Enhancement of the Antigenotoxic and Antioxidant Actions of Eugenol from Spice Clove and the Stabilizer Gum Arabic on Colorectal Carcinogenesis

Spices are defined as any aromatic condiment of plant origin used to alter the flavor and aroma of foods. Besides flavor and aroma, many spices have antioxidant activity, mainly related to the presence in cloves of phenolic compounds, such as flavonoids, terpenoids and eugenol. In turn, the most common uses of gum arabic are in the form of powder for addition to soft drink syrups, cuisine and baked goods, specifically to stabilize the texture of products, increase the viscosity of liquids and promote the leavening of baked products (e.g., cakes). Both eugenol, extracted from cloves, and gum arabic, extracted from the hardened sap of two species of the Acacia tree, are dietary constituents routinely consumed virtually throughout the world. Both of them are also widely used medicinally to inhibit oxidative stress and genotoxicity. The prevention arm of the study included groups: Ia, IIa, IIIa, Iva, V, VI, VII, VIII. Once a week for 20 weeks, the controls received saline s.c. while the experimental groups received DMH at 20 mg/kg s.c. During the same period and for an additional 9 weeks, the animals received either water, 10% GA, EUG, or 10% GA + EUG by gavage. The treatment arm of the study included groups Ib, IIb, IIIb e IVb, IX, X, XI, XII). Once a week for 20 weeks, the controls received saline s.c. while the experimental groups received DMH at 20 mg/kg s.c. During the subsequent 9 weeks, the animals received either water, 10% GA, EUG or 10% GA + EUG by gavage. The novelty of this study is the investigation of their use alone and together for the prevention and treatment of experimental colorectal carcinogenesis induced by dimethylhydrazine. Our results show that the combined use of 10% gum arabic and eugenol was effective, with antioxidant action in the colon, as well as reducing oxidative stress in all colon segments and preventing and treating genotoxicity in all colon segments. This paper presents a proposal for an experimental salt cavern in offshore ultra-deep water for CO2 abatement,including the instrumentation plan and well conceptual design evaluated for carbon capture and storage(CCS)application.These studies are based on applied computational mechanics associated with field experimentation that has contributed to the technical feasibility of the underground potash mine at the State of Sergipe in Brazil.This knowhow allowed the stability analysis of several salt caverns for brine production at the State of Alagoas in Brazil and to the drilling through stratified thick layers of salt of the pre-salt reservoirs in Santos Basin.Now,this knowledge has been applied in the design of onshore and offshore salt caverns opened by dissolution for storage of natural gas and CO2.The geomechanical study,through the application of computational mechanics,of offshore giant salt caverns of 450 m high by 150 m in diameter,shows that one cavern can store about 4 billion Sm3 or 7.2 million tons of CO2.Before the construction of the giant cavern,which will be the first gas storage offshore in the world,it has been decided to develop an experimental one,with smaller size,to obtained field parameters.The experimental cavern will allow the calibration of parameters to be used in the structural integrity analysis of the cavern and well for storage of natural gas which is rich in CO2 under high pressure.

Novel experimental techniques to assess the time-dependent deformations of geosynthetics under soil confinement

A new experimental approach to assess the impact of soil confinement on the long-term behavior of geosynthetics is presented in this paper.The experimental technique described herein includes a novel laboratory apparatus and the use of different types of tests that allow generation of experimental data suitable for evaluation of the time-dependent behavior of geosynthetics under soil confinement.The soil-geosynthetic interaction equipment involves a rigid box capable of accommodating a cubic soil mass under plane strain conditions.A geosynthetic specimen placed horizontally at the mid-height of the soil mass is subjected to sustained vertical pressures that,in turn,induce reinforcement axial loads applied from the soil to the geosynthetic.Unlike previously reported studies on geosynthetic behavior under soil confinement,the equipment was found to be particularly versatile.With minor setup modifications,not only interaction tests but also in-isolation geosynthetic stress relaxation tests and soil-only tests under a constant strain rate can be conducted using the same device.Also,the time histories of the reinforcement loads and corresponding strains are generated throughout the test.Results from typical tests conducted using sand and a polypropylene woven geotextile are presented to illustrate the proposed experimental approach.The testing procedure was found to provide adequate measurements during tests,including good repeatability of test results.The soilegeosynthetic interaction tests were found to lead to increasing geotextile strains with time and decreasing reinforcement tension with time.The test results highlighted the importance of measuring not only the time history of displacements but also that of reinforcement loads during testing.The approach of using different types of tests to analyze the soilegeosynthetic interaction behavior is an innovation that provides relevant insight into the impact of soil confinement on the time-dependent deformations of geosynthetics.

