Selective serotonin re-uptake inhibitor sertraline inhibits bone healing in a calvarial defect model

Bone wound healing is a highly dynamic and precisely controlled process through which damaged bone undergoes repair and complete regeneration. External factors can alter this process, leading to delayed or failed bone wound healing. The findings of recent studies suggest that the use of selective serotonin reuptake inhibitors(SSRIs) can reduce bone mass, precipitate osteoporotic fractures and increase the rate of dental implant failure. With 10% of Americans prescribed antidepressants, the potential of SSRIs to impair bone healing may adversely affect millions of patients’ ability to heal after sustaining trauma. Here, we investigate the effect of the SSRI sertraline on bone healing through pre-treatment with(10 mg·kg-1sertraline in drinking water, n = 26) or without(control, n = 30) SSRI followed by the creation of a 5-mm calvarial defect. Animals were randomized into three surgical groups:(a) empty/sham,(b) implanted with a DermaMatrix scaffold soak-loaded with sterile PBS or(c) DermaMatrix soak-loaded with542.5 ng BMP2. SSRI exposure continued until sacrifice in the exposed groups at 4 weeks after surgery. Sertraline exposure resulted in decreased bone healing with significant decreases in trabecular thickness, trabecular number and osteoclast dysfunction while significantly increasing mature collagen fiber formation. These findings indicate that sertraline exposure can impair bone wound healing through disruption of bone repair and regeneration while promoting or defaulting to scar formation within the defect site.

Blood-brain barrier pathology in cerebral small vessel disease

cerebral small vessel disease is a neurological disease that affects the brain microvasculature and which is commonly observed among the elderly. Although at first it was considered innocuous, small vessel disease is nowadays regarded as one of the major vascular causes of dementia. Radiological signs of small vessel disease include small subcortical infarcts, white matter magnetic resonance imaging hyperintensities, lacunes, enlarged perivascular spaces, cerebral microbleeds, and brain atrophy; however, great heterogeneity in clinical symptoms is observed in small vessel disease patients. The pathophysiology of these lesions has been linked to multiple processes, such as hypoperfusion, defective cerebrovascular reactivity, and blood-brain barrier dysfunction. Notably, studies on small vessel disease suggest that blood-brain barrier dysfunction is among the earliest mechanisms in small vessel disease and might contribute to the development of the hallmarks of small vessel disease. Therefore, the purpose of this review is to provide a new foundation in the study of small vessel disease pathology. First, we discuss the main structural domains and functions of the blood-brain barrier. Secondly, we review the most recent evidence on blood-brain barrier dysfunction linked to small vessel disease. Finally, we conclude with a discussion on future perspectives and propose potential treatment targets and interventions.

The Meat of the Matter: Plant-Based Meat Analogue versus Traditional Meat Product Nutritional Quality

Plant-based meat analogues (PBMA) are formulated to mimic the sensory characteristics of traditional meat products (TMP) using vegetarian ingredients and are increasingly popular with the trend towards plant-based diets， even among meat-eating consumers. This narrative review compared the nutritional quality of PBMA and TMP to broaden the discussion on the suitability of simulated meat products to substitute TMP， while exploring other attributes for their comparison. The complexity of PBMA and TMP means they are not entirely interchangeable. Importantly， PBMA health effects extend beyond their basic nutrient content and quality. Postprandial metabolism and satiety， for example， can be affected by product formulation， ingredient interactions， and thermomechanical processing involved in producing organoleptically acceptable PBMA. There are specific opportunities for low-sodium PBMA products to contribute to a healthy shift toward plant-based diets. In conclusion， PBMA cannot be assumed to be healthier than TMP. They should be designed with comprehensive consideration of composition and processing to ensure they support consumers who are adopting plant-based diets in realizing the theoretical health benefits.

Metal-halide perovskites are revolutionizing the world of X-ray detectors,due to the development of sensitive,fast,and cost-effective ***-powered operation,ensuring portability and low power consumption,has also been recently demonstrated in both bulk materials and thin ***,the signal stability and repeatability under continuous X-ray exposure has only been tested up to a few hours,often reporting degradation of the detection *** it is shown that self-powered direct X-ray detectors,fabricated starting from a FAPbBr_(3)submicrometer-thick film deposition onto a mesoporous TiO_(2)scaffold,can withstand a 26-day uninterrupted X-ray exposure with negligible signal loss,demonstrating ultra-high operational stability and excellent *** structural modification is observed after irradiation with a total ionizing dose of almost 200 Gy,revealing an unexpectedly high radiation hardness for a metal-halide perovskite thin *** addition,trap-assisted photoconductive gain enabled the device to achieve a record bulk sensitivity of 7.28 c Gy^(−1)cm^(−3)at 0 V,an unprecedented value in the field of thin-film-based photoconductors and photodiodes for“hard”***,prototypal validation under the X-ray beam produced by a medical linear accelerator for cancer treatment is also introduced.

