Tissue innervation is a complex process controlled by the expression profile of signaling molecules secreted by tissue-resident cells that dictate the growth and guidance of *** innervation is part of the neuronal network of the bone tissue with a defined spatiotemporal occurrence during bone ***,the current understanding of the mechanisms regulating the map of sensory innervation in the bone tissue is still ***,we demonstrated that differentiation of human mesenchymal stem cells to osteoblasts leads to a marked impairment of their ability to promote axonal growth,evidenced under sensory neurons and osteoblastic-lineage cells *** mechanisms by which osteoblast lineage cells provide this nonpermissive environment for axons include paracrine-induced repulsion and loss of neurotrophic factors *** identified a drastic reduction of NGF and BDNF production and stimulation of Sema3A,Wnt4;and Shh expression culminating at late stage of OB *** noted a correlation between Shh expression profile,OB differentiation stages,and OB-mediated axonal *** of Shh activity and signaling reversed the repulsive action of osteoblasts on sensory ***,to strengthen our model,we localized the expression of Shh by osteoblasts in bone ***,our findings provide evidence that the signaling profile associated with osteoblast phenotype differentiating program can regulate the patterning of sensory innervation,and highlight osteoblast-derived Shh as an essential player in this cue-induced regulation.

Duloxetine(DUl), an antidepressant drug, has been detected in surface water and wastewater effluents, however, there is little information on the formation of its transformation products(TPs). In this work, hydrolysis, photodegradation(UV irradiation) and chlorination experiments were performed on spiked distillated water, under controlled experimental conditions to simulate abiotic processes that can occur in the environment and wastewater treatment plants(WWTPs). Eleven TPs, nine from reaction with UV light and two from chlorine contact, were formed and detected by ultra-high performance liquid chromatography coupled to quadrupole time-of-flight mass spectrometry, and nine of them had their chemical structures elucidated upon analyses of their fragmentation patterns in MS/MS spectra. The formation and degradation of the TPs were observed. The parent compound was completely degraded after30 min in photodegradation and after 24 hr in chlorination. Almost all TPs were completely degraded in the experiments. The ecotoxicity and mutagenicity of the TPs were predicted based on several in silico models and it was found that a few of these products presented more ecotoxicity than DUl itself and six TPs showed positive mutagenicity. Finally, wastewater samples were analyzed and DUl and one TP, possibly formed by chlorination process, were detected in the effluent, which showed that WWTP not only did not remove DUl, but also formed a TP.

Bacterial ability to colonize the rhizosphere of plants in arsenic（As） contaminated soils is highly important for symbiotic and free-living plant growth-promoting rhizobacteria（PGPR）used as inoculants, since they can contribute to enhance plant As tolerance and limit metalloid uptake by plants. The aim of this work was to study the effect of As on growth,exopolysaccharide（EPS） production, biofilm formation and motility of two strains used as soybean inoculants, Bradyrhizobium japonicum E109 and Azospirillum brasilense Az39. The metabolism of arsenate（As（V）） and arsenite（As（III）） and their removal and/or possible accumulation were also evaluated. The behavior of both bacteria under As treatment was compared and discussed in relation to their potential for colonizing plant rhizosphere with high content of the metalloid. B. japonicum E109 growth was reduced with As（III）concentration from 10 μM while A. brasilense Az39 showed a reduction of growth with As（III） from 500 μM. EPS and biofilm production increased significantly under 25 μM As（III）for both strains. Moreover, this was more notorious for Azospirillum under 500 μM As（III）,where motility was seriously affected. Both bacterial strains showed a similar ability to reduce As（V）. However, Azospirillum was able to oxidize more As（III）（around 53%） than Bradyrhizobium（17%）. In addition, both strains accumulated As in cell biomass. The behavior of Azospirillum under As treatments suggests that this strain would be able to colonize efficiently As contaminated soils. In this way, inoculation with A. brasilense Az39 would positively contribute to promoting growth of different plant species under As treatment.

Background: Oxidative stress has been implicated in the progression of severe forms of non-alcoholic fatty liver disease(NAFlD). NADPH oxidase produces reactive oxygen species. In the present study, we investigated for the first time two single nucleotide polymorphisms(SNPs) in the regulatory region of genes encoding NADPH oxidase 4(NOX4) and p22 phox(CYBA) in ***: A total of 207 biopsy-proven NAFlD patients [simple steatosis(n = 27); nonalcoholic steatohepatitis(NASH)(n = 180)] were evaluated. Genomic DNA was extracted from peripheral blood cells, and polymorphisms in CYBA(unregistered) and NOX4(rs3017887) were determined by direct sequencing of ***: Associations of CYBA-675 T/A with high-density lipoprotein(HDl)(TT vs TA vs AA; P lycerides(TGl)(TT vs XA; P ly in NASH patients. For polymorphisms in the NOX4 gene, NOX4(rs3017887) CA + AA genotypes was significant associated with alanine aminotransferase(AlT)(CA + AA vs CC; P = 0.02). However, there was no association of SNPs in the CYBA and NOX4 genes encoding the NADPH oxidase system proteins and the presence of NASH. Regarding the clinical results, it was observed that the most advanced degrees of fibrosis occurred in patients diagnosed with type 2 diabetes mellitus(66.9% vs 37.5%, P lucose and insulin levels increased significantly in the presence of ***: There were associations between the presence of the allele A in the NOX4 SNP and a higher concentration of AlT in the NAFlD population; between the presence of the AA genotype in the polymorphism of the CYBA-675 T/A CYBA gene and a higher level of TGl and lower HDl in NASH patients. The presence of metabolic syndrome was associated with advanced degrees of fibrosis in NAFlD patients.

