Assessing phylogenetic relationships of Lycium samples using RAPD and entropy theory

Aim: To evaluate the phylogenetic relationships among related species of Lycium samples. Methods: Random amplified polymorphic DNA (RAPD) fingerprinting and lab-on-a-chip electrophoresis techniques were used to analyze the characteristics of Lycium species. Seven species and 3 varieties of Lycium were studied. Based on RAPD fingerprint data obtained from 11 primers, we proposed a new index, called dispersivity, using entropy theory and projection methods to depict the diversity of the DNA fingerprints. Results: Using the proposed dispersivity, primers were sorted and the dendrograms of the 7 species and 3 varieties of Lycium were constructed synthetically by merging primer information. Conclusion:Phylogenetic relationships among Lycium samples were constructed synthetically based on RAPD fingerprint data generated from 11 primers.

Mechanistic links between gut microbial community dynamics, microbial functions and metabolic health

Gut microbes comprise a high density, biologically active community that lies at the interface of an animal with its nutritional environment. Consequently their activity profoundly influences many aspects of the physiology and metabolism of the host animal. A range of microbial structural components and metabolites directly interact with host intestinal cells and tissues to influence nutrient uptake and epithelial health. Endocrine, neuronal and lymphoid cells in the gut also integrate signals from these microbial factors to influence systemic responses. Dysregulation of these host-microbe interactions is now recognised as a major risk factor in the development of metabolic dysfunction. This is a two-way process and understanding the factors that tip host-microbiome homeostasis over to dysbiosis requires greater appreciation of the host feedbacks that contribute to regulation of microbial community composition. To date, numerous studies have employed taxonomic profiling approaches to explore the links between microbial composition and host outcomes(especially obesity and its comorbidities), but inconsistent host-microbe associations have been reported. Available data indicates multiple factors have contributed to discrepancies between studies. These include the high level of functional redundancy in host-microbiome interactions combined with individual variation in microbiome composition; differences in study design, diet composition and host system between studies; and inherent limitations to the resolution of r RNA-based community profiling. Accounting for these factors allows for recognition of the common microbial and host factors driving community composition and development of dysbiosis on high fat diets. New therapeutic intervention options are now emerging.

Water and Nitrogen Management Effects on Biomass Accumulation and Partitioning in Two Potato Cultivars

Biomass accumulation and partitioning into different plant parts is a dynamic process during the plant growing period, which is influenced by crop management and climate factors. Adequate knowledge of biomass partitioning is important to manage the crops to gain maximum partitioning of assimilates into plant parts of economic significance, i.e. tubers in potato. This study was conducted using two potato cultivars grown in a sandy soil with center pivot irrigation under full irrigation (FI;irrigation to replenish 100% of water loss by evapotranspiration [ET]) and deficit irrigation (DI;replenish only 80% ET) and two nitrogen(N) rates (pre-plant + in-seasonN rates of 56 + 112 or 168 + 336 kg/ha). Plant samples were taken on 22, 44, 66, and 98 days after seedling emergence (DAE). With high N rate, tuber biomass of ‘Umatilla Russet’ cultivar in relation to total plant biomass varied from 23% - 88% and 25% - 86% over 22 to 98 DAE for the FI and DI treatments, respectively. The corresponding partitioning ranges were 30% - 93% and 38% - 93% at the low N rate. With respect to the‘Ranger Russet’ cultivar, biomass partitioning to tubers ranged from 36% - 82% and 23% - 84% for the FI and DI, respectively, at the high N rate, and 29% - 87% and 39% - 95% at the low N rate. Overall, this study demonstrated that within the range of N rate and irrigation treatments the biomass portioning into tubers was largely similar in both cultivars.

目的:测量糖尿病性视网膜病变(DR)行玻璃体切除术(PPV)患者玻璃体样品中白细胞介素-8(IL-8),肿瘤坏死因子-α(TNF-α)的水平,与对照组比较结果并探讨其对DR的影响。方法:将伊斯坦布尔Bilim大学眼科系的57例(57眼)DR患者和22例黄斑裂孔未伴有增生性玻璃体视网膜病变患者分为研究组和对照组。对所有79例患者行3切口,20G PPV。PPV手术眼内灌注前,用Vitrector抽吸0.5mL玻璃体样品并稀释。样品转至冷藏室,存放在-70℃。IL-8和TNF-α的测量结果用酶联免疫吸附法以pg/mL为单位计算。结果:DR患者玻璃体样本中IL-8水平[82.7891±74.08700(0.08-307.09)pg/mL]明显高于对照组患者[2.9805±3.77546(0.08-18.53)pg/mL](P<0.001)。同样,DR患者TNF-α水平[18.0007±13.90015(2.32-51.11)pg/mL]也显着高于对照组[1.7005±1.26949(0.1-5.17)pg/mL](P<0.001)。结论:在视网膜新生血管形成中起重要作用的TNF-α和作为炎症和血管生成介体的IL-8水平在DR患者玻璃体样本中明显高。
