A transgenic pig model expressing a CMV-ZsGreen1 reporter across an extensive array of tissues

Since genetic engineering of pigs can benefit both biomedicine and agriculture,selecting a suitable gene promoter is critically *** cytomegalovirus(CMV)promoter,which can robustly drive ubiquitous transgene expression,is commonly used at present,yet recent reports suggest tissue-specific activity in the *** objective of this study was to quantify ZsGreen1 protein(in lieu of CMV promoter activity)in tissues from pigs harboring a CMV-ZsGreen1 transgene with a single integration *** samples(n=35)were collected from neonatal hemizygous(n=3)and homozygous(n=3)piglets and ZsGreen1 abundance was determined via ***1 was detected in all tissues,except hypothalamus,kidney cortex and *** expression patterns of homozygous and hemizygous piglets were similar(P>0.05).However,quantification revealed that ZsGreen1 protein levels were *** neural/endocrine tissues,ZsGreen1 abundance was highest in the anterior pituitary gland,intermediate in the cerebellum and lowest in the cerebrum,spinal cord and posterior pituitary(Pthe digestive system,ZsGreen1 was more abundant in the salivary gland than esophagus,stomach,pancreas,duodenum,jejunum,ileum,spleen,colon,gallbladder and liver(Pterestingly,ZsGreen1 amounts also differed within an organ(i.e.,the right ventricle had 3-fold higher levels than the other heart chambers;Pthese results provide useful information for the use of the CMV promoter to drive transgene expression in the ***,this swine model represents a novel resource of ZsGreen1-labeled organs and a valuable tool to advance genome editing research.

COVID-19 posed challenges for global tourism *** in visitor temporal and spatial patterns and their associated determinants pre-and peri-pandemic in Canadian Rocky Mountain National Parks are *** was collected through social media programming and analyzed using spatiotemporal analysis and a geographically weighted regression(GWR)*** highlight that COVID-19 significantly changed park visitation *** tended to explore more remote areas *** GWR model also indicated distance to nearby trails was a significant influence on visitor *** results indicate that the pandemic influenced tourism temporal and spatial *** research presents a novel approach using combined social media big data which can be extended to the field of tourism management,and has important implications to manage visitor patterns and to allocate resources efficiently to satisfy multiple objectives of park management.

Bamboo plants are an essential component of tropical ecosystems,yet their vulnerability to climate extremes,such as drought,is poorly understood due to limited knowledge of their hydraulic *** pergracile,a commonly used tropical bamboo species,exhibited a substantially higher mortality rate than other co-occurring bamboos during a severe drought event in 2019,but the underlying mechanisms remain *** study investigated the leaf and stem hydraulic traits related to drought responses,including leaf-stem embolism resistance(P50leaf;P50stem) estimated using optical and X-ray microtomography methods,leaf pressure-volume and water-releasing ***,we investigated the seasonal water potentials,native embolism level(PLC) and xylem water source using stable *** found that *** exhibited strong resistance to embolism,showing low P50leaf,P50stem,and turgor loss point,despite its rapid leaf water ***,its leaves displayed greater resistance to embolism than its stem,suggesting a lack of effective hydraulic vulnerability segmentation(HVS) to protect the stem from excessive xylem *** the dry season,approximately 49% of the water was absorbed from the upper 20-cm-deep soil ***,significant diurnal variation in leaf water potentials and an increase in midday PLC from 5.87±2.33% in the wet season to 12.87±4.09%in the dry season were *** summary,this study demonstrated that the rapid leaf water loss,high reliance on surface water,and a lack of effective HVS in *** accelerated water depletion and increased xylem embolism even in the typical dry season,which may explain its high mortality rate during extreme drought events in 2019.

