Background:The most used protein sources in ruminant nutrition are considered as having negative impacts in terms of environmental sustainability and competition with human ***,the investigation of alternative and sustainable feedstuffs is becoming a priority in ruminant production ***:This trial was designed to evaluate eight full-fat insect meals(Acheta domesticus–ACD;Alphitobius diaperinus–ALD;Blatta lateralis–BL;Gryllus bimaculatus–GB;Grylloides sygillatus–GS;Hermetia illucens–HI;Musca domestica–MD;and Tenebrio molitor–TM)as potential protein and lipid sources in ruminant *** parameters and fatty acids(FA)of rumen digesta after 24-h in vitro ruminal incubation of the tested insect meals were measured and compared with those of three plant-based meals(soybean meal,rapeseed meal and sunflower meal)and fishmeal(FM).Similarly to FM,the insect meals led to a significantly lower total gas production(on average,1.75vs.4.64 mmol/g dry matter-DM),methane production(on average,0.33 vs.0.91 mmol/g DM),volatile FA production(on average,4.12 vs.7.53 mmol/g DM),and in vitro organic matter disappearance(on average,0.32 vs.0.59 g/g)than those observed for the plant *** insect meals also led to lower ammonia of rumen fluid,when expressed as a proportion of total N(on average,0.74 vs.0.52 for the plant and insect meals,respectively),which could be an advantage provided that intestinal digestibility is *** in ruminal fermentation parameters between the insect meals could be partially explained by their chitin,crude protein and ether extract contents,as well as by their FA *** particular,high content of polyunsaturated FA,or C12:0(in HI),seems to partially inhibit the ruminal ***:The tested full-fat insect meals appear to be potentially an interesting protein and lipid source for ruminants,alternative to the less sustainable and commonly used ones of plant *** FA profile of the rum

抗体是一类Y字形的蛋白分子，在机体内识别数百种自身或非自身的成分。抗体是后天免疫系统的核心成分，以一种可溶性的方式存在于B细胞表面或血液中。人们通过不同类型的哺乳动物表达载体来表达人源化抗体并将其投入使用。对于抗体的研究已经成为生物学及现代医学关注的热点话题。通过基因工程改造而产生的抗体药物的应用为很多疾病的治疗带来了新的希望。抗体药物与化学药物的联合应用刺激了新生药物的产生。本研究的主要内容即构建了人源化抗体的哺乳动物表达载体，并将其转入到大肠杆菌中。反应中用到的寡聚核苷酸来自H1c9序列，并分别在5’及3’端插入BglII和EcoRI限制性内切酶位点。哺乳动物表达载体使用的是pEGFPC1。PCR产物及酶切产物采用1%的琼脂糖凝胶电泳，目的片段通过凝胶回收试剂盒进行回收。连接后将连接产物转化大肠杆菌感受态。从我们所得到的数据可以看出，人源化单链抗体表达载体pEGFP-C1-hscFv构建成功。

Wilson's disease in Lebanon and regional countries: Homozygosity and hepatic phenotype predominance

AIM To determine the phenotypes and predominant diseasecausing mutations in Lebanese patients with Wilson's disease,as compared to regional non-European *** The clinical profile of 36 patients diagnosed in Lebanon was studied and their mutations were determined by molecular *** patients underwent full physical exam,including ophthalmologic slit-lamp examination ultrasound imaging of the liver,as well as measurement of serum ceruloplasmin and 24-h urinaryCu *** addition,genetic screening using PCR followed by sequencing to determine disease-causing mutations and polymorphisms in the ATP7B gene was carried on extracted DNA from patients and immediate family *** phenotypic-genotypic findings were then compared to reported mutations in Wilson's disease patients from regional Arab and non-European countries. The most common mutation,detected in 4 out of 13families,involved the ATP hinge region and was present in patients from Lebanon,Egypt,Iran and ***,mutations in Lebanese patients and those of the region were scattered over 17 exons of *** the homozygous exon 12 mutation Trp939Cys was only detected in patients from Lebanon but none from the regional countries,the worldwide common mutation H1069Q was not present in the Lebanese and was rare in the *** hepatic phenotype was predominant in patients from both Lebanon and the region(25%-65%).Furthermore,the majority of patients,including those who were asymptomatic,had evidence of some hepatic *** neurologic phenotype was rare. CONCLUSION Findings do not support presence of a founder *** and genetic screening is recommended for family members with index patients and unexplained hepatic dysfunction.

