High Sugar-Fat Diet Induces Metabolic-Inflammatory Disorders Independent of Obesity Development

Background: The modern dietary habit, which is rich in refined carbohydrates and saturated fats, increases the risk of chronic diseases due to the proinflammatory effect of these nutrients. Aim: To evaluate the impact of high sugar-fat diet in the development of metabolic-inflammatory disorders in non-obese animals. Methods: Male Wistar rats were distributed into two groups according to the diet: control and high sugar-fat for 30 weeks. It was analyzed: dietary efficiency;chow, water and caloric intake;metabolic and hormonal profile in plasma and inflammatory cytokines in epididymal adipose tissue. Data were compared by Student’s t test or by Mann-Whitney U test with p Results: HSF presented lower chow intake, higher water consumption and dietary efficiency with no difference in the caloric intake. The final body weight (FBW) and weight gain (WG) were lower in the HSF group and there was no difference in the adiposity index (AI). HSF diet-induced hyperglycemia and hyperinsulinemia with no difference for Homeostatic Model Assessment for Insulin Resistance (HOMA-IR). Triglycerides, uric acid, adiponectin and leptin levels were higher in the HSF group. The HSF group showed increased interleukin-6 (IL-6) and tumoral necrosis factor-alpha (TNF-α) levels in epidydimal adipose tissue. The urinary protein-creatinine ratio and albuminuria were higher in the HSF group. Conclusion: HSF diet intake is directly involved in the development of metabolic-inflammatory disorders independent of obesity, dissociating the view that increased adiposity is the major risk factor for complications commonly found in obese individuals.

This work describes the chemical composition of the essential oils extracted from fresh leaves of Lippia origanoides Kunth. and Mentha spicata L. and their antioxidant and insecticidal activities. The essential oils were extracted by steam distillation using a modified Clevenger apparatus and subsequently analyzed by gas chromatography-flame ionization detector and gas chromatography-mass spectrometry. The antioxidant activities were determined by the β-carotene-linoleic acid method and by sequestration of the 2,2-diphenyl-1-picryl-hidrazila radical. The concentrations of the essential oils and the synthetic standard, butylated hydroxyltoluene, were 25, 50, 100, 150, 200, 250, 300 and 500 μg mL-1. Insecticidal activity was analyzed by non-preference with choice and no choice against the aphid Myzus persicae Sulzer. Gas chromatography analysis of the essential oil from Lippia origanoides Kunth. revealed carvacrol (41.51%), p-cymene (18.36%), γ-terpinene (17.03%) and thymol (4.86%) as major constituents, and the essential oil from Mentha spicata L. contained piperitona (81.18%), piperitenone (14.57%) and limonene (1.47%) as the principal components. The essential oils and the standard exhibited dose-dependent antioxidant activities at the concentrations tested. The essential oils were shown to be potential agents in the integrated management of the aphid Myzus persicae Sulzer.

Vitamin D and Secondary Hyperparathyroidism in HIV Infected Patients Taking Antiretroviral Therapy

Objective: Due to the lack of studies assessing hypovitaminosis D and secondary hyperparathyroidism in Brazilian HIV-infected population, especially in the northeastern population, this study aimed to determine the profile of these conditions in patients infected with HIV and its correlation with immuno-virological, sociodemographic data and associated comorbidities. Methods: Comparison studies were obtained from routine clinical samples of HIV infected patients submitted for 25-OH Vitamin D, PTH and alkaline phosphatase determination. Results: A total of 78 patients were included, 42 (53.8%) males, mean age 45.7 years. Antiretroviral regimens most used in this study were Zidovudine/Lamivudine/Efavirenz 17.9%, Tenofovir/Lamivudine/Efavirenz 17.9%,Tenofovir/Lamivudine/Atazanavir-r 15.4%. The mean value CD4 count was 592.1 ± 247.2 cells/mm3, CD8 cell count was 1026.5 ± 467.3 cells/mm3, mean detectable viral load was 2220 ± 15703 copies and CD4/CD8 ratio was 0.63 ± 0.33. A total of 34 vitamin D dosages were collected with 41.2% representing sufficient amount and 58.8% insufficient. Alkaline Phosphatase (ALP) dosage was elevated in 49.3% (N=35) of the patients. Parathormone (PTH) was elevated in 18% (N = 11). Among patients with elevated PTH levels, 81.9% had elevated levels of ALP (p = 0.01). In the group of patients with high levels of ALP, 45.7% had a CD4 count 3 (p = 0.02). There was no significant difference in vitamin D related to gender (p = 0.21), age (p = 0.23), CD4 count (p = 0.26), suppressed viral load (p = 0.44) or blood glucose (p = 0.45). Conclusions: This study evidenced a high prevalence of Vitamin D insufficiency in Northeast Brazil, which suggests HIV infection correlation. A high prevalence of Hyperparathyroidism was detected and related with inflammatory condition persistence and low CD4 count. We suggest improve vitamin D follow up and measurements in this population with better CD4 count control to avoid future osteoarticular complications of HIV