The use of Vis-NIR-SWIR spectroscopy in the prediction of soil available ions after application of rock powder

Some of the problems attributed to traditional laboratory analyses that limit the correct assessment of the nutrient contents in the soil are time requirements and high cost of the soil nutrient *** solve these problems,a study was carried out to evaluate the use of visible,near-infrared,and short-wave infrared(Vis-NIR-SWIR)spectroscopy in the prediction of soil available ions submitted to the application of rock *** study was carried out on an Arenosol in Paranavaícity/***(rock powders)were arranged within a split-plot system designed in randomized blocks with four *** was cultivated for 14 months after the application of rock ***,96 soil samples were collected for measuring the pH and available ions P,K^(+),ca^(2+),Mg^(2+),S-SO_(4)^(2-),Si,cu^(2+),Fe^(2+),Mn^(2+),and Zn^(2+)as well as spectral reading through a Vis-NIR-SWIR spectroradiometer to predict the soil chemical attributes through the partial least square regression(PLS)*** results showed that the elements K^(+),ca^(2+),Mg^(2+),cu^(2+),and Fe^(2+)could be predicted with a reasonable rightness degree(R^(2)_(p)>0.50,RPDp>1.40)from spectral ***,for the attributes pH,P,S-SO_(4)^(2-),Si,Mn^(2+),and Zn^(2+),there were no satisfactory models(R^(2)_(p)cation of rock powder changed the spectral curves and,because of that,allows the building of PLS models to predict the elements K^(+),ca^(2+),Mg^(2+),cu^(2+),and Fe^(2+).Therefore,Vis-NIR-SWIR spectroscopy is a promising alternative to the routine analyses of soil fertility since it has advantages such as fast analytical speed,low cost,easy to operate,non-destructive,and environmentally friendly,because it does not use harmful chemicals.

Effect of β-Glucan (Angel Yeast) compared to a Placebo on cold and Flu Incidence and Symptoms in an Adult Population—A Double Blind, Randomised controlled Trial

Background: 1-3, 1-6 β-glucan derived from Baker’s yeast (Saccharomyces cerevisiae) has been widely studied for its immune stimulatory capabilities and safety. Previous studies found β-glucan to have efficacy at reducing incidence of URTIs as well as being a low risk for negative side effects. The current study aimed to examine the effects of yeast β-glucan (Angel Yeast) on cold and flu incidences and symptoms in healthy adults. Methods: Two hundred and thirty-one males and females aged 18 to 65 years old supplemented with either β-glucan or a placebo for 3-months. Participants completed a general health questionnaire every 4 weeks and in addition, if participants experienced any cold or flu symptoms, these were recorded daily (along with severity) until resolved or up to 2 weeks. Results: Supplementation with β-glucan reduced the self-reported severity of sore throats and improved sleep quality compared to the placebo group. conclusions: Yeast β-glucan supplementation appears to be able to help reduce certain symptoms experienced during a cold or flu episode and is safe and well tolerated.

Wild Nutrition’s Food-Grown® Magnesium Supplementation Increases Sleep Quality and Sleep Duration and Reduces Stress in a Healthy Adult Population: A Double-Blind, Randomised, Placebo-controlled Study

Background: Magnesium, an essential mineral crucial for various bodily functions, has been shown to positively influence sleep patterns. This study aimed to evaluate the efficacy of Food-Grown® magnesium in enhancing sleep quality and duration, as well as overall well-being. Methods: Eighty participants were randomly assigned to receive either 80 mg of Food-Grown® magnesium or a placebo (microcrystalline cellulose) daily for 8 weeks. Participants completed questionnaires assessing sleep quality, daytime drowsiness, quality of life, anxiety, and stress levels. Additionally, participants maintained daily sleep diaries and wore wrist-worn actigraphy devices. The primary outcome measured was the change in sleep quality and duration. Results: Seventy-one participants fulfilled all study requirements (35 in the active group and 36 in the placebo group). Magnesium supplementation significantly improved reported sleep quality, with the active group showing a 32% increase compared to 16% in the placebo group (p = 0.034). Moreover, magnesium supplementation led to a decrease in reported stress scores at week 8 compared to the placebo group (3.7 ± 2.6 vs. 5.5 ± 3.1, respectively). Both the magnesium and placebo groups exhibited significant increases in reported sleep duration and reductions in time to fall asleep, sleep disturbance, sleep latency, sleep medication usage, and total Pittsburgh Sleep Quality Index score at week 8 compared to baseline. conclusion: Magnesium supplementation notably enhanced sleep quality and reduced stress levels compared to the placebo group. These findings highlight the potential of magnesium as a beneficial supplement for improving sleep quality and overall well-being.
