Objective:Bone metastasis occurs in up to 90%of men with advanced prostate cancer and leads to fractures,severe pain and *** metastases induce a spectrum of types of bone lesions which can respond differently to therapy even within individual prostate cancer ***,the special environment of the bone makes the disease more complicated and incurable.A model in which bone lesions are reproducibly induced that mirrors the complexity seen in patients would be invaluable for pre-clinical testing of novel *** microstructural changes in the femurs of mice implanted with PCSD1,a new patient-derived xenograft from a surgical prostate cancer bone metastasis specimen,were ***:Quantitative micro-computed tomography(micro-CT)and histological analyses were performed to evaluate the effects of direct injection of PCSD1 cells or media alone(Control)into the right femurs of Rag2/gc/male ***:Bone lesions formed only in femurs of mice injected with PCSD1 *** volume(BV)was significantly decreased at the proximal and distal ends of the femurs(ply increased along the femur ***:PCSD1 cells reproducibly induced bone loss leading to osteolytic lesions at the ends of the femur,and,in contrast,induced aberrant bone formation leading to osteoblastic lesions along the femur ***,the interaction of PCSD1 cells with different bone region-specific microenvironments specified the type of bone *** approach can be used to determine if different bone regions support more therapy resistant tumor growth,thus,requiring novel treatments.

葡萄糖对小鼠胚胎干细胞生长的影响

目的:目前小鼠胚胎干细胞(mouse embry-onic stem cells,mESC)常规应用高浓度葡萄糖(25 mmol/l)培养基培养,但是在应用干细胞向糖尿病胰岛beta细胞分化的研究中发现,慢性高糖培养可促进干细胞的凋亡、降低细胞分化的效率及分化后胰岛beta细胞对葡萄糖的反应性,因此本研究拟选择合适的较低浓度的葡萄糖以优化胚胎干细胞的培养基、提高胚胎干细胞的生长、分化效率。方法:mESC传代4或12 h后,将传统的25 mmol/l葡萄糖培养基分别换为5、10、15、25 mmol/l葡萄糖培养基培养,均用碱性磷酸酶(AP)染色计数细胞集落形成情况、台盼兰染色测定细胞数目,MTT法测定细胞活力及用4’,6-联脒-2-苯基吲哚(4’,6-diamidino-2-phenylindole,DAPI)染色检测细胞凋亡情况。结果:mESC传代4 h后即调换葡萄糖浓度:(1)各葡萄糖浓度组(5、10、15 mmol/l)与25 mmol/l组相比,集落形成明显减少。(2)各葡萄糖浓度组与25 mmol/l组相比,mESC增殖及细胞活力均受到不同程度明显影响。mESC传代12 h后调换葡萄糖浓度:(1)各葡萄糖浓度组(5、10、15 mmol/l)与25 mmol/l组相比,集落形成无明显变化,且AP染色呈强阳性。(2)15 mmol/l葡萄糖组对mESC增殖及细胞活力均无明显影响。(3)15 mmol/l葡萄糖组与25 mmol/l葡萄糖组细胞核形态正常,均未见明显凋亡。结论:ESC传代12 h后将培养基中传统的高糖(25 mmol/l)降低为15 mmol/l,不影响mESC细胞活力及多能分化潜能和未分化状态。

Bacterial Transfer from Hands While Eating Popcorn

Popcorn is a very social food, often shared with others and offered at many major sporting events, concerts, movies, and fairs. However, sharing may not be safe since microorganisms found on hands may be transferred onto the shared popcorn. This study was conducted to determine if bacteria are transferred from hands to popcorn during handling. Over 30 samplings revealed that bacterial transfer to popcorn from hands was very low;however transfer did occur with large variation between subjects. Since hands and surfaces can carry bacteria in situations where food is being shared, transferring bacteria from one person to another person is always a risk.

In this study we examined the suitability of the 3H-imidazo[4,5-b]pyridine ring system in developing novel anticancer and anti-inflammatory agents incorporating a diaryl pharmacophore. Eight 2,3-diaryl-3H-imidazo[4,5-b]pyridine derivatives retrieved from our in-house database were evaluated for their cytotoxic activity against nine cancer cell lines. The results indicated that the compounds showed moderate cytotoxic activity against MCF-7, MDA-MB-468, K562 and SaOS2 cells, with K562 being the most sensitive among the four cancer cell lines. The eight 2,3-diaryl-3H-imidazo[4,5-b]pyridine derivatives were also evaluated for their COX-1 and COX-2 inhibitory activity in vitro. The results showed that compound 3f exhibited 2-fold selectivity with IC_(50) values of 9.2 and 21.8 mmol/l against COX-2 and COX-1, respectively. Molecular docking studies on the most active compound 3f revealed a binding mode similar to that of celecoxib in the active site of the COX-2 enzyme.

Caspase-1 activation and mature interleukin-1β release are uncoupled events in monocytes