Corneal stem cells and tissue engineering: Current advances and future perspectives

Major advances are currently being made in regenerative medicine for cornea. Stem cell-based therapies represent a novel strategy that may substitute conventional corneal transplantation, albeit there aremany challenges ahead given the singularities of each cellular layer of the cornea. This review recapitulates the current data on corneal epithelial stem cells, corneal stromal stem cells and corneal endothelial cell progenitors. Corneal limbal autografts containing epithelial stem cells have been transplanted in humans for more than 20 years with great successful rates, and researchers now focus on ex vivo cultures and other cell lineages to transplant to the ocular surface. A small population of cells in the corneal endothelium was recently reported to have self-renewal capacity, although they do not proliferate in vivo. Two main obstacles have hindered endothelial cell transplantation to date: culture protocols and cell delivery methods to the posterior cornea in vivo. Human corneal stromal stem cells have been identified shortly after the recognition of precursors of endothelial cells. Stromal stem cells may have the potential to provide a direct cell-based therapeutic approach when injected to corneal scars. Furthermore, they exhibit the ability to deposit organized connective tissue in vitro and may be useful in corneal stroma engineering in the future. Recent advances and future perspectives in the field are discussed.

Gene capture by transposable elements leads to epigenetic conflict in maize

Deciphering the role of PGC-1α in neurological disorders: from mitochondrial dysfunction to synaptic failure

The onset and mechanisms underlying neurodegenerative diseases remain uncertain. The main features of neurodegenerative diseases have been related with cellular and molecular events like neuronal loss, mitochondrial dysfunction and aberrant accumulation of misfolded proteins or peptides in specific areas of the brain. The most prevalent neurodegenerative diseases belonging to age-related pathologies are Alzheimer's disease, Huntington's disease, Parkinson's disease and amyotrophic lateral sclerosis. Interestingly, mitochondrial dysfunction has been observed to occur during the early onset of several neuropathological events associated to neurodegenerative diseases. The master regulator of mitochondrial quality control and energetic metabolism is the transcriptional coactivator peroxisome proliferator-activated receptor gamma coactivator 1-alpha(PGC-1α). Additionally, it has been observed that PGC-1α appears to be a key factor in maintaining neuronal survival and synaptic transmission. In fact, PGC-1α downregulation in different brain areas(hippocampus, substantia nigra, cortex, striatum and spinal cord) that occurs in function of neurological damage including oxidative stress, neuronal loss, and motor disorders has been seen in several animal and cellular models of neurodegenerative diseases. Current evidence indicates that PGC-1α upregulation may serve as a potent therapeutic approach against development and progression of neuronal damage. Remarkably, increasing evidence shows that PGC-1α deficient mice have neurodegenerative diseases-like features, as well as neurological abnormalities. Finally, we discuss recent studies showing novel specific PGC-1α isoforms in the central nervous system that appear to exert a key role in the age of onset of neurodegenerative diseases and have a neuroprotective function in the central nervous system, thus opening a new molecular strategy for treatment of neurodegenerative diseases. Glycine-extended gastrin activates two independent tyrosine-kinases in upstream of p85/p110 phosphatidylinositol 3-kinase in human colonic tumour cells

AIM： To investigate whether Src, JAK2 and phosphatidylinositol 3-kinase （PI3K） pathways are involved in the proliferation of human colonic tumour cells induced by glycine-extended gastrin （G-gly）, the precursor of the mature amidated gastrin and to elucidate the molecular interaction between these three kinases in response to this peptide. METHODS： Using the human colonic tumour cell line HCT116 as a model, we first measured the activation of PI3K, p60-Src and JAK2 in response to G-gly by in vitro kinase assays. Then we investigated the involvement of these kinases in G-gly-induced cell proliferation by MTT test. RESULTS： G-gly stimulation induced p60-Src, JAK2 and PI3K activation in HCT116. The different pathways were involved in proliferation of human colon cancer cells induced by G-gly. Furthermore, we found that both Src and JAK2 were necessary to PI3K regulation by this peptide. However, we did not find any cross-talk between the two tyrosine kinases. CONCLUSION： Our results suggest that the p60-Src/ PI3K and JAK2/PI3K pathways act independently to mediate G-gly proliferative effect on human colonic tumour cells.
