Cytogenetic Mechanism for the Aneuploidy and Mosaicism Foundin Tetraploid Pacific Oyster Crassostrea gigas （Thunberg）

Chromosome constitution was investigated in adult tetraploid Pacific oyster produced by blocking the first polar body oftriploid eggs which were fertilized with haploid sperms. A high incidence of aneuploid and heteroploid mosaics were found amongthe offspring. Of 20 individuals identified, only 9 （45%） were eutetraploid which contained 40 chromosomes; 2 （10%） were ane-uploid （hypotetraploid）, which contained 39 and 38 chromosomes, respectively; and 9 （45%） were heteroploid mosaics. One mosaicwas consisted of cells containing 40 and 39 chromosomes, respectiovely （1：1 in cell number）, while the other 8 were consisted ofcells containing chromosomes varying between tetraploid and triploid. It was also interesting to note that 3 mosaics even containedsome diploid cells with 20 chromosomes. A certain number of cells of 2 tetraploids and 8 mosaics spread with 32-37 well-scatteredand some clumped chromosomes at metaphase. The percentage of aneuploid cells with chromosomes varying between triploid andtetraploid correlated significantly with that of heteroploid mosaics cells with clumping chromosomes （P〈0.05）. Our findings sug-gested that reversion existed in both tetraploid and triploid oyster and chromosome clumping may underline the chromosome elimi-nation in tetraploid oyster. It seems that the reversing cells, at least some of them, continuously eliminate their chromosomes until themost stable diploid state is established.

Analysis of Oxides Formed on the Surface of the Alloy 690 in Hydrogenated Supercritical Water

The oxides formed on the surface of the alloy 690 in hydrogenated supercritical water at 400℃ for 1000 h were investigated using scanning electron microscopy,transmission electron microscopy,scanning transmission electron microscopy and energy-dispersive X-ray *** oxides on me surface of the alloy 690 exhibited multi-layer structure：an outer layer consisted of granular crystallites（NiO and NiFe_2O_4） and a compact inner layer（spinel and Cr_2O_3）.Chemical analysis indicated that the outer layer was enriched in nickel but depleted in chromium,whereas the inner layer was enriched in chromium and iron but depleted in *** inner layer was also characterized as layered structure by Fe-rich spinel on top of continuous Cr_2O_3 ***,Cr_2O_3 nodules were readily observed at the oxides/alloy interface.

Design of Continuous and Discrete LQI Control Systems with Stable Inner Loops

Design approaches were proposed for both continuous and discrete LQI (linear quadratic with integral) controllers, if exist, to stabilize the inner loops in addition to stabilizing the closed loops and minimizing the quadratic cost functionals. Derived from the algebraic Riccati equations involved in continuous and discrete LQI control, the design approaches were straightforward obtained by setting the quadratic cost functionals to decoupled forms with positive definite weighting matrices. Examples were provided to verify the effectiveness of the approaches.

On environmental threats to male infertility

The possibility that environmental pollutants damage human fertility is a topic of both public concern and scientific interest. The effect of environmental factors on male fertility （semen parameters） has been a special focus, stimulated in part by the dramatic damage inflicted by the pcstlcide DBCP on testicular function. Extensive searches for testicular toxicants （in particular, the proposed effects of endocrine-disrupting compounds through fetal exposure） have produced little hard evidence of impairment in humans. This scarcity of evidence may reflect the difficultles of studying human fertility. Future fertifity studies should consider the wide spectrum of environmental exposures that plausibly affect reproduction.

Screening antioxidants for the protection of cochlear sensory cells

Sensory cell damage is a major cause of hearing loss： Sensorineural hearing loss （SNHL） is a common sensory deficit characterized by tissue damage and/or cell death in the cochlea. Moderate and severe SNHL causes deficits in communication, associated with increased isolation from human relationships, and negativly impacts employabflity. Many etiologies have been associated with SNHL： Noise overexposure, certain drugs, inner or middle ear infection or immune-induced inflammation being common. However, the most prevalent form of SNHL is presbycusis or age-related hearing loss, often aggravated by other factors including a history of noise exposure, diabetes or high blood pressure （Wong and Ryan, 2015）. Though systemic steroids are used in the treatment of sudden SNHL, other drugs to prevent or cure hearing loss have not yet been developed. Cochlear sensory cells, known as hair cells （HCs） and neurons have very limited repair capacity. Moreover since these cells do not regenerate, making any loss permanent, prevention of cell damage is critical to protecting hearing.

Adoptive chimeric antigen receptor(CAR)-engineered natural killer(NK)cells have shown promise in treating various ***,limited immunological memory and access to sufficient numbers of allogenic donor cells have hindered their broader preclinical and clinical ***,we first assess eight different CAR constructs that use an anti-PD-L1 nanobody and/or universal anti-fluorescein(FITC)single-chain variable fragment(scFv)to enhance antigen-specific proliferation and anti-tumor cytotoxicity of NK-92 cells against heterogenous solid *** next genetically engineer human pluripotent stem cells(hPSCs)with optimized CARs and differentiate them into functional dual CAR-NK *** tumor microenvironment responsive anti-PD-L1 CAR effectively promoted hPSC-NK cell proliferation and cytotoxicity through antigen-dependent activation of phosphorylated STAT3(pSTAT3)and pSTAT5 signaling pathways via an intracellular truncated IL-2 receptorβ-chain(ΔIL-2Rβ)and STAT3-binding tyrosine-X-X-glutamine(YXXQ)***-tumor activities of PD-L1-induced memory-like hPSC-NK cells were further boosted by administering a FITC-folate bi-specific adapter that bridges between a programmable anti-FITC CAR and folate receptor alpha-expressing breast tumor ***,our hPSC CAR-NK engineering platform is modular and could constitute a realistic strategy to manufacture off-the-shelf CAR-NK cells with immunological memory-like phenotype for targeted immunotherapy.

On the gating of mechanosensitive channels by fluid shear stress

Mechanosensation is an important process in biological fluid-structure interaction. To understand the biophysics underlying mechanosensation, it is essential to quantify the correlation between membrane deformation, membrane tension, external fluid shear stress, and conformation of mechanosensitive (MS) channels. Smoothed dissipative particle dynamics (SDPD) simulations of vesicle/cell in three types of flow configurations are conducted to calculate the tension in lipid membrane due to fluid shear stress from the surrounding viscous flow. In combination with a simple continuum model for an MS channel, SDPD simulation results suggest that shearing adhered vesicles/cells is more effective to induce membrane tension sufficient to stretch MS channels open than a free shear flow or a constrictive channel flow. In addition, we incorporate the bilayer-cytoskeletal interaction in a two-component model to probe the effects of a cytoskeletal network on the gating of MS channels.
