Prime editing(PE)technology enables precise alterations in the genetic code of a genome of *** offers great potential for identifying major agronomically important genes in plants and editing them into superior variants,ideally targeting multiple loci simultaneously to realize the collective effects of the ***,we report the development of a modular assembly-based multiplex PE system in rice and demon-strate its efficacy in editing up to four genes in a single transformation *** duplex PE(DPE)system achieved a co-editing efficiency of 46.1%in the T0 generation,converting TFIIAg5 to xa5 and xa23 to *** resulting double-mutant lines exhibited robust broad-spectrum resistance against multiple Xanthomonas oryzae pathovar oryzae(Xoo)strains in the T1 *** addition,we success-fully edited OsEPSPS1 to an herbicide-tolerant variant and OsSWEET11a to a Xoo-resistant allele,achieving a co-editing rate of 57.14%.Furthermore,with the quadruple PE(QPE)system,we edited four genes—two for herbicide tolerance(OsEPSPS1 and OsALS1)and two for Xoo resistance(TFIIAg5 and OsSWEET11a)—using one construct,with a co-editing efficiency of 43.5%for all four genes in the T0 *** performed multiplex PE usingfive more constructs,including two for triplex PE(TPE)and three for QPE,each targeting a different set of *** editing rates were dependent on the activity of pegRNA and/or *** instance,optimization of ngRNA increased the PE rates for one of the targets(OsSPL13)from 0%to 30%but did not improve editing at another target(OsGS2).Overall,our modular assembly-based system yielded high PE rates and streamlined the cloning of PE reagents,making it feasible for more labs to utilize PE for their editing *** have significant implications for advancing gene editing techniques in plants and may pave the way for future agricultural applications.

Background: Hepatic ischemia-reperfusion injury (HIRI) stands as an unavoidable complication arising from liver surgery, profoundly intertwined with its prognosis. The role of lysine methyltransferase SET domain bifurcated 1 (SETDB1) in HIRI remains elusive, despite its confirmation as a potential therapeutic target for diverse diseases. Here, we investigated the mechanism by which SETDB1 regulated HIRI. Methods: RNA sequencing data were used to identify the expression and potential targets of SETDB1 through bioinformatics analysis. To elucidate the impact of SETDB1 on HIRI, both an in vivo model of HIRI in mice and an in vitro model of hepatocyte hypoxia/reoxygenation were established. Biochemical and histological analyses were used to investigate the influence of SETDB1 on liver damage mediated by HIRI. Chromatin immunoprecipitation and coimmunoprecipitation were implemented to explore the in-depth mechanism of SETDB1 regulating HIRI. Results: We confirmed that hepatocellular SETDB1 was up-regulated during HIRI and had a close correlation with HIRI-related inflammation and apoptosis. Moreover, inhibition of SETDB1 could mitigate HIRI-induced liver damage, inflammation, and apoptosis. Through our comprehensive mechanistic investigation, we revealed that SETDB1 interacts with apoptosis-signal-regulating kinase 1 (ASK1) and facilitates the methylation of its lysine residues. Inhibition of SETDB1 resulted in reduced phosphorylation of ASK1, leading to a marked suppression of downstream c-Jun N-terminal kinase (JNK)/p38 signaling pathway activation. The therapeutic effect on inflammation and apoptosis achieved through SETDB1 inhibition was nullified by the restoration of JNK/p38 signaling activation through ASK1 overexpression. Conclusions: The findings from our study indicate that SETDB1 mediates lysine methylation of ASK1 and modulates the activation of the ASK1–JNK/p38 pathway, thus involved in HIRI-induced inflammation and apoptosis. The balance between metal and acid sites directly affects the preparation of high-performance cracking catalysts with high heat sink and low ***,how to control acid-metal sites balance and its relationship with cracking performance are reported *** this work,a series of Pt/Al2O3-SiO2dual sites catalysts with different metal to acid active sites ratio(CM/CSA) were constructed by ethanolassisted impregnation method and the impact on n-decane cracking under supercritical conditions was systematically and deeply *** results showed that the conversion and carbon deposition increased gradually with varied CM/CSAand reached the balance at CM/CSAof *** proper ratio CM/CSA（0.13） can balance the deep dehydrogenation coking over metal active sites and high heat sink of cracking over acid active sites,the chemical heat sink reaches amazing 1.75 MJ/kg and carbon deposition is only22.03 mg/cm2at 750℃.Meanwhile,the few metal sites at low CM/CSAand the few strong acid sites at high CM/CSAare the main factors limiting the cracking *** CM/CSAlimit the activation of C-H bond and deep dehydrogenation of coking precursor,resulting in relative low cracking activity and carbon deposition,while high CM/CSAlimit the activation of C-C bond and increase the deep *** this contribution,design and construction of metal-acid dual sites can not only provide the technical solution for the preparation of high heat sink and low coking cracking catalyst,but also deepen the understanding of the cracking path of hydrocarbon fuel.

Side-chain engineering has emerged as a highly effective strategy for tailoring the aggregation behavior and charge transport properties of non-fullerene small molecule acceptors(SMAs).In this study,we designed and synthesized two SMAs,namely BTPSi-Bu and BTPSi-Pr,respectively incorporating tributylsilyloxy and trisopropylsilyloxy groups in their outer ***,BTPSi-Bu exhibited better planarity,crystallization,and favorable phase separation when paired with PM6 donor polymer compared to its counterpart,*** resulting organic solar cells,utilizing the PM6:BTPSi-Bu blend,demonstrated a remarkable power conversion efficiency of 17.41%and a high open-circuit voltage of 0.859 *** findings underscore the significance of integrating trialkylsilyloxy side chains into SMAs as a rational design approach for enhancing the performance of photovoltaic systems.
