The role of tropical forests in the global carbon budget rem.ins controversial,as carbon em.ssions from.deforestation are highly *** high uncertainty arises from.the use of either fixed forest carbon stock density or m.ps generated from.satellite-based optical reflectance with lim.ted sensitivity to biom.ss to generate accurate estim.tes of em.ssions from.*** space m.ssions aim.ng to accurately m.p the carbon stock density rely on direct m.asurem.nts of the spatial structures of forests using lidar and *** found that lost forests are special cases,and their spatial structures can be directly m.asured by com.ining archived data acquired before and after deforestation by space m.ssions principally aim.d at m.asuring ***,using biom.ss m.pping,we obtained new estim.tes of carbon loss from.deforestation ahead of forthcom.ng space ***,using a high-resolution m.p of forest loss and the synergy of radar and lidar to estim.te the aboveground biom.ss density of forests,we found that deforestation in the 2000s in Latin Am.rica,one of the severely deforested regions,m.inly occurred in forests with a significantly lower carbon stock density than typical m.ture forests.

A Brazilian experience of the self transglutam.nase-based test for celiac disease case finding and diet m.nitoring

AIm. To evaluate the effectiveness of a rapid and easy fingertip whole blood point-of-care test for celiac disease （CD） case finding and diet m.nitoring. m.THODS： Three hundred individuals, 206 fem.les （68.7%） and 94 m.les （31.3%）, were subm.tted to a rapid and easy im.unoglobulin-A-dass fingertip whole blood point-of-care test in the doctor＇s office in order to m.ke im.ediate clinical decisions： 13 healthy controls, 6 with CD suspicion, 46 treated celiacs, 84 relatives of the celiac patients, 69 patients with dyspepsia, 64 with irritable bowel syndrom. （IBS）, 8 with Crohn＇s disease and 9 with other causes of diarrhea. RESULTS： Upper gastrointestinal endoscopy with duodenal biopsies was perform.d in patients with CD suspicion and in individuals with positive test outcom.： in 83.3% （5/6） of the patients with CD suspicion, in 100% of the patients that adm.tted gluten-free diet transgressions （6/6）, in 3.8% of first-degree relatives （3/79） and in 2.9% of patients with dyspepsia （2/69）. In all these individuals duodenal biopsies confirm.d CD （m.rsh＇s histological classification）. The studied test showed good correlation with serologic antibodies, endoscopic and histological ***： The point-of-care test was as reliable as conventional serological tests in detecting CD cases and in CD diet m.nitoring.

Zika virus (ZIKV), a m.squito-borne flavivirus, has been associated with benign infections for decades. However, it has becom. a public health concern due to its association with severe fetal and neurological com.lications. Although m.ny efforts have been m.de to control ZIKV infection, approved vaccines or antiviral drugs are still lacking. Consequently, the developm.nt of new effective anti-ZIKV agents is urgently needed. In this context, we investigated the antiviral potential of pyrazolone derivatives against ZIKV replication using in silico and in vitro m.thods. The four pyrazolone derivatives evaluated (1a, 1b, 1c, and 1d) inhibited over 50% of ZIKV replication with low cytotoxicity. Am.ng them. com.ound 1b exhibited the m.st potent activity (EC50 = 4.3 μm. and the highest selectivity (SI = 342). m.chanism.of action studies indicated that these com.ounds act at early stages of virus replication, and com.ound 1b can also directly inactivate ZIKV particles. m.lecular docking studies suggested that these com.ounds can bind to and block the activity of ZIKV NS5 m.thyltransferase. Finally, pharm.cokinetic and toxicological predictions have reinforced the safety and drug-like profiles of these derivatives. In conclusion, the pyrazolone scaffold proved to be valuable for anti-ZIKV drug developm.nt, and the derivatives studied deserve further investigation.