Contribution of NMDA receptors to dorsolateral prefrontal cortical networks in primates

Cognitive disorders such as schizophrenia and Alzheimer＇s disease are associated with dysfunction of the highly evolved dorsolateral prefrontal cortex （dlPFC）, and with changes in glutamatergic N-methyI-D-aspartate receptors （NMDARs）. Recent research on the primate dlPFC discovered that the pyramidal cell circuits that generate the persistent firing underlying spatial working memory communicate through synapses on spines containing NMDARs with NR2B subunits （GluN2B） in the post-synaptic density. this contrasts with synapses in the hippocampus and primary visual cortex, where GluN2B receptors are both synaptic and extrasynaptic. Blockade of GluN2B in the dlPFC markedly reduces the persistent firing of the Delay cells needed for neuronal representations of visual space. Cholinergic stimulation of nicotinic （17 receptors within the glutamate synapse is necessary for NMDAR actions. In contrast, a-amino-3-hydroxy-5-methyl-4-isoxazolepropionic acid receptors have only subtle effects on the persistent firing of Delay cells, but contribute substantially to the firing of Cue and Response cells. Systemic administration of the NMDAR antagonist ketamine reduces the persistent firing of Delay cells, but increases the firing of some Response cells. the reduction in persistent firing produced by ketamine may explain why this drug can mimic or worsen the cognitive symptoms of schizophrenia. Similar actions in the medial PFC circuits representing the emotional aspects of pain may contribute to the rapid analgesic and anti-depressant actions of ketamine.

While hypoxic signaling has been shown to play a role in many cellular processes,its role in metabolism-linked extracellular matrix(ECM)organization and downstream processes of cell fate after musculoskeletal injury remains to be ***(HO)is a debilitating condition where abnormal bone formation occurs within extra-skeletal *** andhypoxia-inducible factor 1α(HIF-1α)activation have been shown to promote ***,the underlying molecular mechanisms bywhich the HIF-1αpathway in mesenchymal progenitor cells(MPCs)contributes to pathologic bone formation remain to ***,we used a proven mouse injury-induced HO model to investigate the role of HIF-1αon aberrant cell ***-cell RNA sequencing(scRNA-seq)and spatial transcriptomics analyses of the HO site,we found that collagen ECM organizationis the most highly up-regulated biological process in *** mesenchymal cell-specific deletion of Hif1α(Hoxa11-CreER^(t2);Hif1a^(fl/fl))significantly mitigated HO in ***-seq analysis of these Hoxa11-CreER^(t2);Hif1a^(fl/fl)mice identified the PLOD2/LOXpathway for collagen cross-linking as downstream of the HIF-1αregulation of ***,our scRNA-seq data and mechanisticstudies further uncovered that glucose metabolism in MPCs is most highly impacted by HIF-1α*** a translational aspect,a pan-LOX inhibitor significantly decreased HO.A newly screened compound revealed that the inhibition of PLOD2 activity in MPCssignificantly decreased osteogenic differentiation and glycolytic *** suggests that the HIF-1α/PLOD2/LOX axis linked tometabolism regulates HO-forming MPC *** results suggest that the HIF-1α/PLOD2/LOX pathway represents a promisingstrategy to mitigate HO formation.

A global enterprise must continuously improve the efficiency of logistic operations between supply chain collaborators. Integrating logistic services, resources, and necessary information flows in the supply chain to ensure efficiency and efficacy is critically important to these companies. Global logistic service companies face challenges from their clients to provide logistic services that are cost effective, accurate, and seamlessly integrate material, information and cash flows. In this research, an improved framework for one-stop logistic services is systematically designed, analyzed, and evaluated. the one-stop logistic service framework, defmed in four models, is developed to provide enterprises with integrated and comprehensive services within the global supply chain context. the levels of service importance are assessed using a four categories questionnaire. this research provides a case study of the implementation of one-stop logistic services in the distribution industry and demonstrates the framework operating under different demand conditions. Finally, system dynamics causal evaluation is used to evaluate the advantages of the logistic service framework.

