Comparison of tRMM and Water District Rain Rates over New Mexico

this paper compares monthly and seasonal rain rates derived from the Version 5 （V5） and Version 6 （V6） tRMM Precipitation Radar （tPR, tSDIS reference 2A25）, tRMM Microwave Imager （tMI, 2A12）, tRMM Combined Instrument （tCI, 2B31）, tRMM calibrated IR rain estimates （3B42） and tRMM merged gauge and satellite analysis （3B43） algorithms over New Mexico （NM） with rain gauge analyses provided by the New Mexico water districts （WD）. the average rain rates over the NM region for 1998- 2002 are 0.91 mm d^-1 for WD and 0.75, 1.38, 1.49, 1.27, and 1.07 mm d^-1 for V5 3B43, 3B42, tMI, PR and tCA; and 0.74, 1.38, 0.87 and 0.97 mm d^-1 for V6 3B43, tMI, tPR and tCA, respectively. Comparison of V5 3B43 with WD rain rates and the daily tRMM mission index （tPR and tMI） suggests that the low bias of V5 3B43 for the wet months （summer to early fall） may be due to the non-inclusion of some rain events in the operational gauge analyses that. are used in the production of V5 3B43. Correlation analyses show that the WD rain rates vary in phase, with higher correlation between neighboring WDs. High temporal correlations （〉0.8） exist between WD and the combined algorithms （3B42, 3B43 and tCA for both V5 and V6） while satellite instrument algorithms （PR, tMI and tCI） are correlated best among themselves at the monthly scale. Paired t-tests of the monthly time series show that V5 3B42 and tMI are statistically different from the WD rain rates while no significant difference exists between WD and the other products. the agreements between the tRMM satellite and WD gauge estimates are best for the spring and fall and worst for winter and summer. the reduction in V6 tMI （-7.4%） and tPR （-31%） rain rates （compared to V5） results in better agreement between WD estimates and tMI in winter and tPR during summer.

AIM to characterize the role of apolipoprotein B100(apoB 100) in hepatitis C viral(HCV) infection. MEtHODS In this study, we utilize a gene editing tool, transcription activator-like effector nucleases(tALENs), to generate human hepatoma cells with a stable genetic deletion of APOB to assess of apoB in HCV. Using infectious cell culture-competent HCV, viral pseudoparticles, replicon models, and lipidomic analysis we determined the contribution of apoB to each step of the viral lifecycle. We further studied the effect of mipomersen, an FDAapproved antisense inhibitor of apoB 100, on HCV using in vitro cell-culture competent HCV and determined itsimpact on viral infectivity with the tCID50 method. RESULtS We found that apo B100 is indispensable for HCV infection. Using the JFH-1 fully infectious cell-culture competent virus in Huh 7 hepatoma cells with tALENmediated gene deletion of apoB(APOB KO), we found a significant reduction in HCV RNA and protein levels following infection. Pseudoparticle and replicon models demonstrated that apo B did not play a role in HCV entry or replication. However, the virus produced by APOB KO cells had significantly diminished infectivity as measured by the tCID-50 method compared to wildtype virus. Lipidomic analysis demonstrated that these virions have a fundamentally altered lipidome, with complete depletion of cholesterol esters. We further demonstrate that inhibition of apoB using mipomersen, an FDA-approved anti-sense oligonucleotide, results in a potent anti-HCV effect and significantly reduces the infectivity of the virus. CONCLUSION Apo B is required for the generation of fully infectious HCV virions, and inhibition of apo B with mipomersen blocks HCV. targeting lipid metabolic pathways to impair viral infectivity represents a novel host targeted strategy to inhibit HCV.

We have previously shown that capacitively coupled electrical stimulation of either normal bovine articular chondrocytes or osteoarthritic human articular cartilage explants resulted in up-regulation of cartilage matrix gene expression and down-regulation of metalloproteinase gene expression. In addition, collagen and proteoglycan protein levels were also elevated. to determine visually the effect of specific electric fields on modifying cartilage structure, freshly harvested human full-thickness osteoarthritic cartilage explants were stimulated in the absence or presence of interleukin-1β, an inflammatory cytokine, and were examined photographically and spectrophotometrically. Hexosamine and hydroxyproline contents were also determined. Spectrophotometric analysis was used to quantify any changes in the depth of defects in the cartilage ranging from surface level (red-colored) to the deepest affected layer (blue-colored). Interleukin-1β treatment alone caused significant additional cartilage erosion. Electrical stimulation alone resulted in significant decreases in the cartilage defects. Electrical stimulation in the presence of interleukin-1β resulted in a small, but significant, surface improvement. Meta-analysis also confirmed a significant increase in the hexosamine and hydroxyproline contents (indicating matrix deposition). It was concluded that an appropriate electric field could modify osteoarthritic lesions in full-thickness cartilage plugs by increasing matrix production and/or by decreasing matrix destruction. Furthermore, it appears that spectrophotometric analysis is a relatively easy method for quantifying the “filling in” or healing of articular cartilage defects.

Formation potential and analysis of 32 regulated and unregulated disinfection by-products: two new simplified methods

Water disinfection is an essential process that provides safe water by inactivating pathogens that cause waterborne diseases. However, disinfectants react with organic matter naturally present in water, leading to the formation of disinfection by-products(DBPs). Multi-analyte methods based on mass spectrometry(MS) are preferred to quantify multiple DBP classes at once however, most require extensive sample pre-treatment and significant resources. In this study, two analytical methods were developed for the quantification of 32 regulated and unregulated DBPs. A purge and trap(P&t) coupled with gas chromatography mass spectrometry(GC-MS) method was optimized that automated sample pre-treatment and analyzed volatile and semi-volatile compounds, including trihalomethanes(tHMs), iodinated trihalomethanes(I-tHMs), haloacetonitriles(HANs), haloketones(HKts) and halonitromethanes(HNMs). LOQs were between 0.02-0.4 μg/L for most DBPs except for 8 analytes that were in the low μg/L range. A second method with liquid chromatography(LC) tandem mass spectrometry(MS/MS) was developed for the quantification of 10 haloacetic acids(HAAs) with a simple clean-up and direct injection. the LC-MS/MS direct injection method has the lowest detection limits reported(0.2-0.5 μg/L). Both methods have a simple sample pre-treatment, which make it possible for routine analysis. Hyperchlorination and uniform formation conditions(UFC) formation potential tests with chlorine were evaluated with water samples containing high and low tOC. Hyperchlorination formation potential test maximized tHMs and HAAs while UFC maximized HANs. Ascorbic acid was found to be an appropriate quencher for both analytical methods. Disinfected drinking water from four water utilities in Alberta, Canada were also evaluated.