Heart diseases are the main cause of mortality in Mexico, being coronary heart disease the most frequent in the country. Its high prevalence makes important the study of the pathophysiology and the search for prognostic factors. Different genes and polymorphisms promote atherogenesis and coronary artery disease, they affect inflammatory and vascular pathological processes. ana;">Interferon regulatory factor 5 (IRF5) is associated with coronary heart disease, it promotes chronic inflammation and cytokines release;it could trigger immune reactions and its activating receptors express in the vascular endothelium. Besides, polymorphisms in the renin-angiotensin-aldosterone system (RAAS) are implied with coronary disease, they are found in angiotensinogen (AGT), angiotensin II type 1 receptor (AT1R), angiotensin II type 2 receptor (AT2R), and angiotensin-converting enzyme (ACE) genes. These genetic polymorphisms are associated with a prothrombotic state, endothelial dysfunction, and immune activation. Multiple experimental studies showed that chronic activation of RAAS and chronic expression of IRF5 generates an environment prone to the development of atherosclerosis, and autoimmune and cardiovascular diseases. Studying these specific genes and their relationship with coronary heart disease will allow a better understanding of the pathological process and possibly the quest for new treatments.

Purpose: The primary aim of this paper is to design, create, and improve functional and artificial Bruch’s membranes (BM) using bioengineering techniques, which can be applied in the treatment of maculopathies by supporting the growth and maintenance of retinal pigment epithelium (RPE) cells, thereby potentially enabling subretinal implantation in patients. Methods: We fabricated by electrospinning ultrathin 3D nanofibrous membranes from Polycaprolactone (PCL), and different concentrations of gelatin (5%, 15% and 30%). ARPE-19 cells were seeded onto these artificial membranes. MTT assays were performed in order to evaluate ARPE-19 cell viability and cytotoxicity. IF assays were realized to observe the ARPE-19 cells onto each membrane. Ultrastructure of the modified Bruch’s membrane and ARPE-19 morphology after 25 days of culture were studied with transmission and scanning electron microscopy. To evaluate expression changes in markers of ARPE-19 (RPE65 and ZO-1) qRT-PCR assays were realized. Data from three independent experiments were pooled and expressed as the mean SD. A confidence level of P 0.05 was considered to be statistically significant. Results ARPE-19 cells grew on PCL/Gelatin membranes mainly in PCL/30% gelatin combination, which had not cytotoxic effect. RPE65/cytokeratin-18/ and actin-positive ARPE-19 cells formed a correctly orientated monolayer of polygonal cells with morphological polarity. The apical cell surfaces exhibited abundant protruding microfolds. Moreover, zones of polygonal border look as if ARPE-19 cells fused among, suggesting the presence of tight junctions. The expression of RPE65 and ZO-1 was unchanged. Conclusions: PCL/30% Gelatine membranes may imitate the natural BM to such extent that they support RPE-cells and exhibited RPE-like morphology. 目的：评估2种粘接系统[Adper Single Bond plus（3MESPE）和Clearfil SEBOND（Kuraray）]与预先是否经过Nd：YAG激光或Nd：YAG激光加氟处理的正常或龋坏牙本质粘接后的微拉伸力。材料和方法：将60颗离体第三磨牙的牙本质表面暴露后分为12组。1～6组经过pH循环产生人工龋，7～12组保持正常牙本质。牙本质表面采用3种处理方法：Nd：YAG激光照射（60mJ，15Hz．09w）1min：Nd：YAG激光照射联合氟凝胶处理：不处理（对照组）。实验组根据产品说明书要求应用粘接材料并分层堆塑制成复合树脂块（FiItek Z250．3M ESPE），牙齿沿X和Y轴方向做连续片切．应用万能试验仪进行微拉伸力试验。结果：通过ANOVA和Tukey统计分析（P〈0.05）发现正常牙本质应用Clearfil SE Bond组的平均粘接力最大（4065MPa）．SingleBond组次之（342MPa）。龋坏牙本质组无论是否事先经过激光照射应用两种粘接系统后的粘接力明显下降。但激光照射后应用Clearfil SE Bond组微拉伸粘接力最高。另外．激光照射联合氟化物处理后应用两种粘接系统的微拉伸力有所下降。结论：与全酸蚀粘接系统和激光照射相比．临床上在备洞时用激光照射龋坏牙本质．然后应用自酸蚀粘接系统可以获得较高的粘接力。

