A new animal model for uterine torsion and uterine ischemia-reperfusion studies, but not fetal hypoxia studies

The aim of the present study was to develop a new animal model for use in uterine torsion, uterine ischemia-reperfusion, and fetal hypoxia studies in rats. A total of 14 pregnant rats on their 18 th-19 th gestational days were used. The animals were randomly divided into two groups: those undergoing the shame operation(group 1),and those in which a 360 uterine torsion was performed using a novel technique,which was corrected 6 hours later(group 2). Subsequently, seven female and seven male rat pups aged 1 month were separated from the mothers in each group. The female rats were monitored until puberty via measuring the vaginal apertures. The 1-month old male rats and the female rats on reaching puberty were decapitated and histopathological tests were performed on the dissected organs, including the cerebral, visceral and genital organs. At the end of the study, no differences were observed between the groups with regard to abortions, offspring death rates and congenital abnormalities. It was observed that the time to reach puberty in female rats born from mothers with uterine torsion was longer, but the difference was statistically insignificant. No microscopic lesions were detected in the cerebral, visceral or genital organs of the offspring. Accordingly, it was concluded that offspring of mothers with the uterine torsion were not affected, at least in the short term. It was generally concluded that this animal model is appropriate for use in uterine torsion and ischemia-reperfusion studies, but is not appropriate for fetal hypoxia studies.

AIM: To investigate the oxidative-stress-related changes in rats with portal hypertension with particular emphasis on nitric oxide (NO) and trace metals. METHODS: Cirrhosis was induced by partial portal vein ligation (PVL) in Wistar rats. The lipid peroxidation marker (malondialdehyde, MDA), antioxidant defense enzymes including superoxide dismutase (SOD), catalase (CAT), glutathione (GSH), and agents known to have antioxidant features including nitric oxide (NO), zinc (Zn), copper (Cu) were determined both in serum and in liver tissue at 4 wk after surgery in PVL and sham-operated rats. Portal pressure of all experimental animals was measured. MDA was detected by thiobarbituric acid reactivity *** activity was determined by inhibition of nitroblue tetrazolium reduction with xanthine/xanthine oxidase used asa superoxide generator. CAT activity was determined by the breakdown of hydrogen peroxide. GSH concentrations were measured by using metaphosphoric acid for protein precipitation and 5'-5'-dithio-bis-2-nitrobenzoic acid for color development. NO was detected by the Griess method after reduction of nitrate to nitrite with nitrate reductase, and the concentrations of Zn and Cu were measured by a Shimadzu 680 AA atomic absorption spectrometer. Histopathological confirmation was done under light microscope. Statistical analyses were done by Student's t-test, and significance of the difference was tested by the unpaired Mann-Whitney test. P＜0.05 was considered statistically ***: Histopathological studies confirmed PVL induced cirrhotic changes. There was a statistically significant difference in portal pressure between PVL and control groups (P＜0.001). The results showed significant increases in the levels of MDA and NO in both tissue and serum (P＜0.05 and P＜0.001, respectively in tissue; P＜0.001 for each in serum), and Zn only in tissue (P＜0.001)in rats with PVL compared with sham-operated rats. Disease modifying anti-rheumatic agents are the cornerstone in management of Rheumatoid Arthritis and when used correctly they are life-saving. As the number of the agents increases, detailed guidelines become more and more important for clinicians to set safe and effective regimens. Herein, we combined the EULAR and ACR recommendations for clinicians and also pointed some important facts peculiar to our country.

Takayasu arteritis is a large vessel vasculitis of the young women with giant cells and granuloma formation. The diagnosis and management of the disease are really not so easy because of the insidious onset and the difficulties in assessment of disease activity. Nearly 60% of the patients are corticosteroid resistant or dependent and relapses are very frequent during taper of the dose [1]. The novel biologic agents as Anti-TNF, Rutiximab and Tocilizumab provide acceptable response rates with low toxicity. Herein, we reviewed the efficacy of Tocilizumab in Takayasu arteritis.

This observational cohort study investigated the potential of a novel sperm-washing medium(SWM)enriched with serotonin(5-HT),L-carnitine(L-C),and coenzyme Q10(CoQ10)to enhance sperm motility and reduce DNA *** compared this innovative medium(5-HT/L-C/CoQ10 SWM)with two widely used commercial media(SWM 1 and SWM 2).Ninety-eight volunteers from an infertility clinic provided semen samples,which were divided into three aliquots for analysis in different SWMs:group 1,SWM was composed of hydroxyethyl piperazineethanesulfonic acid(HEPES),sodium bicarbonate,human serum albumin(HSA),taurine,and gentamicin sulfate(SWM 1);group 2,SWM was composed of HEPES,sodium bicarbonate,and HSA(SWM 2);and group 3,SWM was composed of HEPES-buffered human tubal fluid supplemented with 5-HT,L-C,and CoQ10(5-HT/L-C/CoQ10 SWM).Sperm motility was categorized as progressive,nonprogressive,or ***,reactive oxygen species(ROS)production,and DNA fragmentation were also *** were no significant differences in total or progressive sperm motility among the *** in group 3 exhibited reduced apoptosis,necrosis,and ROS levels and increased *** significant differences were observed in the DNA fragmentation index among *** 5-HT/L-C/CoQ10 SWM reduced sperm oxidative stress and apoptosis compared with those of the two commercially available SWMs,suggesting that 5-HT/L-C/CoQ10 SWM could be useful for enhancing in vitro fertilization success rates.