Global climate changes have increased temperatures,radiation indexes,and consequently,irregularities in rainfall in mainly tropical countries,considerably hindering plant establishment in recovering degraded areas.The objective of this study was to evaluate the growth and physiological characteristics of one species of each successional group:pioneer,secondary,and climax when subjected to different light intensities and hydrogel as a soil conditioner during rainy and dry periods.The experiment was conducted in the ecotone between Brazil’s two largest biomes,the Cerrado and the Amazon in the State of Maranh?o.The parameters consisted of three species:Guazuma ulmifolia Lam.(pioneer),Astronium fraxinifolium Schott(secondary),and Cariniana rubra Gardner ex Miers(climax).There were two light intensities:70%and 100%,and two planting conditions:with and without soil conditioner(hydrogel).Gas exchanges were higher during the rainy season;the pioneer and secondary species had greater heights and photosynthetic rates in the dry period;the climax species had the lowest gas exchange and lowest recovery as rainfall resumed.The pioneer and secondary species showed higher physiological plasticity,denoting better adaption to environments with high irradiance.Hydrogel improved the photosynthetic performance of these species in the dry season and in areas with 100%sunlight.

Cytomegalovirus is the most frequent agent of congenital viral infections,affecting approximately 0.2%-2.4%of live births in different countries[1-7].Universal screening for congenital cytomegalovirus infection in newborns is not a standard practice considering cost-benefit issues and viabil-ity of diagnostic tests.Besides,most newborns are asympto-matic and there is no effective treatment for this population[2-5].

Patients’ perceptions, health and psychological changes with obesity treatment: Success and failure in a triangulation study

The present study aims to understand changes in health problems, health complaints and coping strategies, during the obesity treatment process with qualitative and quantitative data. Thirty bariatric patients were interviewed before bariatric surgery and at a 12-month follow-up, and fulfilled self-report measures about health problems, health complaints and coping strategies before surgery, at 6-and 12-month follow-ups. Before surgery, failure cases differ from success on the conceptualization of obesity, However, there are no other differences between groups. At 6-and 12-month follow-ups, failure cases had the highest BMI, health problems and complaints and less % EWL than success cases. One year after the surgery, one in each three persons did not lose the expected weight, i.e., are failure cases. Before surgery, there are no differences between success and failure cases in the report of health problems, health complaints and coping strategies, but they have different conceptualizations of their obesity and treatment. One year after the surgery, success cases understood bariatric surgery as an important moment in their lives related to their expected results, whereas failures valued unexpected dimensions and still waiting for a miracle surgery without their personal commitment. Accordingly, it is necessary to consider lifestyle changes in the obesity treatment process.

Latent tuberculosis:Risk factors,screening and treatment in liver transplantation recipients from an endemic area

BACKGROUND Patients undergoing solid organ transplantation,particularly those who live or have lived in tuberculosis(TB)endemic areas,are at a high risk of developing TB.The majority of post-transplantation TB cases are associated with reactivation of latent TB infection(LTBI).Brazil is in a single position with overlapping areas of high TB endemicity and high transplant activity.In liver transplant(LT),one should be aware of the potential hepatotoxicity associated with the treatment regimens for LTBI.AIM To evaluate the frequency of LTBI in LT patients and treatment-related issues.METHODS This was a retrospective analysis of a cohort of cirrhotic patients aged≥18 years,who underwent LT at a high-complexity teaching hospital from January 2005 to December 2012.RESULTS Overall,429 patients underwent LT during the study period.Of these,213(49.7%)underwent the tuberculin skin test(TST)during the pre-transplant period,and 35(16.4%)of them had a positive result.The treatment for LTBI was initiated after LT in 12(34.3%)of the TST-positive patients;in 3(25.0%),treatment was maintained for at least 6 mo.CONCLUSION The prevalence of LTBI was lower than expected.Initiation and completion of LTBI treatment was limited by difficulties in the management of these special patients.

