Chemical and spectroscopic characterization of humic substances from sediment and riparian soil of a highly polluted urban river(Suquía River,Córdoba,Argentina)

The Suquia River,the largest urban river in Cordoba(Argentina),has been severely polluted for *** must be taken to restore its environmental quality by managing riparian zones for increased water-self *** current study aimed to characterize organic matter(OM) dynamics and humic substances(HS) spectrochemical properties along the lower-middle basin of the Suquia *** soil(0-20 cm) and sediment(0-10 cm) samples were collected from a reference location(S1)and four polluted sites(S2-S5) during a low-flow *** contents of soil and sedimentary OM and HS fractions were analyzed by wet oxidation,as well as HS Fourier transform infrared(FT-IR) and ultraviolet-visible(UV-Vis) spectrochemical *** OM and HS fractions from riparian soil were high upstream of Cordoba City(S1 and S2,50.2-50.4 g/kg OM) and within a 50 km downstream location(S5,30.9 g/kg OM) owing to a surplus of fresh plant biomass-carbon(C) *** heterogeneous sediment samples did not show any significant differences among sites(P> 0.05).The lowest values of the ratio of absorbances at 465 and 665 nm(E4/E6)(1.78) and the Δ log K(0.15) coefficient(a measure of HS maturity degree) were obtained downstream of Cordoba City,for both riparian soil and sediment,indicating that HS were enriched by more condensed aromatic structures within highly degraded portions of the *** samples exhibited similar IR spectra,implying overlapping recalcitrant-C structures at the functional group level,but with different absorbance *** from the current study constitute a baseline for understanding the chemical nature of HS from sediment and riparian soil along the Suquia River and can be used as a reference for future studies tracking OM compositional changes over time.

This study aims to compare the grain protein profile of four Brazilian cowpea cultivars (BRS Aracê, BRS Itaim, BRS Pajeú, and BRS Xiquexique) by two-dimensional electrophoresis (2-de) and principal component analysis (PCA). 2-de efficiently separate cowpea protein profiles, showing high homogeneity among the four cultivars. In addition, the principal component analysis indicated that there is a difference in abundance of proteins among the cultivars. The cultivars BRS Aracê and BRS Xiquexique, both biofortified in iron and zinc, were separated from the cultivars BRS Itaim and BRS Pajeú. These results demonstrate that protein profiles can be used to discriminate cowpea varieties.

The suicide phenomenon involves complex interactions between psychological,biological,cultural and socio-environmental *** study aimed to assess the epidemiological and toxicological profiles of officially confirmed suicide *** study was performed using autopsy reports,forensic anthropology examinations and police summaries of all suicide cases that underwent toxicological analysis at an Official Forensic Laboratory(Minas Gerais,Brazil)in 1 *** sample set was composed of 351 suicide victims,predominantly men(70.32%),most of them were adults between 31 and 64 years old(62.11%),with mixed skin colour(48.89%)and low educational level(66.44%).The most common suicide method was hanging(57.79%),followed by intoxication(30.45%).Most victims presented positive toxicological results(56.41%),especially for the presence of medicines(37.6%),illicit drugs(36.3%)and pesticides(26.1%).Our study corroborated previous data that most suicide victims have low educational *** most common toxicological findings were medicines,especially prescription drugs,followed by illicit drugs and *** hope this study contributes to reflections and planning of preventive suicide programmes,considering the described profiles of victims.

Tolerogenic dendritic cells （DCs） are widely studied for their possible use in the treatment of inflammatory disorders, such as autoimmune diseases. One of the obstacles for the use of this cell-based therapy is the characterization of drugs that are able to modulate DCs. We have previously shown that chloroquine （CQ）, an antimalarial agent, has the ability to modulate DCs towards a tolerogenic phenotype.1 These tolerogenic DCs are able to suppress the development of experimental autoimmune encephalomyelitis （EAE）, a T cell-driven mouse model of human multiple sclerosis. In addition, several studies have proposed that nitric oxide （NO） plays a major role in the dif- ferentiation of regulatory T cells （Tregs） and the suppression of Thl/Th17 cells.2＂3 However, little is known about the role of DC-derived NO in the modulation of inflammatory auto- immune responses. Thus, we aimed to evaluate whether NO plays a role in the tolerogenic activity of CQ-treated DCs （CQ- DCs）. We found that CQ induces DC production of NO and expression of indoleamine 2,3-dioxygenase （IDO）, as well as inducible nitric oxide syrtthase （iNOS）.

Early detection of infected asymptomatic plants could prevent the dissemination of tomatobacterial canker throughout the crop and, thus, reduce associated economic losses. Reflectance spectroscopy has been proposed for the detection of diseases in plants, mainlycaused by foliar fungal pathogens. In contrast, bacterial canker is a vascular disease thatdoes not produce visible foliar symptoms until it is too late to avoid contagion to otherplants. The effects of the disease on the conductivity of the xylem have an impact onthe water content of the leaves;initially, a subtle effect that cannot be assessed with thenaked eye but can be evaluated by reflectance spectroscopy. The objective of this workwas to identify indices, based on the reflectance spectral signature of the plants, for thedetection of tomato plants affected by bacterial canker prior to the appearance of symptoms. To verify that the proposed methodology is applicable to a broad range of tomatogenotypes, two experiments were carried out with different tomato cultivars grown in potsand another on the ground in a greenhouse. Plants were inoculated with the pathogenicbacteria and the reflectance spectrum of the apical leaflet of two leaves per plant wereobtained before symptoms expression;several spectral indices were calculated from thereflectance spectra. Three indices, of the shortwave-infrared zone, allowed the detectionof bacterial canker inoculated plants in a fast and non-destructive way, up to one weekbefore symptoms were visible: Normalized Difference Water Index (NDWI), Simple Ratioof Water Index (SRWI) and Water Index 1 180 (WI1180). Our research demonstrates, forthe first time, the usefulness of spectral indices sensitive to water content for the identifi-cation of diseased plants. These indices arise as a promising tool for the early detection ofvascular plant diseases, that could be integrated into sustainable management plans.

Escherichia coli and other Proteobacteria are augmented and several other bacteria are diminished in Crohn’s (CD) disease patients’ intestine. This imbalance in bacterial species composition—termed dysbiosis—seems to be determinant of CD manifestation. Since a great part of intestinal bacteria are not cultivable, detection of CD dysbiosis is accomplished by molecular tools, involving sequences analysis of the 16SrRNA gene (16SrDNA) present in the patient’s clinical samples, which can be done by sequencing or electrophoresis in denaturing gels of 16SrDNA amplicons. By analyzing, by temperature gradient gel electrophoresis (TGGE) and next generation sequencing of 16SV6-V8rDNA amplicons present in gram negative cultures from four distinct clinical samples of a control subject and a CD patient, this study demonstrates that both techniques were able to detect E. coli overgrowth and reduction in species richness in CD and that TGGE can discriminate sequences collectively labeled as “unclassified” in 16SrDNA databases. Although TGGE per se does not identify the sequences, the discriminatory power that it confers represents valuable accessory information to next generation DNA sequencing (NGS), and as such must be used as a NGS complementary tool.
