Expression of Angiopoietin-1, 2 and 4 and Tie-1 and 2 in gastrointestinal stromal tumor, leiomyoma and schwannoma

AIM: To investigate the role of angiopoietin (Ang) -1, -2 and -4 and its receptors, Tie-1 and -2, in the growth and differentiation of gastrointestinal stromal tumors (GISTs).METHODS: Thirty GISTs, seventeen leiomyomas and six schwannomas were examined by immunohistochemistry in this ***: Ang-1, -2 and -4 proteins were expressed in the cytoplasm of tumor cells, and Tie-1 and -2 were expressed both in the cytoplasm and on the membrane of all tumors. Immunohistochemical staining revealed that 66.7% of GISTs (20 of 30), 76.5% of leiomyomas (13 of 17) and 83.3% of schwannomas (5 of 6) were positive for Ang-1. 83.3% of GISTs (25 of 30), 82.4% of leiomyomas (14 of 17) and 100% of schwannomas (6 of 6) were positive for Ang-2. 36.7% of GISTs (11 of 30), 58.8% of leiomyomas (10 of 17) and 83.3% of schwannomas (5 of 6) were positive for Ang-4. 60.0% of GISTs (18 of 30), 82.4% of leiomyomas and 100% of schwannomas (6 of 6) were positive for Tie-1. 10.0% of GISTs (3 of 30), 94.1% of leiomyomas (16 of 17) and 33.3% of schwannomas (2 of 6) were positive for Tie-2. Tie-2 expression was statistically different between GISTs and leiomyomas (P < 0.001). However, there was no correlation between expression of angiopoietin pathway components and clinical risk ***: Our results suggest that the angiopoietin pathway plays an important role in the differentiation of GISTs, leiomyomas and schwannomas.

Objective:To investigate the effects of soft agar on in-vitro proliferation of neonate mouse spermatogonial stem cells co-cultured with Sertoli ***:Tissues of neonate NMRI male mice testes were used for harvesting spermatogonial stem cells and Sertoli *** cell harvest,flow cytometry using promyelocytic leukemia zinc-finger(PLZF)protein antibody was used to assess the purity of the *** isolated testicular cells were cultured in the absence(the control group)or presence of soft agar-coated dishes(the experimental group)supplemented with leukemia inhibitory factor and glia cell line–derived neurotrophic factor for two *** phosphatase activity was assessed in the colonies formed after two weeks of culture by alkaline phosphatase *** day 14 of culture,the expression levels of DNA-binding protein inhibitor(ID-4)and PLZF genes in the undifferentiated cells were evaluated by the detection of PLZF protein antibody using real-time PCR and immunocytochemistry *** number and diameter of the colonies of spermatogonial stem cells were assessed by ImageJ ***:In the experimental group,the number and the diameter of colonies significantly increased as compared with those in the control group(P<0.05,P<0.01,respectively).In addition,the level of expression of ID-4 and PLZF genes in the undifferentiated cells significantly increased in the experimental group as compared with the control group(P<0.01).However,the expression of tyrosine-protein kinase kit(c-kit)gene in differentiated cells decreased in the experimental group as compared with the control group,but there was no significant difference between the two ***:Spermatogonial stem cells can be efficiently proliferated by culturing the stem cells in soft agar-coated *** new protocol used in this study can be a valuable method for future studies.

The Influence of Surrounding Ground Floor Facilities on the Use of Privately Owned Public Spaces in Seoul

Seoul’s privately owned public space (POPS) initiative has achieved quantitative success since its introduction in the early 1990s. However, POPSs have been frequently criticized for failing to correspond with the urban context in which they are created and are thus often underused. Association between surrounding urban context and the use of POPS is acknowledged, but specific elements of the urban context associated with the use of POPS and the extent of association are unclear. In this study, the relationship between constituent elements of 48 POPSs including the surrounding ground floor facility use and the use of those spaces in Seoul’s central business district was measured by multiple regression analysis. The results present empirical evidence of a substantial relationship between assembly, mercantile, and business facilities on ground floors of surrounding buildings and the use of POPS. As the surrounding ground floor facility use showed an overriding relationship with the use of POPS that possibly overshadowed the influence of secondary determinants, the 48 POPSs were divided into two groups: those surrounded by commercial facilities and those surrounded by business facilities. The use of POPSs surrounded by commercial facilities was associated with additional variables including the proximity to transit stations and crosswalks. Models using the sample of POPSs surrounded by business facilities were not statistically significant. Based on these results, this paper emphasizes the importance of creating POPSs in connection with desirable surrounding facilities. This study also suggests that anticipation of the usage patterns of POPSs and establishment of guidelines supporting those patterns may be viable by investigating preexisting urban contexts.

By performing a statistical change-point analysis of activities of the tropical cyclones(TCs)that have affected Korea(K-TCs),it was found that there was a signifi cant change between 1983 and *** the period of 1984-2004(P2),more TCs migrated toward the west,recurved in the southwest,and affected Korea,compared to the period of 1965-1983(P1).These changes for P2 were related to the southwestward expansion of the subtropical western Pacifi c high(SWPH)and,simultaneously,elongation of its elliptical shape toward *** of these changes,the central pressure and lifetime of K-TC during P2 were deeper and longer,respectively,than fi gures for *** stronger K-TC intensity for P2 was related to the more southwestward genesis due to the southwestward expansion of the *** weaker vertical wind shear environment during P2 was more favorable for K-TC to maintain a strong intensity in the mid-latitudes of East Asia.

