-mar.er genes were immunohistochemically ***： BMSC and ADSC exhibited a fibroblastic morphology that changed to a polygonal shape when cells differentiated. Expression of stem cell mar.er Thyl decreased in differentiated ADSC and BMSC. However, the expression of the hepatic mar.ers, albumin and CYPs increased to a similar extent in differentiated BMSC and ADSC. Hepatic gene activation could be attributed to increased liver-enriched transcription factors （C/EBPβ and HNF4α）, as demonstrated by adenoviral expression ***： Mesenchymal stem cells can be induced to hepatogenic transdifferentiation in vitro. ADSCs have a similar hepatogenic differentiation potential to BMSC, but a longer culture period and higher proliferation capacity. Therefore, adipose tissue may be an ideal source of large amounts of autologous stem cells, and may become an alternative for hepatocyte regeneration, liver cell transplantation or preclinical drug testing.

Clk/STY is a LAMMER protein kinase capable to phosphorylate serine/arginine-rich (SR) proteins that modulate pre-mRNA splicing. Clk/STY alternative splicing generates transcripts encoding a full-length kinase and a truncated catalyti-cally inactive protein. Here we showed that clk/STY, as well as other members of the family (e.g. clk2, clk3 and clk4),are up-regulated during HMBA-induced erythroleukemia cell differentiation. mRNAs coding for the full-length and thetruncated forms were responsible for the overall increased expression. In clk/STY, however, a switch was observed forthe ratio of the two alternative spliced products. In undifferentiated cells the full-length transcript was more abundantwhereas the transcript encoding for the truncated form predominated at latter stages of differentiation. Surprisingly,overexpression of clk/STY did not alter the splicing switch upon differentiation in MEL cells. These results suggest thatclk/STY might contribute to control erythroid differentiation by a mechanism that implicates a balance between thesetwo isoforms.

Overlap of reflux and eosinophilic esophagitis in two patients requiring different therapies:A review of the literature

Eosinophilic esophagitis (EE) and gastroesophageal reflux disease (GERD) have overlapping clinical, manometric, endoscopic and histopathologic features. The diagnosis of EE is nowadays based upon the presence of 15 or more eosinophils per high power field (eo/HPF) in esophageal biopsies. We report the cases of two young males suffering from dysphagia and recurrent food impaction with reflux esophagitis and more than 20 eo/HPF in upper-mid esophagus biopsies, both of which became asymptomatic on proton pump inhibitor (PPI) therapy. The first patient also achieved a histologic response, while EE remained in the other patient after effective PPI treatment, as shown by 24-h esophageal pH monitoring. Topical steroid therapy combined with PPI led to complete remission in this latter patient. GERD and EE may be undistinguishable, even by histology, so diagnosis of EE should only be established after a careful correlation of clinical, endoscopic and pathologic data obtained under vigorous acid suppression. These diagnostic difficulties are maximal when both diseases overlap. Limited data are available about this topic, and the interaction between EE and GERD is a matter of debate. In this setting, upper-mid esophagus step biopsies and esophageal pH monitoring of patients on PPI therapy are pivotal to evaluate the role of each disease. A PPI trial is mandatory in patients with a histopathologic diagnosis of EE; in those unresponsive to PPI treatment, EE should be suggested. However, a clinical response to PPI may not rule out quiescent EE, as shown in this report.

AIM： To evaluate the relationship between leptin and systemic inflammation in acute pancreatitis. METHODS： Consecutive patients with acute pancreatitis were included. Body mass index and serum samples were obtained at admission. Leptin, TNF-α, IL-6, -8 and -10 levels were determined by ELISA. Severity was defined according to Atlanta criteria. RESULTS： Fifty-two （29 females） patients were studied. Overall body mass index was similar between mild and severe cases, although women with severe pancreatitis had lower body mass index （P = 0.04） and men showed higher body mass index （P = 0.05）. No difference was found in leptin levels regarding the severity of pancreatitis, but higher levels tended to appear in male patients with increased body mass index and severe pancreatitis （P = 0.1）. A multivariate analysis showed no association between leptin levels and severity. The strongest cytokine associated with severity was IL-6. Correlations of leptin with another cytokines only showed a trend for IL-8 （P = 0.058）. CONCLUSION： High body mass index was associated with severity only in males, which may be related to android fat distribution. Serum leptin seems not to play a role on the systemic inflammatory response in acute pancreatitis and its association with severe outcome in males might represent a mar.er of increased adiposity.

SCADA Data-Based Support Vector Machine for False Alarm Identification for Wind Turbine Management

Maintenance operations have a critical influence on power gen-eration by wind turbines(WT).Advanced algorithms must analyze large volume of data from condition monitoring systems(CMS)to determine the actual working conditions and avoid false *** paper proposes different support vector machine(SVM)algorithms for the prediction and detection of false alarms.K-Fold cross-validation(CV)is applied to evaluate the classification reliability of these *** Control and Data Acquisition(SCADA)data from an operating WT are applied to test the proposed *** results from the quadratic SVM showed an accuracy rate of 98.6%.Misclassifications from the confusion matrix,alarm log and maintenance records are analyzed to obtain quantitative information and determine if it is a false *** classifier reduces the number of false alarms called misclassifications by 25%.These results demonstrate that the proposed approach presents high reliability and accuracy in false alarm identification.

