IL-6 prevents Th2 cell polarization by promoting SOCS3-dependent suppression of IL-2 signaling

Defective interleukin-6 (IL-6) signaling has been associated with Th2 bias and elevated IgE levels. However, the underlying mechanism by which IL-6 prevents the development of Th2-driven diseases remains unknown. Using a model of house dust mite (HDM)-induced Th2 cell differentiation and allergic airway inflammation, we showed that IL-6 signaling in allergen-specific T cells was required to prevent Th2 cell differentiation and the subsequent IgE response and allergic inflammation. Th2 cell lineage commitment required strong sustained IL-2 signaling. We found that IL-6 turned off IL-2 signaling during early T-cell activation and thus inhibited Th2 priming. Mechanistically, IL-6-driven inhibition of IL-2 signaling in responding T cells was mediated by upregulation of Suppressor Of Cytokine Signaling 3 (SOCS3). This mechanism could be mimicked by pharmacological Janus Kinase-1 (JAK1) inhibition. Collectively, our results identify an unrecognized mechanism that prevents the development of unwanted Th2 cell responses and associated diseases and outline potential preventive interventions.

Introduction:Climate factors play an important role in the transmission of viruses,such as influenza viruses,MERS-CoV,and *** study aimed to determine the relationship between changes in temperature,humidity,rainfall,and SARS-CoV-2 *** ecologically and climatically distinct regions were considered—Karachi,Lahore,Islamabad,Peshawar,and ***:Data on daily COVID-19 cases and deaths were retrieved from government officials,while meteorological information was collected from Pakistan Meteorological *** analysis was performed using SPSS version 20 and the Spearman rank correlation test was used to analyze the correlation between the meteorological factors and COVID-19 cases and ***:Positive correlation of COVID-19 incidence was observed with all the temperature ranges(maximum,minimum and average)and negative correlation was seen with humidity,DTR and ***-19 deaths were positively associated with temperature and were negatively associated only with *** regression showed that for every unit increase in humidity,there was a3.345 daily signifi-cant decrease in COVID-19 cases,while in Karachi for every unit increase in humidity,there remained a 10.104 daily significant increase in *** Gilgit-Baltistan,for every unit increase in average temperature and rainfall respectively,significant increases of 0.534 and 1.286 in daily cases were ***:This study signifies the effect of climate factors on COVID-19 incidence and mortality rate,but climate factors are not the only variable and several other interlinked factors enhance the spread of ***,effective mitigation policies,enhancing testing capacities,and developing public attitudes toward adopting precautionary measures are important to overcome this overwhelming pandemic.

In this paper, we propose a novel method to enhance the OCR (Optical Character Recognition) readability of public signboards captured by smart-phone cameras—both outdoors and indoors, and subject to various lighting conditions. A distinct feature of our technique is the detection of these signs in the HSV (Hue, Saturation and Value) color space, done in order to filter out the signboard from the background, and correctly interpret the textual details of each signboard. This is then binarized using a thresholding technique that is optimized for text printed on contrasting backgrounds, and passed through the Tesseract engine to detect individual characters. We test out our technique on a dataset of over 200 images taken in and around the campus of our college, and are successful in attaining better OCR results in comparison to traditional methods. Further, we suggest the utilization of a method to automatically assign ROIs (Regions Of Interest) to detected signboards, for better recognition of textual information.

Role of Clonal Integration among Different Environmental Conditions (A Review)

Most of the ecosystems are dominated by clonal species. The most unique feature of clonal plants is their capability for clonal integration (translocation of vital resources among connected ramets), implying that integration may play an essential role in their success. However, a general effect of clonal integration on plant performance is lacking. We conducted a text review on the effects of clonal integration on different habitats and species. Overall, clonal integration increased performance of clonal plants in different habitats. However clonal integration has also some limitations under stressful environments. Benefits of clonal integration may lack somehow when environmental stress increases. But connected ramets placed in unfavorable patches benefited more from integration compared to severed ramet placed in nutrient rich patches. Climate change and temperature increase have positive effects on biomass of clonal species.