A seasonal probability prediction model for tropical cyclone(TC)genesis in the western North Pacifi c(5°-25°N,110°-180°E)was developed using a simple logistic regression *** predictors used in this model were total fi ve:850 h Pa relative vorticity,200-850 h Pa vertical wind shear,600 h Pa relative humidity,300 h Pa equivalent potential temperature,and sea surface temperature(SST).Four predictors except for SST were obtained from differences of spatial-averaged value between May and January,and time average of Ni?o-3.4 index from February to April was used to consider the SST *** a result of prediction for the TC genesis from June to December during 1951 to 2007,years that the model forecasts are 21 years among the total 28 years when the observed TC genesis frequency was higher than normal *** overall predictability was about 75%,and the model was also verifi ed statistically through cross validation analysis using a method of the hindcast.

Invasive infection caused by Streptococcus pyogenes emm89 strains has been increasing in several countries linked to a recently emergent clade of emm89 strains,designated clade *** Japan,the features of emm89 *** strains,such as clade classification,remains *** this study,we collected emm89 strains isolated from both streptococcal toxic shock syndrome(STSS)(89 STSS isolates)and noninvasive infections(72 non-STSS isolates)in Japan from 2011 to 2019,and conducted wholegenome sequencing and comparative analysis,which resulted in classification of a large majority into clade 3 regardless of disease *** addition,invasive disease-associated factors were found among emm89 strains,including mutations of control of virulence sensor,and absence of the hylP1 gene encoding *** findings provide new insights into genetic features of emm89 strains.

The Influence of Exchange Rate on the Volume of Japanese Manufacturing Export

This paper investigates the influence of exchange rate volatility on the volume of Japanese manufacturing export. The volatility in yen is shown by conditional variance from EGARCH （Exponential Generalized Autoregressive Conditional Heteroscedasticity） model, allowing for asymmetric effects that a shock of an appreciation of the yen is different from that of a depreciation of the yen. The export action model including exchange rate volatility is constructed based on VAR （Vector Auto Regressive） model to examine the relationship between exchange rate uncertainty and the volume of export. Tests are performed for typical eight kinds of industry in Japan. Few empirical studies focus on each Japanese industry export. Results indicate significant negative effects of exchange rate volatility on most manufacturing exports. In addition, this paper analyzes the each industry, featurc of the influence of exchange rate on the volume of Japanese export. The authors find that equipment industries occupying 60% or more of total Japanese exports especially tend to receive negative influence of exchange.

The present study was designed to clarify the roles of N-type and P/Q-type calcium channels in the increased sympathetic activity of spontaneously hypertensive rats (SHR/Izm). We also tested in SHR/Izm the contribution to increased vascular tone of α1A adrenoceptor-linked L-type calcium channels and α1B receptor-mediated calcium mobilization from the sarcoplasmic reticulum. Methods: Six-week-old SHR/Izm and Wistar-Kyoto rats (WKY/Izm) were used. A superior mesenteric arterial preparation was electrically stimulated before and after treatment with ω-conotoxin GVIa (N-type calcium channel blocker [CgTX]) and ω-agatoxinIVa (P/Q-type calcium channel blocker [AgaTX]). Pressor response to norepinephrine was measured before and after treatment with the α1A blocker WB-4101 and the α1B blocker chloroethyl clonidine (CEC). To determine the intracellular calcium store size, the effects of ryanodine on pressor response and caffeine-induced vascular contraction were also tested. Results: Norepinephrine overflow evoked by electrical stimulation was increased in SHR/Izm. CgTX but not AgaTX suppressed the increased NE overflow in SHR/Izm. WB-4101 suppressed the pressor response to norepinephrine in SHR/Izm but not WKY/Izm rats. CEC had no effects on pressor response to norepinephrine in both types of rats. Caffeine-induced contraction to a high potassium-induced maximal contraction ratio was reduced in SHR/Izm. The effect of ryanodine on pressor response was reduced in SHR/Izm. Conclusion: N-type calcium channels but not P/Q-type calcium channels play an important role in the increased sympathetic tone in SHR/ Izm. Although α1A adrenoceptor-linked L-type calcium channels contribute to the increased vascular tone, the intracellular calcium store size was reduced in SHR/Izm.

Methionine and Threonine Requirements of Dutch Rabbits Fed under a Cecotrophy Prevention Program

It is suggested that the difference in body size between domestic-type rabbits and small pet-type rabbits results in different nutrient requirements. The nutritional requirements of pet rabbits have been assessed, although such assessments require evaluation throughout the rabbit life span. The present study was conducted under a cecotrophy prevention program with young and adult rabbits. Six male Dutch rabbits were housed individually in a dormitory-type cage, and they were randomly fed graded levels of dietary methionine and threonine, at ratios of 4:0, 3:1, 2:2, 1:3, and 0:4 mixed with two types of feed, 4:0 and 0:4. Four days after switching diets and 4 hrs after starting morning feeding, approximately one milliliter of blood was collected from the vein of the ear. Free amino acid concentrations in the plasma were determined with a high-speed amino acid analyzer. Plasma concentrations of methionine and threonine compared to dietary methionine and threonine levels are shown in young rabbits. The plasma concentration of methionine remained constant at a low level and then increased linearly. The intersection was estimated as 0.16 g/d. In the same manner, the intersection of the plasma threonine value was estimated as 0.27 g/d. These values were calculated as 0.27% and 0.47% of the diets, respectively. In the case of adult rabbits, the baseline was not obtained for dietary methionine and threonine levels. The methionine requirement was estimated as 0.11 g/d. The threonine requirement was estimated as 0.22 g/d. These values were calculated as 0.15% and 0.30% of the diets, respectively. In comparison with young and adult rabbits, both methionine and threonine showed a low baseline level in young rabbits, while their variations in plasma levels of adult rabbits were not determined.
