C-reactive protein and telomerase reverse transcriptase(TERT)associate with chronic disease markers in a sample from low-income neighborhoods in Detroit,Michigan

Racial and ethnic minorities in economically deprived inner cities experience high rates of chronic diseases compared to neighborhoods with higher socioeconomic status(SES).However,these economically deprived populations are understudied in terms of biomarkers associated with chronic disease risk which include C-reactive protein(CRP),telomerase reverse transcriptase(TERT),and glycosylated hemoglobin(A1C).We examined relationships between CRP and TERT and chronic disease indicators(body mass index[BMI]and A1C)in two lowincome,predominantly African American(AA)neighborhoods in Detroit,***-nine adults(43 females,26 males,mean age 46 years[y],standard deviation[SD]=15.9)completed a health survey,anthropometry,and finger stick blood tests.A1C was measured using A1CNow test strips,and CRP and TERT levels were measured using enzyme-linked immunosorbent assay(ElISA)with samples extracted from dried blood *** examined CRP(mean=4.9,SD=3.1),TERT(mean=32.5,SD=15.1),and A1C(mean=5.4,SD=1.0)by BMI *** fitted restricted maximum likelihood regression models to evaluate associations between CRP,TERT,BMI,and A1C,after adjustment for demographics and inclusion of a random effect for the *** this predominantly AA sample(91%,63/69),68%had levels of CRP(means=4.8 mg/l,SD=3.0 for AAs;6.4 mg/l,SD=3.9 for all others)indicative of chronic inflammation(CRP greater than 3 mg/l).BMI was significantly associated with CRP(p=0.004)and TERT(p=0.026).TERT levels indicate that being overweight is associated with markers of chromosome remodeling,suggestive of chronic *** followed a similar trend with overweight individuals having higher inflammation and risk of chronic *** findings warrant further exploration of additional factors that may influence CRP and ***,examining populations in a more ethnically and/or economically diverse,yet still high proportion minority,sample will fill a knowledge gap in this understudied field.

Decision-Making and Management of Self-Care in Persons with Traumatic Spinal Cord Injuries: A Preliminary Study

Patients and physicians understand the importance of self-care following spinal cord injury (SCI), yet many individuals with SCI do not adhere to recommended self-care activities despite logistical supports. Neurobehavioral determinants of SCI self-care behavior, such as impulsivity, are not widely studied, yet understanding them could inform efforts to improve SCI self-care. We explored associations between impulsivity and self-care in an observational study of 35 US adults age 18 - 50 who had traumatic SCI with paraplegia at least six months before assessment. The primary outcome measure was self-reported self-care. In lASSO regression models that included all neurobehavioral measures and demographics as predictors of self-care, dispositional measures of greater impulsivity (negative urgency, lack of premeditation, lack of perseverance), and reduced mindfulness were associated with reduced self-care. Outcome (magnitude) sensitivity, a latent decision-making parameter derived from computationally modeling successive choices in a gambling task, was also associated with self-care behavior. These results are preliminary;more research is needed to demonstrate the utility of these findings in clinical settings. Information about associations between impulsivity and poor self-care in people with SCI could guide the development of interventions to improve SCI self-care and help patients with elevated risks related to self-care and secondary health conditions.

Disease-resistance(R)gene cloning in wheat(Triticum aestivum)has been accelerated by the recent surge of genomic resources,facilitated by advances in sequencing technologies and ***,with the challenges of population growth and climate change,it is vital not only to clone and functionally characterize a few handfuls of R genes,but also to do so at a scale that would facilitate the breeding and deployment of crops that can recognize the wide range of pathogen effectors that threaten *** populations are continually changing,and breeders must have tools and resources available to rapidly respond to those changes if we are to safeguard our daily *** meet this challenge,we propose the creation of a wheat R-gene atlas by an international community of researchers and *** atlas would consist of an online directory from which sources of resistance could be identified and deployed to achieve more durable resistance to the major wheat pathogens,such as wheat rusts,blotch diseases,powdery mildew,and wheat *** present a costed proposal detailing how the inter-acting molecular components governing disease resistance could be captured from both the host and the pathogen through biparental mapping,mutational genomics,and whole-genome association *** explore options for the configuration and genotyping of diversity panels of hexaploid and tetraploid wheat,as well as their wild relatives and major pathogens,and discuss how the atlas could inform a dynamic,durable approach to R-gene *** against the current magnitude of wheat yield losses worldwide,recently estimated at 21%,this endeavor presents one route for bringing R genes from the lab to the field at a considerable speed and quantity.

