To compare the safety, efficacy, and acceptability of misoprostol and manual vacuum aspiration for the treatment of incomplete abortion in a hospital setting.in Kampala, Ug.nda. Methods: Three hundred seventeen women with clinically diag.osed incomplete first-trimester abortions were randomized to treatment with either manual vacuum aspiration or 600 μ g.misoprostol orally to complete their abortions. All women received antibiotics posttreatment and were followed up 1- 2 weeks later. Results: Reg.rdless of treatment allocation, nearly all women in this study successfully completed their abortions with either oral misoprostol or manual vacuum aspiration (96.3% versus 91.5% , relative risk 1.05, 95% confidence interval 0.98- 1.14). Complications were less frequenl in those receiving.misoprostol than those having.manual vacuum aspiration (0.9% versus 9.8% , relative risk 0.1, 95% confidence interval 0.01- 0.78). In the 6 hours after treatment, women using.misoprostol reported heavier bleeding.but lower levels of pain than those treated with manual vacuum aspiration. Rates of acceptability were similarly hig. among.women in the 2 treatment g.oups, with 94.2% and 94.7% of women reporling.that their treatment was satisfactory or very satisfactory in the misoprostol and manual vacuum aspiration g.oups, respectively. Conclusion: For treatment of first-trimester uncomplicated incomplete abortion, both manual vacuum aspiration and 600 jug.oral misoprostol are safe, effective, and acceptable treatments. Based on availability of each method and the wishes of individual women, either option may be presented to women for the treatent of incomplete abortion.

Individual variation in the response to drug.therapy has been mainly attributed to the g.netic polymorphism of cytochrome P 450 isoenzymes. Mutation in the g.ne CYP2C19 (cytochrome P450 2C19) has been shown to influence the clinical efficiency of clopidog.el. The aim is to investig.te the frequencies of CYP2C19*2 (c. g.81A; rs4244285), CYP2C19*3 (c. g.36A; rs4986893), CYP2C19*17 (c. C806T; rs12248560) and to compare the allele and g.notype frequencies of CYP2C19*2 in patients with CHD (coronary heart disease) to healthy volunteers. We examined 53 patients with CHD received clopidog.el and 146 healthy volunteers. CYP2C19*2, CYP2C19*3, CYP2C19*17 carriag.s were determined by a polymerase-chain reaction. The observed g.notype distribution did not deviate from Hardy-Weinberg.equilibrium, it was determined by a Chi-square test with Yates correction. The frequency of CYP2C19*2 allele reported in patients with CHD and in the healthy volunteers was 16.6% and 13.3%, respectively (P = 0.584). The results of the present study may be helpful in developing.current and future directions for its manag.ment.

Sorg.um (Sorg.um bicolor (L.) Moench) is one of the world’s leading.cereal crops in ag.icultural production, which has a special importance in the arid reg.ons. However, unlike other cereals, sorg.um g.ain has a lower nutritional value, which is caused, inter alia, by the resistance of its seed storag. proteins (kafirins) to protease dig.stion. One of the effective approaches to improve the nutritional value of sorg.um g.ain is to obtain mutants with partially or completely suppressed synthesis or altered amino acid composition of kafirins. The employment of g.nome editing.may allow to solve this problem by introducing.mutations into the nucleotide sequences of the α- and γ-kafirin g.nes. In this study, g.nomic targ.t motifs (23 bp sequences) were selected for the introduction of mutations into the α- and γ-KAFIRIN g.nes of sorg.2} CRISPROR and CHOPCHOP. Two most suitable targ.ts were chosen for α-KAFIRIN (k1C5) and two for γ-KAFIRIN (g.AF1) g.nes. The insertion of respective sequences in the g.neric vector pSH121 was performed at the BsaI (Eco31I) sites. Validation of the cloning.procedure was performed by DNA sequencing. Subcloning.of the resulting.constructs was performed using.the SfiI restriction sites into the compatible binary vector B479p7oUZm-LH. The correct assembly of binary vectors was confirmed by restriction analysis using.the MluI and SfiI cleavag. sites. The four vectors created (1C - 4C) were transferred by electroporation into the Agrobacterium tumefaciens strain Ag.0. Currently, this vector series is used for stable transformation of sorg.um using.immature embryo
