The perennial Medicago sativa cv.Gabès is widely grown on saline soils in Tunisian oases.Morphological and physiological analyses of two populations (Mareth and Gannouch) were conducted in order to study the efiect of salinity and gamma radiation (350 Gy) inter-action on two populations of this species.It has been shown that the two irradiated populations of Medicago sativa are fairly tolerant to salt at growth phase compared to the non-irradiated.Exposure to gamma irradiation (350 Gy), alone or in combination with salt stress, increased significantly (p alinity around 5 g/l.Such a mechanism refiects probably an inclusive behaviour of the plants and a good aptitude to use the dominant ions (Na+) for the osmotic adjustment.However, the non-irradiated populations are unable to adjust their internal osmotic potential.Consequently, irradiated plants hav

Genetic diversity of rhizobial populations recovered from three Lotus species cultivated in the infra-arid Tunisian soils

Eighty-three bacterial strains isolated from root nodules of Lotus creticus, L. pusillus, and L. arabicus grown in infra-arid Tunisian soils were characterized using a polyphasic approach including phenotypic analysis, rep-PCR and PCR-RFLP analyses of the 16S rRNA gene. Phenotypically, all isolates are fast growers the majority of which grow at a pH of between 5.5 and 9. Most of the tested isolates tolerate NaCl concentrations from 1.39% to 3.48%. By rep-PCR fingerprinting, the genomic similarity varied from 30% to 98%. All tested isolates were clustered into 32 rep-PCR clusters at the similarity level of 80%. The genomic divergence of strains revealed by rep/PCR analysis appeared to be very important since a molecular polymorphism delimiting symbionts for each species of Lotus was identified. With the high-resolution of rep-PCR profiles of the isolates obtained using Pearson’s/UPGMA analysis, the isolates were resolved into 60 different profiling groups to undergo 16S ARDRA analyses. The analysis of all restriction fragments from each strain based on the UPGMA algorithm from the combined patterns showed that Lotus isolates are very diverse and that they were affiliated to Sinorhizobium, Rhizobium, and Mesorhizobium genera.

Genetic Diversity of Tunisian and Chinese Alfalfa (Medicago sativa L.) Revealed by RAPD and ISSR Markers

Medicago sativa L.) is one of the most important forage crops in the world. The genetic variability analysis of 19 alfalfa populations collected from three sites in South Tunisia (Gabes, Kebili, Tozeur) and 1 from North West China were carried out using Random Amplified Polymorphic DNA (RAPD) and Inter Simple Sequence Repeat (ISSR) markers. Five RAPD primers amplified 44 bands of which 22 were polymorphic;and five ISSR primers amplified 51 bands of which 33 were polymorphic. The percentage of polymorphic bands detected by RAPD and ISSR was 50% and 64.7%, respectively. The resolving power (Rp) varied between 0.6 and 4.1 with an average of 2.02 for RAPD marker and between 0.7 and 6.5 with an average of 2.28 for ISSR marker. However the Average Informativeness band (AvIb) was ranged from 0.2 to 0.9 with an average of 0.5 in RAPD marker and from 0.29 to 0.7 with an average of 0.624 in ISSR marker. The RAPD marker revealed less within population genetic diversity than ISSR marker. Although Cluster (UPGMA) and Correspondence Factorial Analyses (CFA) indicate that populations’ clustering made independently both from the geographical origin.

The Saharian ecosystems present an important intensity of rising sensitization to: Erosion, and desertification whose impacts are irreversible. On the one hand, the weakness of the yield and the poverty of soil lead to a limited biodiversity. In addition to these difficulties, hydra is the main cause of the rarification of certain pastoral species such as Cenchrus ciliaris and Digitaria commutata. The ecological study underlines a distribution of these species which are very dependent on water resources in the dry regions of Tunisia. The bioclimatic (temperature, pluviometry) variations lead to modifications to these species from one area to another which are translated through several parameters. Concerning the number of bundles, the difference is important. The national park of Bouhedma records the highest number, compared with Bni khdach, Jerba, Khanguit aicha and Matmata respectively. The variation inter-site of study also is considerable by the study of the morphological parameters (height, number and length of leaves by bundle, number of ear) whose bundles of the national park Bouhedma occupy the first class.

Relationships between Fruit Acceptability and Health-Case of Seven Pomegranate (Punica granatumL.) Juices

The objective of this study was to evaluate acceptability and instrumental measurements (titratable acidity, total soluble sugar, total phenolic, flavonoids, anthocyanins, and condensed tannin contents as well as antioxidant capacity) of seven Tunisian pomegranate cultivars to relate fruit acceptability and beneficial health. Seventy one participants were asked to complete a questionnaire and to indicate their level of acceptance, on a nine point-hedonic scale, for fruit size, sweetness, bitterness, juice color and juice overall acceptability. Taste and aril color were the main criteria to select Pomegranate. Significant differences occurred for all the acceptability tests. Titratable acidity determines the taste and important inter-specific variability in the phenolic contents as well as for the antioxidant capacities was noticed. Anthocyanin and condensed tannin were contributing to the organoleptic properties while bitterness acceptability was negatively correlated to the antioxidant capacity. Also, a relationship was found between the fruit size acceptability and the antioxidant capacity.
