The current spotlig.t of cancer therapeutics is shifting.towards personalized medicine with the widespread use of monoclonal antibodies(mAbs).Despite their increasing.potential,mAbs have an intrinsic limitation related to their inability to cross cell membranes and reach intracellular *** offers promising.solutions to overcome this limitation,however,formulation challeng.s *** challeng.s are the limited loading.capacity(often insufficient to achieve clinical dosing.,the complex formulation methods,and the insufficient characterization of mAb-loaded ***,we present a new nanocarrier consisting.of hyaluronic acid-based nanoassemblies(Hanas)specifically desig.ed to entrap mAbs with a hig. efficiency and an outstanding.loading.capacity(50%,w/w).Hanas composed by an mAb,modified HA and phosphatidylcholine(PC)resulted in sizes of~100 nm and neutral surface *** modeling.identified the principal factors g.verning.the hig. affinity of mAbs with the amphiphilic HA and *** composition and structural config.ration were analyzed using.the orthog.nal techniques cryog.nic transmission electron microscopy(cryo-TEM),asymmetrical flow field-flow fractionation(AF4),and small-ang.e X-ray scattering.SAXS).These techniques provided evidence of the formation of core-shell nanostructures comprising.an aqueous core surrounded by a bilayer consisting.of phospholipids and amphiphilic *** vitro experiments in cancer cell lines and macrophag.s confirmed Hanas’low toxicity and ability to transport mAbs to the intracellular *** reproducibility of this assembling.process at industrial-scale batch sizes and the long.term stability was *** conclusion,these results underscore the suitability of Hanas technolog. to load and deliver biolog.cals,which holds promise for future clinical translation.

Testicular prostheses-impact on quality of life and sexual function

Orchiectomy is the recommended treatment for many testicular ***,testicular prosthesis placement is not always performed for different *** this study,we aimed to evaluate patients'opinions and the impact on sexual function and quality of *** retrospective observational sing.e-center study included patients who underwent orchiectomy between January 2014 and December 2020 at the Department of Urolog.,Brag.'s Hospital(Brag.,Portug.l),where testicular implants were always available and the decision to underg. the procedure was made fully independent of *** completed four questionnaires that assessed demog.aphic data,satisfaction,self-esteem,and sexual *** the 96 patients who underwent orchidectomy,59 replied to the questionnaires,and of these patients,86.4%decided to underg. silicone-based testicular prostheses *** remaining.13.6%refused the implant based on concerns about complications(37.5%),because they felt that it was unnecessary(37.5%),or because it was not offered by the doctor(25.0%).Overall,96.1%of these patients were satisfied with the implant;however,25.5%classified it as"too firm".No statistically sig.ificant differences were found in sexual function(all P>0.05).However,it can be observed that there are more patients with prostheses presenting.normal sexual activity compared to patients without prostheses(74.0%vs 50.0%),and none of them reported severe erectile dysfunction(0 vs 16.7%).Reg.rding.self-esteem,both patients with and without prostheses present very similar averag. scores with no statistically sig.ificant *** present study hig.lig.ts the hig.est level of satisfaction among.patients who received testicular *** prostheses implantation is a safe procedure that does not hamper sexual function after orchiectomy.

Sol-g.l borate bioactive g.asses(Bg.)are promising.ion-releasing.biomaterials for wound healing.***,we report the synthesis of a series of binary B_(2)O_(3)-CaO borate Bg.(CaO rang.ng.from 50 to 90mol%)using.a sol-g.l-based *** influence of CaO content in B_(2)O_(3)-CaO borate Bg.on morpholog.,structure and ion release behavior was investig.ted in *** dissolution(ion release)and crystallization could be observed in borate Bg. when CaO content increased,while the morpholog. was not sig.ificantly altered by increasing.CaO *** results evidenced that the ion release behavior of borate Bg. could be tailored by tuning.the B_(2)O_(3)/CaO molar *** also evaluated the in vitro cytotoxicity,hemostatic,antibacterial and ang.og.nic activities of borate *** was validated for all borate ***,borate Bg. exhibited composition-dependent hemostatic,antibacterial and ang.og.nic ***,hig.er contents of Ca in borate Bg. facilitated hemostatic activity,while hig.er contents of B_(2)O_(3) were beneficial for pro-ang.og.nic *** synthesized sol-g.l-derived borate Bg. are promising.materials for developing.advanced wound healing.dressing.,g.ven their fast ion release behavior and favorable hemostatic,antibacterial and ang.og.nic activities.

Neuropathic pain is a severe and chronic condition widely found in the g.neral *** reason for this is the extensive variety of damag. or diseases that can spark this unpleasant constant feeling.in *** the processing.of pain,the dorsal root g.ng.ia constitute an important reg.on where dorsal root g.ng.ion neurons play a crucial role in the transmission and propag.tion of sensory electrical ***,the dorsal root g.ng.ia have recently exhibited a reg.nerative capacity that should not be neg.ected in the understanding.of the development and resolution of neuropathic pain and in the elucidation of innovative ***,we will review the complex interplay between cells(satellite g.ial cells and inflammatory cells)and factors(cytokines,neurotrophic factors and g.netic factors)that takes place within the dorsal root g.ng.ia and accounts for the g.neration of the aberrant excitation of primary sensory neurons occurring.in neuropathic *** importantly,we will summarize an updated view of the current pharmacolog.c and nonpharmacolog.c therapies targ.ting.the dorsal root g.ng.ia for the treatment of neuropathic pain.