AIM: To test antibiotic-loaded coating for efficacy in reducing bacterial biofilm and development of osteomyelitis in an orthopaedic model of implant infection. METHODS: Phosphatidylcholine coatings loaded with 25% vancomycin were applied to washed and sterilized titanium wires 20 mm in length. A 10 mm segment was removed from rabbit radius(total = 9; 5 coated, 4 uncoated), and the segment was injected with 1 × 106 colony forming units(CFUs) of Staphylococcus aureus(UAMS-1 strain). Titanium wires were inserted throughthe intramedullary canal of the removed segment and into the proximal radial segment and the segment was placed back into the defect. After 7 d, limbs were removed, X-rayed, swabbed for tissue contamination. Wires were removed and processed to determine attached CFUs. Tissue was swabbed and streaked on agar plates to determine bacteriological ***: Antibiotic-loaded coatings resulted in significantly reduced biofilm formation(4.7 fold reduction in CFUs; P < 0.001) on titanium wires and reduced bacteriological score in surrounding tissue(4.0 ± 0 for uncoated, 1.25 ± 0.5 for coated; P = 0.01). Swelling and pus formation was evident in uncoated controls at the 7 d time point both visually and radiographically, but not in antibiotic-loaded ***: Active antibiotic was released from coated implants and significantly reduced signs of osteomyelitic symptoms. Implant coatings were well tolerated in bone. Further studies with additional control groups and longer time periods are warranted. Antibiotic-loaded phosphatidylcholine coatings applied at the point of care could prevent implant-associated infection in orthopaedic defects.

Anti-yeast activities of Origanum oil against human pathogenic yeasts

Outbreak of autoimmune diseases by pathogenic yeasts has led to a serious medical threat. As these organisms evolve resistance to existing antifungal drugs, the concern could be further compounded. The realm of plant derived products offers a wide spectrum of potentially valuable alternatives to the existing synthetic fungicides. Essential oils from sev-eral medicinal plants have been shown to exhibit pharmacological attributes. In the present study, anti-yeast properties of Oregano essential oil (OEO) were examined in vitro against four human patho-genic yeasts i.e., Candida albicans, Cryptococcus al-bidus, Cryptococcus neoformans and Rhodotorula ru-brum. OEO concentration of 200 μg/mL was found to be growth inhibitory against all four yeasts examined, thereby showing its potential to function as a natural anti-yeast agent.

Water and Nitrogen Management Effects on Biomass Accumulation and Partitioning in Two Potato Cultivars

Biomass accumulation and partitioning into different plant parts is a dynamic process during the plant growing period, which is influenced by crop management and climate factors. Adequate knowledge of biomass partitioning is important to manage the crops to gain maximum partitioning of assimilates into plant parts of economic significance, i.e. tubers in potato. This study was conducted using two potato cultivars grown in a sandy soil with center pivot irrigation under full irrigation (FI;irrigation to replenish 100% of water loss by evapotranspiration [ET]) and deficit irrigation (DI;replenish only 80% ET) and two nitrogen(N) rates (pre-plant + in-seasonN rates of 56 + 112 or 168 + 336 kg/ha). Plant samples were taken on 22, 44, 66, and 98 days after seedling emergence (DAE). With high N rate, tuber biomass of ‘Umatilla Russet’ cultivar in relation to total plant biomass varied from 23% - 88% and 25% - 86% over 22 to 98 DAE for the FI and DI treatments, respectively. The corresponding partitioning ranges were 30% - 93% and 38% - 93% at the low N rate. With respect to the‘Ranger Russet’ cultivar, biomass partitioning to tubers ranged from 36% - 82% and 23% - 84% for the FI and DI, respectively, at the high N rate, and 29% - 87% and 39% - 95% at the low N rate. Overall, this study demonstrated that within the range of N rate and irrigation treatments the biomass portioning into tubers was largely similar in both cultivars.

We show three-dimensional reconstructions of a region of an integrated circuit from a 130 nm copper *** reconstructions employ x-ray computed tomography,measured with a new and innovative high-magnification x-ray *** instrument uses a focused electron beam to generate x-rays in a 100 nm spot and energy-resolving x-ray detectors that minimize backgrounds and hold promise for the identification of materials within the *** x-ray generation target,a layer of platinum,is fabricated on the circuit wafer itself.A region of interest is imaged from a limited range of angles and without physically removing the region from the larger *** reconstruction is consistent with the circuit's design file.