SlC11A1 polymorphisms in inflammatory bowel disease and Mycobacterium avium subspecies paratuberculosis status

AIM: To test for association of SlC11A1 with inflammatory bowel disease (IBD) and Mycobacterium avium subspecies paratuberculosis (MAP) status in a Caucasian cohort. METHODS: Five hundred and seven Crohn's disease (CD) patients, 474 ulcerative colitis (UC) patients, and 569 healthy controls were genotyped for SlC11A1 1730G>A and SlC11A1 469+14G>C using pre-designed TaqMan  SNP assays. χ 2 tests were applied to test for association of single nucleotide polymorphisms (SNPs) with disease, and the presence of MAP ***:SlC11A1 1730G>A and SlC11A 1469+14G>C were not associated with CD, UC, or IBD. The SlC11A1 1730A minor allele was over-represented in patients who did not require immunomodulator therapy (P = 0.002, OR: 0.29, 95% CI: 0.13-0.66). The frequency of the SlC11A1 469+14C allele was higher in the subset of study participants who tested positive for MAP DNA (P = 0.02, OR: 1.56, 95% CI: 1.06-2.29). No association of SlC11A1 1730G>A with MAP was ***: Although SlC11A1 was not associated with IBD, association with MAP suggests that SlC11A1 is important in determining susceptibility to bacteria implicated in the etiology of CD.

AIM： TO assess whether polymorphisms in NOD2 and ATG16l1 affect cytokine responses and mycobacterium avium subspecies paratuberculosis （MAP） survival in monocytes from Crohn＇s disease （CD） patients METHODS： Monocytes were isolated from peripheral blood of CD patients of known genotype for common single nucleotide polymorphisms of NOD2 and ATG16l1, Monocytes were challenged with MAP and bacterial per- sistence assessed at subsequent time-points. Cytokine responses were assayed using a Milliplex multi-analyte profiling assay for 13 cytokines. RESUlTS： Monooltes heterozygous for a NOD2 polymorphism （R702W, P268S, or 1007fs） were more permissive for growth of MAP （P = 0.045） than those without. There was no effect of NOD2 genotype on subsequent cytokine expression. The T300A polymorphism of ATG16l1 did not affect growth of MAP in our model （P = 0.175）, but did increase expression of cytokines interleukin （Il）-10 （P = 0.047） and Il-6 （P = 0.019）. CONClUSION： CD-associated polymorphisms affected the eliminaUon of MAP from ex v/vo monooltes （NOD2）, or expression of certain oltokines （ATG16lI）, implying independent but contributory roles in the pathogenesis of CD.

Temporal changes in inflammatory mitochondria-enriched microRNAs following traumatic brain injury and effects of miR-146a nanoparticle delivery

BACKGROUND Autoimmunity has emerged as a probable disease modifier in patients with clinically diagnosed type 2 diabetes mellitus(T2DM),that is,patients who have insulin resistance,obesity,and other cardiovascular risk factors,suggesting that the presence of glutamic acid decarboxylase(anti-GAD65),islet antigen 2(anti-IA2),and zinc transporter 8(anti-Zn8T)antibodies could have deleterious effects on beta cell function,causing failure and earlier requirement for insulin *** To evaluate anti-GAD65,anti-IA2 and anti-Zn8T as predictors of early insulin requirement in adolescents with a clinical diagnosis of *** This was a case–control study in patients with clinically diagnosed with T2DM(68 cases and 64 controls with and without early insulin dependence respectively),male and female,aged 12–18 ***,blood pressure,glucose,insulin,C-peptide,glycated hemoglobin A1c,and lipid profiles were *** was used to measure anti-GAD65,anti-IA2,and anti-Zn8T *** statistics,pearson'sχ2 test,Student's t test,and logistic regression was performed.Plly *** There were 132 patients(53.8%female),with a mean age was 15.9±1.3 years,and there was a disease evolution time of 4.49±0.88 *** presence of anti-GAD65,anti-IA2,and anti-Zn8T positivity was found in 29.5%,18.2%,and 15.9%,*** the groups by early or no insulin dependence showed that the group with insulin had a higher frequency of antibody positivity:anti-GAD65 odds ratio(OR):2.42(1.112–5.303,P=0.026);anti-IA2:OR:1.55(0.859–2.818,P=0.105);and anti-Zn8T:OR:7.32(2.039–26.279,P=0.002).CONClUSION Anti-GAD65 positivity was high in our ***-GAD65 and anti-Zn8T positivity showed a significantly depleted beta cell reserve phenotype,leading to an increased risk of early insulin dependence.