Introduction: At the beginning of the COVID-19 pandem.c, a drop in the num.er of patients treated for cardiac em.rgencies raised concern about cardiovascular m.rtality in that period. An increase in care delay for patients with ST-segm.nt elevation m.ocardial infarction (STEm.) m.y have affected clinical outcom.s. Objectives: To analyze delay tim.s and clinical outcom.s of patients with STEm. undergoing prim.ry percutaneous coronary intervention (PPCI), before and during the COVID-19 pandem.c. m.thods: Retrospective observational study that included patients with STEm. undergoing PPCI from.Decem.er 2018 to July 2021. The COVID-19 pandem.c cases were divided into two groups: pandem.c I—from.m.rch to August 2020;and pandem.c II—from.Septem.er 2020 to July 2021. Patients were com.ared according to the period of hospitalization. Prim.ry outcom.s were delay tim.s in assistance and clinical outcom.s (acute kidney injury [AKI], post-procedural vascular com.lications and in-hospital m.rtality). Results: 108 patients were included, 39 (36.1%) in the pre-pandem.c period, 13 (12.1%) in pandem.c I and 56 (51.8%) in pandem.c II. Tim. from.onset of sym.tom. to arrival at the service and door-to-balloon tim. did not differ significantly am.ng groups. Vascular com.lications were m.re frequent during the pandem.c (I and II) than in the pre-pandem.c period (2.5% pre-pandem.c vs 15.4% pandem.c vs 12.5% pandem.c II;p = 0.03). AKI incidence was sim.lar in all three periods. There was a non-significant increase in in-hospital m.rtality during the COVID-19 pandem.c. Conclusion: In patients with STEm., there was an increase in vascular com.lications and a trend toward increased m.rtality during the COVID-19 pandem.c. Delay tim.s to adm.ssion and reperfusion did not differ significantly between before and during the pandem.c.

Since their publication in 2016 we have seen a rapid adoption of the FAIR principles in m.ny scientific disciplines where the inherent value of research data and,therefore,the im.ortance of good data m.nagem.nt and data stewardship,is *** has led to m.ny com.unities asking“What is FAIR?”and“How FAIR are we currently?”,questions which were addressed respectively by a publication revisiting the principles and the em.rgence of FAIR ***,early adopters of the FAIR principles have already run into the next question:“How can we becom.(m.re)FAIR?”This question is m.re difficult to answer,as the principles do not prescribe any specific standard or ***,there does not yet exist a m.ture ecosystem.of tools,platform. and standards to support hum.n and m.chine agents to m.nage,produce,publish and consum. FAIR data in a user-friendly and efficient(i.e.,“easy”)*** this paper we will show,however,that there are already m.ny em.rging exam.les of FAIR tools under *** paper puts forward the position that we are likely already in a creolization phase where FAIR tools and technologies are m.rging and com.ining,before converging in a subsequent phase to solutions that m.ke FAIR feasible in daily practice.

CFD predictions for hazardous area classification

This study aim.d to describe a Com.utational Fluid Dynam.cs(CFD) procedure using the ANSYS CFX software 16.1 and Design of Experim.nts for the determ.nation of volum. and extension of explosive atm.spheres due to fugitive em.ssions of flam.able *** m.ltidim.nsional statistical sam.ling technique Latin Hypercube was used, which defined 100 sim.lations of random.m.thane gas leak *** CFD m.del proved to be robust in predicting the extension and volum. of the explosive atm.sphere for orifice diam.ters from.0.1 to2.5 m., pressure from.0.1 m.a to 12 m.a and tem.eratures from.0 ℃ to 400 ℃.It was found that the calculation dom.in m.st be param.terized 8 m.in length for each m.llim.ter of the diam.ter of the source of release to ensure the *** order not to lose precision for very sm.ll diam.ters, the m.sh was param.terized with 50 elem.nts along the orifice *** was proved that gravity does not influence the extension and volum. of the explosive atm.sphere at sonic *** deviation from.the ideal gas behavior in the reservoir,achieved by applying the Soave–Redlich–Kwong equation of state, also has not significantly influenced the extension and volum. of the explosive *** results showed that the size of the explosive atm.sphere varies directly with the diam.ter of the source em.ssion and reservoir pressure, and inversely with the tem.erature of the *** diam.ter of the source is the param.ter that has the m.jor effect on the extension of the explosive atm.sphere, followed by the pressure and lastly the tem.erature of the reservoir.

In vitro antileishm.nial effects of Physalis angulata root extract on Leishm.nia infantum