Biofilm analyses and exoproduct release by clinical and environmental isolates of Burkholderia pseudomallei from Brazil

Objective:To characterize biofilm production by clinical(n=21)and environmental(n=11)isolates of Burkholderia pseudomallei and evaluate the production of proteases,hemolysins and siderophores.Methods:Initially,the 32 strains were evaluated for biofilm production in Müller-Hinton broth-1%glucose(MH-1%glucose)and BHI broth-1%glucose,using the crystal violet staining technique.Subsequently,growing(48 h)and mature(72 h)biofilms were evaluated by confocal microscopy.Finally,the production of proteases,hemolysins and siderophores by planktonic aggregates,growing biofilms and mature biofilms was evaluated.Results:All isolates produced biofilms,but clinical isolates had significantly higher biomass in both MH-1%glucose(P<0.001)and BHI-glucose 1%(P=0.005).The structural analyses by confocal microscopy showed thick biofilms,composed of multiple layers of cells,homogeneously arranged,with mature biofilms of clinical isolates presenting higher biomass(P=0.019)and thickness of the entire area(P=0.029),and lower roughness coefficient(P=0.007)than those of environmental isolates.Protease production by growing biofilms was significantly greater than that of planktonic(P<0.001)and mature biofilms(P<0.001).Hemolysin release by planktonic aggregates was higher than that of biofilms(P<0.001).Regarding siderophores,mature biofilms presented higher production than growing biofilms(P<0.001)and planktonic aggregates(P<0.001).Conclusions:Clinical isolates have higher production of biofilms than their environmental counterparts;protease and siderophores seem important for growth and maintenance of Burkholderia pseudomallei biofilms.

Concordance of the point-of-care circulating cathodic antigen test for the diagnosis of intestinal schistosomiasis in a low endemicity area

Background:The Kato-Katz technique is recommended worldwide for the diagnosis of intestinal schistosomiasis,detecting parasite eggs in feces of infected people.However,new tests have been developed in order to facilitate diagnosis,e.***.by detection of specific antigens secreted by schistosomes,such as the circulating cathodic antigen(CCA).The aim of this study was to evaluate the performance of the point-of-care circulating cathodic antigen test(POC-CCA)compared to the Kato-Katz technique in a low prevalence area in the Amazon Region,located in the municipality of Primavera,State of Para,Brazil.Methods:Positivity rates of the POC-CCA test and the Kato-Katz technique were calculated.The sensitivity,specificity,accuracy and kappa coefficient were determined by comparing both methods.The reference standard was established using 16 Kato-Katz slides,12 of the first fecal sample,two of the second and two of the third one.The study also included the concordance between POC-CCA results and different numbers and combinations of Kato-Katz slides.Results:The prevalence of schistosomiasis according to the reference standard or POC-CCA test reached a rate of 9.4%or 23.9%,respectively,among a total of 372 participants.The positivity rates by the Kato-Katz technique increased from 2.4 to 9.4%,according to the increase in the number of slides examined and fecal samples colleaed.A sensitivity of 55.6%,specificity 76.9%,accuracy 76%and k coefficient of 0.06 was observed by comparing one slide of the first sample and POC-CCA.Comparing 6 slides from three different samples,two slides of each,with POC-CCA resulted in a sensitivity of 583%,specificity 78.4%,accuracy 77%and k coefficient of 0.16.Finally,the comparison of 16 slides from three different samples with POC-CCA revealed a sensitivity of 65.7%,specificity 80.4%,accuracy 79%,and k coefficient of 0.27.Conclusions:The immunochromatographic test has the potential to be an important tool to combat schistosomiasis because of its practicality and applicability but should be applied w