The extraction of essential oils was performed by using the technique of hydrodistillation modified Clevenger apparatus, identification and quantification of the constituents was achieved by Gas Chromatography coupled to Mass Spectrometry, and Gas Chromatography equipped with a flame ionization detector. Assessment of allelopathic activity was evaluated with the use of the method that assesses the direct contact of essential oils on germination and vigor of lettuce seeds. The major constituents that characterize the essential oil from the flowers of Callistemon viminalis were 1, 8-cineole, α-pinene and limonene at concentrations of 66.9%, 16.0% and 10.0%, respectively. The essential oil presented allelopathic activity at intensities that varied proportionately to the concentration of the essential oil, with a reduction in the percentage of germination and the germination speed index (GSI) of lettuce seeds and in the dry mass and length of shoots and roots of lettuce seedlings.

盘状星系中的棒结构特征与星系自身所处环境及整体恒星形成特征之间存在复杂的相互关系。利用细致的星系形态特征测量计划Galaxy Zoo DECaLS中的数据构建了一组目前最大的具有同样恒星质量分布的强棒、弱棒和无棒星系样本。通过引入星系所处暗晕质量及相空间特征来控制星系所处的环境效应之后,进一步对比研究强棒、弱棒和无棒星系的恒星形成特征。研究表明,有棒星系中的恒星形成熄灭的比例系统地高于无棒星系,且强棒的效应更为明显。但是,对于仍处于恒星形成主序上的星系来说,强棒、弱棒和无棒星系的恒星形成特征并没有显著差别。以上结果表明,星系中的棒特征在其存在的时标中可能使部分星系迅速地从恒星形成转入熄灭状态。

Determination of Essential (Ca, Fe, I, K, Mo) and Toxic Elements (Hg, Pb) in Brazilian Rice Grains and Estimation of Reference Daily Intake

The levels of Hg and Pb (toxic elements) and Ca, Fe, I, K, Mo (essential elements) were measured in 44 Brazilian rice samples divided among white polished (W), parboiled white (PW) and parboiled brown (PB). Rice grains were ana- lyzed by inductively coupled plasma mass spectrometry (ICP-MS). Concentrations of toxic elements varied from 0.3 to 13.4 and 0.4 to 14.5 ng?g–1 for Hg and Pb, respectively. The values for essential elements were in the same range of those for rice produced in other countries. In general parboiled samples presented higher levels of all elements in study compared to white samples. The Estimated Daily Intake through rice consumption was 0.44 μg for Pb, 0.22 μg for Hg, 0.65 μg for I, 39.5 μg for Mo, 0.43 mg for Fe, 4.1 mg for Ca and 88.9 mg for K. Our findings demonstrated that rice can contribute significantly to the RDIs of molybdenum and potassium, but it can not be considered an important source of I, Fe and Ca. Moreover, the estimated
