Co-delivery of PARP and PI3K inhibitors by nanoscale metal-organic frameworks for enhanced tumor chemoradiation

Due to their complementary activity,the use of DnA damage repair(DDR)inhibitors during radiotherapy(RT)has yielded promising ***,this approach is often hindered by toxicity and poor in vivo stability of the DDR in *** scale metal-organic frameworks(nMOFs)represent an emerging class of crystalline materials which exhibit advantageous properties over traditional nanomaterials and demonstrate great potential in ***,a unique,synergistic treatment strategy for enhancing the therapeutic efficacy of RT vianMOF-mediated drug delivery and RT enhancement was evaluated.A nMOF containing the high-Zelement Hf and the ligand 1,4-dicarboxybenzene(Hf-BDC)was synthesized and loaded with the two DDR inhibitor drugs,talazoparib and buparlisib(TB@Hf-BDC-PEG).TB@Hf-BDC-PEGaugmented RT by increasing reactive oxygen species generation and thus DnA damage of which repair was inhibited *** enhancement was demonstrated in vivo where the combination of concurrent radiation with in travenous TB@Hf-BDC-PEG administration resulted in improved tumor control and increased ***,no apparent toxicity was observed with nMOF treatment,supporting its potential as an attractive candidate over traditional nano *** work provides the first report of nMOFs employed to enhance thetherapeutic response to RT through DDR inhibitor delivery and physical radiation dose enhancement.

Effect of protease supplementation on apparent ileal crude protein and amino acid digestibility of over-processed soybean meals in broilers

Background: nutritional value of proteins in feed ingredients can be negatively affected by hydrothermal processing, which causes large variation in the bioavailability of amino acids(AA) and negatively affects animal productive performance. Supplementation of exogenous proteases could increase the rate of digestion of damaged proteins, thereby increasing overall AA digestibility and bioavailability. The aim was to determine the effect of exogenous protease supplementation on the apparent ileal digestibility(AID) of crude protein(CP) and AA of soybean meals(SBM) with different degrees of hydrothermal processing in ***: The experiment involved a 3 × 2 factorial arrangement, with SBM processing time(commercial SBM or autoclaved for 30 or 60 min at 120 ℃) and protease supplementation(not supplemented and supplemented) as factors. Protease was included at three times the recommended dose(0.06%) and the experimental diets were fed from 15 to 21 ***: The interaction between the effects of SBM processing and protease supplementation was significant for the AID of CP(P = 0.01), Trp(P = 0.01), Gly(P = 0.03) and Pro(P = 0.03), and also for the average daily gain(P = 0.01)and feed conversion ratio(P = 0.04). Increasing the processing time of SBM decreased(P no acids, whilst the effect of protease supplementation was only significant for the AID of Phe(P = 0.02) and Tyr(P = 0.01).Conclusions: Exogenous protease supplementation at three times the commercial dose does not seem to offset the negative effects of hydrothermal processing of SBM on the apparent ileal digestibility of CP and amino acids or performance of broilers. Whilst positive numerical improvements of digestibility and performance(ADG and FCR)were noticed with protease supplementation at relatively mild processing levels, negative results were obtained with the harsh-processed meals.

Preoperative virtual video visits only:a convenient option that should be offered to caregivers beyond the pandemic

Objective During the COVID-19 pandemic,our group implemented preoperative video visits(VVs)to limit physical *** aim of this study was to determine caregivers’and providers’perceptions of this practice and to determine feasibility for *** All patients who had only a preoperative VV prior to an elective surgery were identified from March-October ***,surgeons,and clinic staff were surveyed about their *** Thirty-four preoperative VVs were followed by an elective surgery without a preceding in-person *** the 31 caregiver surveys completed,the majority strongly agreed that the VV was more convenient(87%,n=27).Eighty-one percent(n=25)strongly agreed or agreed that the VV saved them ***-four percent(n=29)strongly agreed or agreed that they would choose the VV option *** saved an average travel distance of 60.3 miles one way(range 6.1-480).Of the 13/17 providers who responded,77%(n=10)expressed that the practice should *** Virtual health became a necessity during the pandemic,and caregivers were overwhelmingly *** VVs as an option beyond the pandemic may be a reasonable and effective way to help eliminate some of the hurdles that impede healthcare-seeking behavior and should be offered.

