Novel wild edible mushroom Astraeus hygrometricus(Pers.)Morgan induces robust apoptosis on human acute lymphoblastic leukemia cells through a RONS-subsisted mitochondria-dependent pathway

Objective:To explore the therapeutic effects of the novel wild edible mushroom Astraeus hygrometricus(Pers.)Morgan(A.hygrometricus)on human acute lymphoblastic leukemia cells.Methods:Extensive screening of the antiproliferative and chemopreventive potential of different extracts from 5 wild mushrooms,A.hygrometricus,Phallus sp.,Lentinus sp.,Tricholoma sp.,and Serpula sp.was performed against a panel of 6 cancer cell lines and normal cells using the 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide(MTT)assay.Apoptosis determination,cell cycle profiling,intracellular reactive oxygen species(ROS)and reactive nitrogen species(RNS),and mitochondrial membrane potential were analyzed by flow cytometry.The activity of caspases was measured colorimetrically,and the expression pattern of mitochondrial proteins was analyzed.Results:The methanol extract of A.hygrometricus and MOLT-4 cells were identified as the most potent extract exhibiting antiproliferative activity and most sensitive cell line,respectively.The mushroom extract induced robust selective apoptosis in MOLT-4 cells and arrested cell cycle progression at the G0/G1 stage.The extract disrupted the mitochondrial membrane potential and enhanced ROS production in MOLT-4 cells.The methanol extract induced apoptosis by downregulating the expression of Bcl-2,increasing the expression of Bax,and activating the caspase cascade.Conclusion:The novel wild edible mushroom is a potential repository of biomolecules for the development of antileukemic drugs.

目标:探讨新型野生食用菌硬皮地星（Astraeus hygrometricus）对人急性淋巴细胞白血病细胞的治疗作用。方法：采用3-(4,5-二甲基噻唑-2)-2,5-二苯基四氮唑溴盐(MTT)试验，对5种野生菌类（硬皮地星、竹荪、香菇、口蘑和干朽菌）的提取物针对6种癌细胞系和正常细胞的抗增殖及化学预防的潜力进行了广泛筛选。采用流式细胞术分析细胞凋亡、细胞周期、细胞内活性氧(ROS)和活性氮(RNS)以及线粒体膜电位进行检测。采用比色法测定半胱天冬酶活性，分析线粒体蛋白的表达模式。结果：硬皮地星甲醇提取物的抗增殖活性最强，且MOLT-4细胞系最为敏感。菌类提取物可诱导MOLT-4细胞的强选择性凋亡，并在G0/G1期阻滞细胞周期进程。提取物可破坏线粒体膜电位，增强MOLT-4细胞的ROS生成。甲醇提取物通过下调Bcl-2的表达，增加Bax的表达，以及激活caspase级联反应来诱导细胞凋亡。结论：这种新型野生食用菌为抗白血病药物的开发提供了潜在的生物分子库。

Therapeutic neovascularization promoted by injectable hydrogels

The aim of therapeutic neovascularization is to repair ischemic tissues via formation of new blood vessels by delivery of angiogenic growth factors,stem cells or expansion of pre-existing cells.For efficient neovascularization,controlled release of growth factors is particularly necessary since bolus injection of molecules generally lead to a poor outcome due to inadequate retention within the injured site.In this regard,injectable hydrogels,made of natural,synthetic or hybrid biomaterials,have become a promising solution for efficient delivery of angiogenic factors or stem and progenitor cells for in situ tissue repair,regeneration and neovascularization.This review article will broadly discuss the state-of-the-art in the development of injectable hydrogels from natural and synthetic precursors,and their applications in ischemic tissue repair and wound healing.We will cover a wide range of in vitro and in vivo studies in testing the functionalities of the engineered injectable hydrogels in promoting tissue repair and neovascularization.We will also discuss some of the injectable hydrogels that exhibit self-healing properties by promoting neovascularization without the presence of angiogenic factors.

Sensing Behaviour of Some Nanocomposite Systems

Silver nanoparticles of diameters 3.4 to 13.2 nm grown at the interfaces between silicate glass and some oxide crystallites exhibited about six orders of magnitude reduction in resistivity for a relative humidity change from 25% to 80%. Sn-SnO2 nano core-shell structure prepared within a gel-derived silica glass film by electrodeposition technique followed by heat treatment showed large change in resistivity as a function of humidity. The resistivity also changed due to gas flow of CO2, C2H5OH and NO2, respectively. The latter arose because of reduction/oxidation of Sn4+/Sn2+ species present at the shell layer of the nanostructures. Nickel nanosheets of thickness ~0.6 nm grown within the interlayer spaces of Na-4 mica crystallites showed a change of dielectric permittivity (5%) for an applied magnetic field of 1.2 Tesla. An inhomogeneous model was used to explain this behavior. Two dimensional CuO phase was grown within the channels of diameter ~5 nm of mesoporous SiO2 structure. A magnetodielectric (MD) parameter M.D. of 4.4% was obtained in this case. BaTiO3 nanoparticles of diameter ~25 nm having pores with diameter 10 nm showed multiferroic behavior which arose due to the presence of oxygen vacancies as a result of large surface area present. An M.D. parameter of 11% was found. Similarly mesoporous LiNbO3 of 10 nm diameter showed an M.D. parameter of ~4.5% at a magnetic field 1 Tesla. A giant magnetocapacitance effect with a value of 44% at 1.5 T was observed in nickel zinc ferrite (NZF) impregnated mesoporous silica. A magnetocapacitance of 51% at magnetic field 1.7 T was found in the case of nanocomposites comprising of iron ion containing silica based nanoglass and mesoporous silica. In the last two examples the behavior was explained on the basis of Catalan model of space-charge polarization with extracted values of magnetoresistance of the NZF and nanoglass phases being 58%.

Determination of upper reference value of urinary calcium-creatinine ratio for the paediatric population in Burdwan district

To estimate the rate of excretion of urinary calcium, a 24-hour sample of urine is required and this is not always easy to collect accurately in infant and children. So, random urine calcium to creatinine ratio (Ca/Cr ratio) has been developed. But as the ratio varies worldwide, reference values of the parameter in paediatric population are not developed. To determine reference value, the present study was conducted in healthy paediatric population in Burdwan district, West Bengal. This study was performed on 693 healthy paediatric subjects, aged between 3 months to 18 years and divided into five groups. Early morning non-fasting urine samples from all study groups were analyzed for Ca/Cr ratio. A negative correlation was observed between age and urinary Ca/Cr ratio, but there was no significant difference of urinary Ca/Cr ratio between two sexes. Considering 97.5th percentile of the underlying distribution of values as the upper limit of reference range, upper reference values of urinary Ca/Cr ratio for age groups
