Background The clinical utility and safety of sargramostim has previously been reported in cancer,acute radiation syndrome,autoimmune disease,inflammatory conditions,and Alzheimer’s *** safety,tolerability,and mecha-nisms of action in Parkinson’s disease(PD)during extended use has not been *** As a primary goal,safety and tolerability was assessed in five PD patients treated with sargramostim(Leukine®,granulocyte-macrophage colony-stimulating factor)for 33 *** goals included numbers of CD4+T cells and monocytes and motor ***,metabolic,immune,and neurological evaluations were assessed during a 5-day on,2-day off therapeutic regimen given at 3μg/*** 2 years,drug use was discon-tinued for 3 *** was then followed by an additional 6 months of *** Sargramostim-associated adverse events included injection-site reactions,elevated total white cell counts,and bone *** drug,blood analyses and metabolic panels revealed no untoward side effects linked to long-term *** Parkinson’s Disease Rating Scale scores remained stable throughout the study while regulatory T cell number and function were *** the initial 6 months of treatment,transcriptomic and proteomic mono-cyte tests demonstrated autophagy and sirtuin *** finding paralleled anti-inflammatory and antioxidant activities within both the adaptive and innate immune profile *** Taken together,the data affirmed long-term safety as well as immune and anti-inflammatory responses reflecting clinical stability in PD under the sargramostim *** in larger patient populations is planned in a future phase II evaluation.

Array-based comparative genomic hybridization (array CGH),a method used to detect gains or losses of genetic material,has recently been applied to prenatal diagnosis of genomic imbalance in the clinical laboratory setting. This new and exciting diagnostic tool represents a major technological step forward in cytogenetic testing and addresses many of the limitations of current cytogenetic methods. Conventional chromosome analysis,the current gold standard in prenatal diagnosis,focuses primarily on the detection of common aneuploidies and is limited by its capacity to detect only those copy number changes that are large enough to be microscopically visible (typically 5-6 mb in size at the 500 band level). In contrast,array CGH analysis simultaneously evaluates regions across the entire genome and allows for detection of unbalanced structural and numerical chromosome abnormalities of less than one hundred kb. Array CGH analysis also overcomes some of the limitations of chromosome analysis,such as the requirement for cell culture and longer reporting time,by using direct uncultured fetal specimens. With many diagnostic laboratories now embracing this technology,the past year has seen tremendous growth in the use of array CGH analysis for prenatal diagnosis. This review aims to summarize array CGH methodo-logy and its current applications in prenatal diagnosis.

In this study,we demonstrated the label-free continuous separation and enrichment of Bacillus subtilis populations based on length using viscoelastic ***,a gram-positive,rod-shaped bacterium,has been widely used as a model organism and an industrial *** can be arranged in different morphological forms,such as single rods,chains,and clumps,which reflect differences in cell types,phases of growth,genetic variation,and changing environmental *** ability to prepare *** populations with a uniform length is important for basic biological studies and efficient industrial ***,we systematically investigated how flow rate ratio,poly(ethylene oxide)(PEO)concentration,and channel length affected the length-based separation of *** *** lateral positions of *** cells with varying morphologies in a straight rectangular microchannel were found to be dependent on cell length under the co-flow of viscoelastic and Newtonian ***,we evaluated the ability of the viscoelastic microfluidic device to separate the two groups of *** cells by length(i.e.,1-5μm and>5μm)in terms of extraction purity(EP),extraction yield(EY),and enrichment factor(EF)and confirmed that the device could separate heterogeneous populations of bacteria using elasto-inertial effects.

Effect of spatial scale and topography on spatial heterogeneity of soil seed banks under grazing disturbance in a sandy grassland of Horqin Sand Land, Northern China

Soil seed banks play an important role in the distribution and composition of plant communities in semiarid grassland ecosystems. However, information on how spatial scale influences the spatial heterogeneity of soil seed banks in a grassland under grazing disturbance is still lacking. Based on field sampling and greenhouse germination, we measured the species composition and seed density of soil seed banks at different spatial scales （30 mx30 m, 30 mx60 m and 30 mx90 m） along a topographical gradient in a sandy grassland in Horqin Sand Land, Northern China. By applying geostatistical methods, we examined how spatial scale and topography affected the spatial distribution of soil seed banks in the study area. Our results showed that the total number of species in soil seed banks, as well as the number of dominant annuals, increased with the increase of spatial scales. Seed density in soil seed banks decreased with the increase of spatial scales due to an increase in the slopes and relative heights of the sampling points. Geostatistical analysis showed that the relative structural variance （C/（C0＋C）） of seed density and species richness were over 65% for all spatial scales, indicating that these variables had an ob- vious spatial autocorrelation and the spatial structured variance accounted for the largest proportion of the total sample variance. Spatial autocorrelation of seed density in soil seed banks increased with the increase of measured scales, while that of species richness showed a reverse trend. These results suggest that the total number of spe- cies in soil seed banks is spatial scale dependent and lower topography may accommodate more seeds. Spatial distribution of seed density in soil seed banks is also scale dependent due to topographic variation. Grassland management, therefore, needs to consider local grazing disturbance regime, spatial scale and topography.

Intensity-modulated radiation therapy with concurrent chemotherapy for locally advanced cervical and upper thoracic esophageal cancer

AIm： To evaluate the dosimetry, efficacy and toxicity of intensity-modulated radiation therapy （ImRT） and concurrent chemotherapy for patients with locally advanced cervical and upper thoracic esophageal cancer. mETHODS： A retrospective study was performed on 7 patients who were definitively treated with ImRT and concurrent chemotherapy. Patients who did not receive ImRT radiation and concurrent chemotherapy were not included in this analysis. ImRT plans were evaluated to assess the tumor coverage and normal tissue avoidance. Treatment response was evaluated and toxicities were assessed. RESULTS： Five- to nine-beam ImRT were used to deliver a total dose of 59.4-66 Gy （median： 64.8 Gy） to the primary tumor with 6-mV photons. The minimum dose received by the planning tumor volume （PTV） of the gross tumor volume boost was 91.2%-98.2% of the prescription dose （standard deviation [SD]： 3.7%-5.7%）. The minimum dose received by the PTV Of the clinical tumor volume was 93.8%-104.8% （SD： 4.3%-11.1%） of the prescribed dose. With a median follow-up of 15 rno （range： 3-21 too）, all 6 evaluable patients achieved complete response. Of them, 2 developed local recurrences and 2 had distant metastases, 3 survived with no evidence of disease. After treatment, 2 patients developed esophageal stricture requiring frequent dilation and 1 patient developed tracheal-esophageal fistula. CONCLUSION： Concurrent ImRT and chemotherapy resulted in an excellent early response in patients with locally advanced cervical and upper thoracic esophageal cancer. However, local and distant recurrence and toxicity remain to be a problem. Innovative approaches are needed to improve the outcome.