Comparation of the eutectic mixture of lidocaine/prilocain versus benzocaine gel in children

Objective: To compare the anesthetic effect of a non commercial eutectic mixture of 4% lidocaine/ prilocaine (PLO 4%) and 20% benzocaine gel (Hurricaine?), as topical anesthetic, prior to inferior alveolar nerve block and buccal infiltration anesthe-sia in 5-12 year old children. Study design: Infiltrative anesthesia was applied in 50 children, divided in two groups (n = 25) using PLO 4% and Hurricaine? as topical anesthesia prior to infiltration. Physical reac-tions were registered using the Sound-Eyes- Motor Scale. Physiological changes expressed by ar-terial pressure and heart rate. Subjective pain re-sponse was scored on a Facial Image Scale. Physical physiological and subjective response was related to the type of topical anesthetic, age and sex using χ2 and Mann-Whitney U test. Results: Physical responses to puncture were similar and localized in the state of comfort with both anesthetics. Girls showed more ocular response than boys. Subjective pain perception and physiological reactions showed no anesthetic- or sex-related differences, except for heart rate before and after the procedure which was significantly higher in girls. Conclusions: PLO 4% showed the same capacity as Hurricaine? in reducing pain response to needle puncture. Girls expressed more needle puncture-related pain than boys. The young children showed most prior comfort and less discomfort to the puncture than older children.

In-depth analysis of soil management and farmers’perceptions of related risks in two olive grove areas in southern Spain

This manuscript presents a questionnaire-based study aimed to provide a detailed analysis on the different soil management carried out by olive farmers in two representative olive-growing areas in southern Spain(Cordoba and Estepa),their perceptions on cover crop use and the possible influence of the different types of farms and farmers’typologies on these *** results show a relatively large variability of soil management,with fourteen options,as a result of a combination of different alternatives for bare soil and cover crops with the use or not of pruning residues,but with a great similarity between both *** results indicate a high adoption of soil conservation measures in the two study areas,with 63%of farmers using cover crops and 80%a mulch of pruning residues,higher than that reported in previous studies in Southern Spain,and a trend of lower use of these techniques by less experienced and younger *** high penetration of soil conservation measures resulted in a significant reduction of soil erosion risk,as indicated by the relatively low values for the cover and management factor(C)of RUSLE,also calculated and presented in this study,but also the possibility of focusing further efforts on fanners with less *** results indicate the persistence of a minor,but relevant,percentage of farmers using bare soil management(37%)and no mulching(20%),with a moderate concern on the impact of soil erosion on soil degradation and provision of ecosystem *** suggests the need to concentrate efforts also on this cluster of farmers to enhance the success of what seems to be a remar.able expansion of the use of soil conservation measures in recent decades in Southern Spain,but also in similar areas in the Mediterranean basin.

Colorectal cancer(CRC)is the second leading cause of cancer-related death *** five-year survival rate of CRC patients depends on the stage at diagnosis,being higher than 80%when CRC is diagnosed in the early stages but lower than 10%when CRC is diagnosed in advanced *** against specific CRC autoantigens(tumor-associated antigens(TAAs))in the sera of patients have been widely demonstrated to aid in early ***,we herein aim to identify autoantigens target of autoantibodies specific to CRC that possess a significant ability to discriminate between CRC patients and healthy individuals by means of liquid *** that end,we examined the protein content of the exosomes released by five CRC cell lines and tissue samples from CRC patients by means of immunoprecipitation coupled with mass spectrometry analysis.A total of 103 proteins were identified as potential autoantigens specific to *** bioinformatics and meta-analysis,we selected 15 proteins that are more likely to be actual CRC autoantigens in order to evaluate their role in CRC prognosis by Western blot(WB)and immunohistochemistry(IHC).We found dysregulation at the protein level for 11 of these proteins in both tissue and plasma exosome samples from patients,along with an association of nine of these proteins with CRC *** validation,all but one showed a statistically significant high diagnostic ability to distinguish CRC patients and individuals with premalignant lesions from healthy individuals,either by luminescence Halotag-based beads,or by a multiplexed biosensing platform involving the use of magnetic microcarriers as solid support modified with covalently immobilized Halotag fusion proteins constructed for CRC *** together,our results highlight the usefulness of the approach defined here to identify the TAAs specific to chronic diseases;they also demonstrate that the measurement of autoantibody levels in plasma against the TAAs identified here

Whole Chia Flour as Yield Enhancer, Potential Antioxidant and Input of n-3 Fatty Acid in a Meat Product

In this paper, the effect of the addition of whole chia flour and water at cooked fish burgers on yield, water and fat retention, n-3 fatty acids content and oxidative stability were studied. The results showed that 94% of yield, 92% of water retention and 97% of fat retention were achieved with the addition of 5.92 g of whole chia flour and 15 g of water per 100 g of burger. The n-3 fatty content increased from 704.57 ± 21.66 mg to 1551.71 ± 47.71 mg/100g of cooked burger when the chia flour was added, enhancing nutritional quality indices of lipid. A good oxidative stability, with minimum formation of conjugated dienes, hydroperoxides and thiobarbituric acid reactive substances during 30 days frozen storage was obtained too, probably due to the presence of polyphenolic compounds with antiradical activity and reducing power in the fibrous fraction of the flour chia. The results from the present study highlight remar.able technological applications of whole chia flour as food ingredient in the design of healthier fish meat commodities.
