There is increasing evidence that pathogens do not only elicit direct defense responses, but also cause pronounced changes in primary carbohydrate metabolism. Cell-wall-bound invertases belong to the key regulators of carbohydrate partitioning and source-sink relations. Whereas studies have focused so far only on the transcriptional induction of invertase genes in response to pathogen infection, the role of post-translational regulation of invertase activity has been neglected and was the focus of the present study. Expression analyses revealed that the high mRNA level of one out of three proteinaceous invertase inhibitors in source leaves of Arabidopsis thaliana is strongly repressed upon infection by a virulent strain of Pseudomonas syringae pv. tomato DC3000. This repression is paralleled by a decrease in invertase inhibitor activity. The physiological role of this regulatory mechanism is revealed by the finding that in situ invertase activity was detectable only upon infection by P. syringae. In contrast, a high invertase activity could be measured in vitro in crude and cell wall extracts prepared from both infected and non-infected leaves. The discrepancy between the in situ and in vitro invertase activity of control leaves and the high in situ invertase activity in infected leaves can be explained by the pathogen-dependent repression of invertase inhibitor expression and a concomitant reduction in invertase inhibitor activity. The functional importance of the release of invertase from post-translational inhibition for the defense response was substantiated by the application of the competitive chemical invertase inhibitor acarbose. Posttranslational inhibition of extracellular invertase activity by infiltration of acarbose in leaves was shown to increase the susceptibility to P. syringae. In Arabidopsis thaliana,canonical auxin-dependent gene regulation is mediated by 23 transcription factors from the AUXIN RESPONSE FACTOR(ARF)family that interact with auxin/indole acetic acid repressors(Aux/IAAs),which themselves form co-receptor complexes with one of six TRANSPORT INHIBITOR*!/AUXIN-SIGNALLING F-BOX(TIR1/AFB)*** combinations of co-receptors drive specific sensing outputs,allowing auxin to control a myriad of ***6 and ARF8 are positive regulators of adventitious root initiation upstream of jasmonate,but the exact auxin co-receptor complexes controlling the transcriptional activity of these proteins has remained ***,using loss-of-function mutants we show that three Aux/IAA genes,IAA6,IAA9,and IAA17,act additively in the control of adventitious root(AR)*** three IAA proteins interact with ARF6 and/or ARF8 and likely repress their activity in AR *** show that TIR1 and AFB2 are positive regulators of AR formation and TIR1 plays a dual role in the control of jasmonic acid(JA)biosynthesis and conjugation,as several JA biosynthesis genes are up-regulated in the tir1-1 *** results lead us to propose that in the presence of auxin,TIR1 and AFB2 form specific sensing complexes with IAA6,IAA9,and/or IAA17 to modulate JA homeostasis and control AR initiation.

Artemisinin, an endoperoxide sesquiterpene lactone has proven effective in treating drug resistant cases of malaria and cancer. Artemisia annua [sweet wormwood] is the sole source for artemisinin production in many countries. To counter the low content in leaves and costly chemical synthesis process in India, alternative ways to produce artemisinin have been sought. In current study, we collected A. pallens, A. japonica and A. nilagirica from Western Ghats of Maharashtra, India and analyzed artemisinin content. Samples were extracted from leaves and florets in various extraction conditions and analyzed using different chromatographic techniques. Thin layer chromatography (TLC) and high performance thin layer chromatography (HPTLC) analysis showed the presence of compound with endoperoxide linkage in A. nilagirica. High performance liquid chromatography (HPLC) analysis showed the detection of artemisinin in methylene dichloride florets extract of A. japonica, but the concentration was too low [1.3 mg/g dry wt.] for further analyses. Gas chromatography/mass spectrometry (GC/MS) analysis identified structurally important components in the A. nilagirica ethyl acetate extract which explores the biosynthetic pathway of artemisinin from its most important precursor amorpha-4,11-diene. This is the first report of chromatographic screening of these Indian varieties of Artemisia spp. for artemisinin content.

AACA Element Negatively Regulates Expression of Protein Phosphatase 2C (PP2C) like Promoter in Arabidopsis thaliana

Genome wide analysis of Arabidopsis thaliana reveals a unique genetic arrangement of ACGT motif in the protein phosphatase 2C (PP2C) like promoter (accession number AT5G59220). In the present study, full length, 900 base pair, 500 base pair, 400 base pair and NRM deletion variants of PP2C like promoter were constructed to investigate the activity of PP2C like promoter in the presence of inducers like abscisic acid, jasmonic acid and salicylic acid. The PP2C promoter has three ACGT elements in close vicinity. They are so positioned that they are separated by a spacer of 30 base pair and 5 base pair respectively from each other. The study has shown that ACGTN30ACGT genetic architecture is essential for the promoter to be induced in response to abscisic acid. The synergistic and antagonistic effects of cis elements were observed. AACA is a positive regulatory element in endosperm and is known to act as negative regulatory element in other tissues. In this study AACA, have been found to negatively regulate the expression of reporter gene EGFP in both induced and under uninduced conditions.

Association of Obesity in Children with Pancreatitis in a Predominantly Hispanic Population at the US-Mexican Border: A Single Center Outpatient Study

Importance: The incidence of pancreatitis has been increasing over the last decade. Limited data are available for pancreatitis in the Hispanic population. The clinical profile of pancreatitis and its association with obesity in a Mexican American population has not been well studied. Objective: To describe prevalence, the characteristics of pancreatitis cases, and determine its association with obesity. Design and Setting: Retrospective review of children from February 2012 to February 2015, at the Texas Tech University Health Sciences Center of El Paso (TTUHSC-El Paso) outpatient center. Case/non-case study was used to determine the association of overweight and obesity with pancreatitis. Participants: Children ages 2 - 18 years, only the newly diagnosed pancreatitis cases irrespective of its type excluding patients with traumatic and repeat visit for pancreatitis. Main Outcome and Measures: Pancreatitis and obesity. Results: The prevalence of pancreatitis was estimated as 20 cases in 21,444 patients seen in the outpatient clinic. In multivariable analysis, only obesity and ethnicity were significantly associated with pancreatitis. Obesity was found to be more than 7-fold likely with pancreatitis as compared to non-cases. Overweight was found to be more than 6-fold likely with pancreatitis as compared with non-cases. Our study demonstrates an association between obesity and pancreatitis in the study population. Conclusions and Relevance: Appropriate intervention for managing obesity may reduce the prevalence of pancreatitis and associated co-morbidities. A larger prospective study should be done to determine underlying causes of association between obesity and pancreatitis.

Dynamic Analysis of Overhead Transmission Lines under Turbulent Wind Loading

Transmission tower-line systems are designed using static loads specified in various codes. This paper compares the dynamic response of a test transmission line with the response due to static loads given by Eurocode. Finite element design software SAP2000 was used to model the towers and lines. Non-linear dynamic analysis including the large displacement effects was carried out. Macroscopic aspects of wind coherence along element length and integration time step were investigated. An approach is presented to compare the probabilistic dynamic response due to 7 different stochastically simulated wind fields with the response according to EN-50341. The developed model will be used to study the response recorded on a test line due to the actual wind speed time history recorded. It was found that static load from EN overestimated the strength of conductor cables. The response of coupled system considering towers and cables was found to be different from response of only cables with fixed supports.