Simultaneous vs staged treatment of urolithiasis in patients undergoing radical prostatectomy

AIM: to assess the outcomes of men treated for urolithiasis at the time of radical ***: From 1991 to 2010, 22 patients were retrospectively identified who were treated simultaneously(n = 10) at radical prostatectomy, or(n = 12) within 120 d prior to prostatectomy, for urolithiasis. Clinical characteristics were reviewed including: type of prostatectomy and stone surgery, location and amount of stone burden, perioperative change in hemoglobin and creatinine, stent frequency, total hospital d, stone-free rates, additional stone procedures and complications. Long-term functional outcomes including stress urinary incontinence and bladder neck contracture were reported. Differences between cohorts(simultaneous vs staged treatment) were assessed. RESULtS: Among men undergoing radical prostatectomy, primary stone procedures included 12 ureteroscopy, 6 shock wave lithotripsy, 2 open nephrolithotomyand 2 percutaneous nephrolithotomy. In staged shock wave lithotripsy there were 4 complications and 3 additional procedures vs 1(P = 0.5) and 0(P = 0.2) in the simultaneous cohort. Meanwhile in staged ureteroscopy there were 5 complications and 1 additional procedure vs 1(P = 0.2) and 1(P = 0.9) in the simultaneous cohort. Additional procedures for residual stones was greater among patients with asymptomatic upper tract calculi 3(60%) relative to patients with symptomatic stones 2(13%; P = 0.02). Likewise, patients with proximal or multiple calculi had a greater total hospital days 5.5 vs 4.1(P = 0.04), additional procedures 6 vs 0(P = 0.04) and lower stone-free rates 39% vs 89%(P = 0.02) relative to men with distal stones. Finally, there was no difference in the incidence of bladder neck contracture(P = 0.4) or stress urinary incontinence(P = 0.7) between ***: Ureteroscopic treatment of symptomatic distal urolithiasis at radical prostatectomy appears to be safe and efficacious with a low rate of adverse postoperative outcomes.

Evacuating a First Nation Due to Wildfire Smoke:the Case of Dene tha’ First Nation

Almost every year,First Nations are evacuated in Canada because of wildfire proximity and *** of wildfires,and remote locations,unique sociocultural characteristics,and limited emergency management resources present challenges for evacuation organizers and *** study explores how Dene tha’First Nation evacuated their tachécommunity in July 2012 due to wildfire smoke and how the evacuation process affected *** were completed with 31 evacuation organizers and residents to examine the factors that helped and hindered the evacuation *** of information about the nearby wildfire,smoke,and evacuation of the nearby small community of Zama City,combined with a generic evacuation plan,delayed and posed challenges during the evacuation of this Dene tha’*** leadership and its role in community organizing,keeping families together,providing the social support they needed,and using familiar host communities,demonstrated and contributed to the community’s resilience during the *** to improve evacuations and emergency management in the community and other First Nations in Canada are identified and discussed.

Microscopic colitis:A large retrospective analysis from a health maintenance organization experience

AIM： to examine the demographic data on a large multi-ethnic population of patients with microscopic colitis （MC） in Southern California and to determine the association of MC with inflammatory bowel disease （IBD） and colorectal cancer. MEtHODS： All patients diagnosed with MC by co- Ionic biopsy from 1996-2005 were identified utilizing a pathology database. All biopsies were reviewed by experienced pathologists utilizing standard histologic criteria. Patients＇ medical records were reviewed and data regarding patient age, co-morbidities, sex, ethnic- ity, and medications were analyzed. An age- and sex- matched standard control group was also generated. Chi-square test was used to evaluate the associations of co-morbidities between lymphocytic colitis （LC）, col- lagenous colitis （CC） and the control group. RESULtS： A total of 547 cases of MC were identified, 376 patients with LC and 171 patients with CC. the fe- male/male ratio was 3：1 in CC and 2.7：1 in LC patients. Celiac disease （P 〈 0.001）, irritable bowel syndrome （IBS）（P 〈 0.001）, and thyroid diseases （P 〈 0.001） were found to have a higher occurrence in MC com- pared to the control group. No statistical differences in the occurrence of colorectal cancer, diabetes and IBD were found between the MC group and the control group. CONCLUSION： this is the largest group of patients with MC known to the authors that has been studied to date. Conditions such as celiac disease, IBS, and thyroid diseases were found to be related to MC. Fur- thermore, neither an increased risk of colorectal cancer nor IBD was associated with MC in this study.