Crambe is a promising crop for biodiesel production. However, there is no much information available about techniques and research regarding the best performance in different regions to explore its potential. The objective of this study was to evaluate the effect of cadmium, chromium, copper and zinc in the development of crambe. The study was carried out in a greenhouse and it consisted of four independent experiments in completely randomized design, with three replications. Four metals with five levels were applied: 0, 10, 15, 20 and 25 mg·kg-1 for copper, cadmium, chromium;and 0, 20, 30, 40 and 50 mg·kg-1 for zinc, in order to evaluate the growth of crambe. Data were subjected to analysis of variance and comparison of means by Tukey test at 1 and 5% significance. Despite doses of cadmium, chromium, copper and zinc in growing crambe have influenced in some of the evaluated variables of the plants, in general, their development was similar in relation to doses and the metals applied. The importance of cultivating crambe is related to grain production, based on this, copper and cadmium decreased this production.

Many studies have been dedicated to the development of scaffolds for improving post-traumatic nerve regeneration. The goal of this study was to assess the effect on nerve regeneration, associating a hybrid chitosan membrane with non-differentiated human mesenchymal stem cells isolated from Wharton＇s jelly of umbilical cord, in peripheral nerve reconstruction after crush injury. Chromosome analysis on human mesenchymal stem cell line from Wharton＇s jelly was carried out and no structural alterations were found in metaphase. Chitosan membranes were previously tested in vitro, to assess their ability in supporting human mesenchymal stem cell survival, expansion, and differentiation. For the in vivo testing, Sasco Sprague adult rats were divided in 4 groups of 6 or 7 animals each： Group 1, sciatic axonotmesis injury without any other intervention （Group 1-Crush）; Group 2, the axonotmesis lesion of 3 mm was infiltrated with a suspension of 1 250 -1 500 human mesenchymal stem cells （total volume of 50 pl）（Group 2-CrushCell）; Group 3, axonotmesis lesion of 3 mm was enwrapped with a chitosan type Ill membrane covered with a monolayer of non-differentiated human mesenchymal stem cells （Group 3-CrushChitlllCell） and Group 4, axonotmesis lesion of 3 mm was enwrapped with a chitosan type III membrane （Group 4-CrushChiUll）. Motor and sensory functional recovery was evaluated throughout a healing period of 12 weeks using sciatic functional index, static sciatic index, extensor postural thrust, and withdrawal reflex latency. Stereological analysis was carded out on regenerated nerve fibers. Results showed that infiltration of human mesenchymal stem cells, or the combination of chitosan membrane enwrapment and human mesenchymal stem cell enrichment after nerve crush injury provide a slight advantage to post-traumatic nerve regeneration. The negative cardiorespiratory health outcomes due to extreme temperatures and air pollution are widely stud-ied,but knowledge about the effectiveness of the implementation of adaptive mechanisms remains *** objective of this paper is to explore the evidence of adaptive mechanisms for cardiorespiratory diseases regard-ing extreme temperatures and air pollution by comparing the results of two systematic literature review(SlR)processes sharing the same initial research question but led by two research groups with different academic back-grounds working in the same multidisciplinary *** start by presenting the methodological procedures and the results of the SlR triggered by the research group mainly composed by researchers with a background in ge-ography(named geographical strategy).We then compare these results with those achieved in the SlR led by the research group with a background in epidemiology(named epidemiological strategy).Both SlR were developed under the EU Horizon 2020 Project“EXHAUSTION”.The results showed:1)the lack of evidence regarding the effectiveness of adaptation measures,namely due to the limited number of studies about the topic,the prepon-derance of studies dedicated to heat extremes or the unbalance between different adaptation measures;2)that the choice of search terms in the geographical strategy,despite being more comprehensive at first sight,ended up retrieving less results,but it brought new studies that can complement the results of the epidemiological ***,it is suggested that to strengthen the empirical evidence of the effectiveness of adaptation measures,powerful multidisciplinary teams should work together in the preparation of SlR in topics of great complexity,such as the one presented in this paper.

Aims Mediterranean coastal dunes are habitats of great conservation interest,with a distinctive and rich *** the last century,Acacia spp.,native from Australia,have been introduced in Portugal,with the objective of stabilizing sand dunes,and since have become dominant in numerous sand dune *** invasion process led to the reduction of native plant species richness,changed soil characteristics and modified habitat’s microclimatic *** aim of this research was to typify and compare,in Mediterranean sand dune ecosystems,the ecophysiological responses to drought of Helichrysum italicum and Corema album,two native species,and Acacia longifolia,an exotic invasive *** addressed the following specific objectives:(i)to com-pare water relations and water use efficiencies,(ii)to evaluate water stress,(iii)to assess water use strategies and water sources used by plants and(iv)to evaluate the morphological adaptations at leaf and phyllode *** In order to obtain an integrative view of ecophysiological patterns,water relations and performance measuring methods have been applied:predawn(ψPD)and midday(ψMD)water potential,chloro-phyll a fluorescence,oxygen isotopic composition of xylem,rain and groundwater(δ18O)and leaf carbon isotopic discrimination(Δ13C).The leaf characteristics of the three species,as well as the histochemistry of non-glandular trichome cell walls,were also studied to identify morpho-traits related to drought *** Findings The results support our initial hypothesis:although *** clearly possesses a degree of resistance to water stress,such ability is provided by a different water strategy,when compared to native *** relied on morphological adaptations to restrict water loss,whereas the invasive species adjusted the water uptake as a way to balance their limited ability of restricting water *** corroborate that woody native species(i)have a conservative water-saving strategy an