Ketamine,a noncompetitive n-methyl-D-aspartate(nMDA)receptor antagonist,has been exclusively used as an anesthetic in medicine and has led to new insights into the pathophysiology of neuropsychiatric *** studies have shown that low subanesthetic doses of ketamine produce antidepressant effects for individuals with ***,its use as a treatment for psychiatric disorders has been limited due to its reinforcing effects and high potential for diversion and *** studies have focused on understanding the molecular mechanisms underlying ketamine’s antidepressant effects,but a precise mechanism had yet to be *** we review different hypotheses for ketamine’s mechanism of action including the direct inhibition and disinhibition of nMDA receptors,aminomethylphosphonic acid receptors(AMPAR)activation,and heightened activation of monoaminergic *** proposed mechanisms are not mutually exclusive,and their combined influence may exert the observed structural and functional neural *** term use of ketamine induces brain structural,functional impairments,and neurodevelopmental effects in both rodents and *** misuse has increased rapidly in the past 20 years and is one of the most common addictive drugs used in *** proposed mechanisms of action and supporting neuroimaging data allow for the development of tools to identify‘biotypes’of ketamine use disorder(KUD)using machine learning approaches,which could inform intervention and treatment.

The design with intent(DwI)toolkit assists designers in creating novel designs and ***,however,is not constrained to any degree,making it impossible to know whether the produced designs adequately account for users’*** contrast,cognitive work analysis(CWA)is a human factors research tool that seeks to map a system and account for users’needs,yet does not provide clear guidelines for progressing such analysis into workable designs with which users can *** paper seeks to present a proof-of-concept investigation to demonstrate that DwI can be suitably constrained and validated by insights gained from ***,in turn,benefits by having a suitable toolkit for progressing *** teams of individuals without design backgrounds were able to develop mock-up in-vehicle interfaces aimed at reducing fuel *** teams were able to use DwI toolkit to articulate the genesis of their ideas,which in turn could be directly linked to system needs identified within CWA.

The epididymal immune balance:a key to preserving male fertility

Experimental Investigation of the Effects of Acute Exercise on Real-World Ecological Memory

Purpose To evaluate the effects of acute moderate-intensity exercise on ecological memory,as assessed from a face-name memory *** A two-arm,parallel-group,randomized controlled intervention was ***(n=40;Mage=20.8 years)were randomized into a seated control task or a bout of acute moderate-intensity treadmill exercise(15-min).Thereafter,participants completed a 3-phase face-name memory task,involving a study phase and two test phases(immediate and delayed recall,with the delay occurring 15 min after the immediate recall).Results For the immediate memory recall,the mean(SD)scores for the exercise and control conditions,respectively,were 6.60(2.5)and 6.20(2.5).For the 15-min delayed assessment,the respective scores were 6.25(2.6)and 5.75(1.9).There was a significant main effect for time(F=4.06,P=0.05,η^(2)_(p)=0.10).However,there were no main effects for group(F=0.33,P=0.56,η^(2)_(p)=0.01)or time by group interactions(F=0.12,P=0.72,η^(2)_(p)=0.003).Conclusions Despite the exercise group having slightly higher immediate and delayed face-name memory scores,we did not observe robust evidence of acute exercise enhancing face-name memory performance.

The PRISMS-Fatigue open-source framework for simulation-based analysis of microstructural influences on fatigue resistance for polycrystalline metals and alloys is presented *** framework uses the crystal plasticity finite element method as its microstructure analysis tool and provides a highly efficient,scalable,flexible,and easy-to-use ICME community *** PRISMS-Fatigue framework is linked to different open-source software to instantiate microstructures,compute the material response,and assess fatigue indicator *** performance of PRISMS-Fatigue is benchmarked against a similar framework implemented using *** indicate that the multilevel parallelism scheme of PRISMS-Fatigue is more efficient and scalable than ABAQUS for large-scale fatigue *** performance and flexibility of this framework is demonstrated with various examples that assess the driving force for fatigue crack formation of microstructures with different crystallographic textures,grain morphologies,and grain numbers,and under different multiaxial strain states,strain magnitudes,and boundary conditions.
