Outbreaks of Monkeypox(mpox)in over 100 non-endemic countries in 2022 represented a serious global health *** a neglected disease,mpox has become a global public health issue.A42R profilinlike protein from mpox(PDB ID:4QWO)represents a potential new lead for drug development and may interact with various synthetic and natural *** this report,the interaction of A42R profilinlike protein with six phytochemicals found in the medicinal plant Ficus religiosa(abundant in India)was *** on the predicted and compared protein–ligand binding energies,biological properties,IC_(50) values and toxicity,two compounds,kaempferol(C-1)and piperine(C-4),were *** characteristics and quantitative structure–activity relationship(QsAR)of these two compounds were determined,and molecular dynamics(MD)simulations were *** silico examination of the kaempferol(C-1)and piperine(C-4)interactions with A42R profilin-like protein gave best-pose ligandbinding energies of–6.98 and–5.57 kcal/mol,*** predicted IC_(50) of C-1 was 7.63 lM and 82 lM for *** data indicated that kaempferol and piperine are non-mutagenic,and the QsAR data revealed that piperlongumine(5.92)and piperine(5.25)had higher log P values than the other compounds *** simulations of A42R profilin-like protein in complex with C-1 and C-4 were performed to examine the stability of the ligand–protein ***/C and C-4 showed the highest affinity and activities,they may be suitable lead candidates for developing mpox therapeutic *** study should facilitate discovering and synthesizing innovative therapeutics to address other infectious diseases.

Response surface methodology-based optimization of ultrasound-assisted extraction of β-sitosterol and lupeol from Astragalus atropilosus(roots) and validation by HPTLC method

Objective:To optimize the ultrasonication method for efficient extraction ofβ-sitosterol and lupeol from the roots of Astragalus atropilosus using Box-Behnken design of response surface methodology(RsM),and its validation by high performance thin layer chromatography(HPTLC)***:Ultrasonication method was used to extractβ-sitosterol and lupeol from Astragalus atropilosus(roots).RsM was used to optimize the different extraction parameters *** to solid ratio(10–14 m L/g),temperature(60-80℃)and time(40–60 min)to maximize the yield ofβ-sitosterol and *** quantitative estimation ofβ-sitosterol and lupeol was done in chloroform extract of Astragalus atropilosus by validated HPTLC method on 10 cm×20 cm glass-backed silica gel 60 F254 plate using hexane and ethyl acetate(8:2,v/v)as mobile ***:A quadratic polynomial model was found to be most appropriate with regard to R1(yield of total extraction;R2/%CV=0.9948/0.28),R2(β-sitosterol yield;R2/%CV=0.9923/0.39)and R3(lupeol yield;R2/%CV=0.9942/0.97).The values of adjusted R2/predicted R2/signal to noise ratio for R1,R2,and R3 were 0.9782/0.9551/48.77,0.9904/0.9110/31.33,and 0.9927/0.9401/36.08,respectively,indicating a high degree of correlation and adequate *** linear correlation plot between the predicted and experimental values for R1,R2,and R3 showed high values of R2 ranging from 0.9905-0.9973.β-sitosterol and lupeol in chloroform extract of Astragalus atropilosus were detected at Rf values of 0.22 and 0.34,respectively,atλmax=518 *** optimized ultrasonic extraction produced 8.462%w/w of R1,0.451%w/w of R2 and 0.172%w/w of R3 at 13.5 m L/g liquid to solid ratio,78℃of temperature and 60 min of ***:The experimental findings of RsM optimized extraction and HPTLC analysis can be further applied for the efficient extraction ofβ-sitosterol and lupeol in other species of Astragalus.

沙特阿拉伯王国是一个信奉传统医学的国家,药用植物的使用十分普遍。《沙特药典》中收载了47科96种药用植物,其中苋科植物最多,有7种,菊科、夹竹桃科和豆科分别有5种;草本植物和亚灌木植物分别占43%和30%;最常用的制剂方式为汤剂和灌肠剂;使用最多的药用部位为全草、叶、种子、地上部分,分别占29%、28%、7%和5%。通过查阅文献,对沙特阿拉伯王国常用药用植物的基本信息进行总结,为传统医学的合理用药提供参考。

Internet of Things(IoTs)provides better solutions in various fields,namely healthcare,smart transportation,home,*** Denial of service(Dos)outbreaks in IoT platforms is significant in certifying the accessibility and integrity of IoT *** learning(DL)models outperform in detecting complex,non-linear relationships,allowing them to effectually severe slight deviations fromnormal IoT activities that may designate a Dos *** uninterrupted observation and real-time detection actions of DL participate in accurate and rapid detection,permitting proactive reduction events to be executed,hence securing the IoT network’s safety and ***,this study presents pigeon-inspired optimization with a DL-based attack detection and classification(PIODL-ADC)approach in an IoT *** PIODL-ADC approach implements a hyperparameter-tuned DL method for Distributed Denial-of-service(DDos)attack detection in an IoT ***,the PIODL-ADC model utilizes Z-score normalization to scale input data into a *** handling the convolutional and adaptive behaviors of IoT,the PIODL-ADCmodel employs the pigeon-inspired optimization(PIO)method for feature selection to detect the related features,considerably enhancing the recognition’s ***,the Elman Recurrent Neural Network(ERNN)model is utilized to recognize and classify DDos ***,reptile search algorithm(RsA)based hyperparameter tuning is employed to improve the precision and robustness of the ERNN method.A series of investigational validations is made to ensure the accomplishment of the PIODL-ADC *** experimental outcome exhibited that the PIODL-ADC method shows greater accomplishment when related to existing models,with a maximum accuracy of 99.81%.

In the contemporary era,the global expansion of electrical grids is propelled by various renewable energy sources(REss).Efficient integration of stochastic REss and optimal power flow(OPF)management are critical for network *** study introduces an innovative solution,the Gaussian Bare-Bones Levy Cheetah Optimizer(GBBLCO),addressing OPF challenges in power generation systems with stochastic *** primary objective is to minimize the total operating costs of REss,considering four functions:overall operating costs,voltage deviation management,emissions reduction,voltage stability index(VsI)and power loss ***,a carbon tax is included in the objective function to reduce carbon *** scrutiny,using modified IEEE 30-bus and IEEE 118-bus systems,validates GBBLCO’s superior performance in achieving optimal *** results demonstrate GBBLCO’s efficacy in six optimization scenarios:total cost with valve point effects,total cost with emission and carbon tax,total cost with prohibited operating zones,active power loss optimization,voltage deviation optimization and enhancing voltage stability index(VsI).GBBLCO outperforms conventional techniques in each scenario,showcasing rapid convergence and superior solution ***,GBBLCO navigates complexities introduced by valve point effects,adapts to environmental constraints,optimizes costs while considering prohibited operating zones,minimizes active power losses,and optimizes voltage deviation by enhancing the voltage stability index(VsI)*** research significantly contributes to advancing OPF,emphasizing GBBLCO’s improved global search capabilities and ability to address challenges related to local *** emerges as a versatile and robust optimization tool for diverse challenges in power systems,offering a promising solution for the evolving needs of renewable energy-integrated power grids.