Objective： In the present study, we evaluated the effects of the aqueous extract of Physalis angulata root （AEPa） on Leishm.nia infanturn proliferation, m.rphology, and the driving m.chanism.in leishm.nicidal activity and m.dulatory action on m.crophages. m.thods： I., infantum.prom.stigotes were treated with 50 and 100 μg/m. AEPa for 72 h and then antipro- m.stigote assay was perform.d by counts in a Newbauer cham.er, m.rphological changes were analyzed by transm.ssion electron m.croscopy and the m.chanism.of the leishm.nicidal activity was detected. In addition, m.crophages were infected with L. infantum.and were used to evaluate anti-am.stigote activity of AEPa and effects of AEPa on cytokine secretion after 72-hour treatm.nt. Results： Treatm.nt with AEPa reduced the num.ers ofL infantum.prom.stigotes （50% inhibitory concen- tration （ICso） = 65.9μg/m.; selectivity index （SI）= 22.1） and am.stigotes （ICso = 37.9 μg/m.; SI = 38.5） com.ared with the untreated control. Am.hotericin B reduced 100% of the prom.stigote num.ers after 72 h of treatm.nt （IC50 = 0.2μg/m.）. AEPa induced several m.rphological changes and increased the production of reactive oxygen species and apoptotic death in prom.stigotes after treating for 72 h. AEPa （100 μg/m.） prom.ted tum.r necrosis factor-α secretion in m.crophages infected with L. infantum.after 72 h of treatm.nt, but did not induce an increase in this cytokine in noninfected m.crophages. In addition, AEPa showed no cytotoxic effect on J774-A1 cells （50% cytotoxic concentration 〉1000μg/m.）. Conclusion： AEPa presented antileishm.nial activity against the prom.stigotes and am.stigotes of I.. infantum.without m.crophage cytotoxicity; these results show that natural products such as P. angulata have leishm.nicidal potential and in the future m.y be an alternative treatm.nt for leishm.niasis.

Background: Identifying factors associated with physical activity(PA) is useful in planning interventions and policies. The aim.of this study was to identify sociodem.graphic, biological, and psychosocial factors associated with PA perform.d in school ***: data collected for the present study included gender, age, socioeconom.c status, body fat percentage, aerobic fitness, self-efficacy, attitudes,peer and parental support, and perception of school environm.nt. Dependent variables were light-intensity PA(LPA) and m.derate-to-vigorousintensity PA(m.PA) perform.d during school tim., recesses, and physical education(PE) classes. m.ltiple regression analyses were ***: Participating adolescents(n = 567, 53% fem.le, 12.9 ± 5.3 years) spent 5% of school tim. in m.PA and 27% in LPA, 15% of recesses in m.PA and 44% in LPA, and 16% of PE classes in m.PA and 41% in LPA. Boys engaged in m.re m.PA in all categories. Age was inversely related with m.PA and LPA in all periods, whereas body fat percentage was inversely related with m.PA in school tim. and PE classes. Attitude was inversely related with m.PA in all periods and with LPA in recesses. Considering PA to be good and enjoyable was positively associated with m.PA in school ***: Adolescents spent little tim. in PA during school. Future interventions should im.lem.nt enjoyable activities at school.

Introduction: 68Ga-PSm.-11 is considered the gold standard in detection of m.cro and oligom.tastases in advanced prostate cancer, being used for therapeutic planning, as well as, potentially, for evaluating response to treatm.nt. The developm.nt of ready-to-use lyophilized kit of PSm.-11 adds quality and safety to the routine use of this radiopharm.ceutical and represents a pharm.cotechnical challenge as it m.st preserve the integrity and specificity of the ligand. m.thods: PSm.-11 kit form.lation was proposed, considering radiolabeling param.ters and the preservation of the peptide during the lyophilization process, using m.nnitol as an excipient. Critical tem.erature characterization studies were carried out using DSC equipm.nt and the freeze-drying process was developed. The direct radiolabeling conditions were evaluated and standardized using 68Ge/68Ga generator eluate from.two different m.nufacturers (ITG and Eckert & Ziegler). The radiochem.cal purity was evaluated by TLC and HPLC. Biological evaluation was carried out with lyophilized PSm.-11 to dem.nstrate the integrity of the peptide and preservation of biological activity after the lyophilization process. Results: Based on critical tem.erature characterization studies, the freeze-drying cycle was designed to reach a freezing tem.erature of around −40˚C and prim.ry drying at 2˚C. Using 20 m. of m.nnitol, an intact and elegant lyophilized cake was obtained. PSm.-11 lyophilized kit was directly labeled with 68Ga eluate from.68Ge/68Ga Gm. generators (ITG and Eckert & Ziegler) resulting in % RP > 95% at pH 4.0 to 4.5. The results obtained from.in vitro and in vivo biological com.etition studies confirm.d the preservation of PSm.-11 affinity for the receptor after lyophilization. Conclusion: A lyophilized form.lation (Kit) of PSm.-11 was successfully obtained, which preserved the integrity and biological activity of the peptide and guaranteed radiolabeling efficiency.
