A New Ratio-Metric pH Probe,“ThiAKS Green”for Live-Cell pH Measurements

Deviation of the H+concentration from optimum values within the organelles is closely associated with irregular cellular functions that cause the onset of various diseases.Therefore,determining subcellular pH values in live cells and tissues is valuable for diagnostic purposes.In this study,we report a novel ratiometric fluorescence probe 1H-pyrazole-3-carboxylic acid,4-(benzo[d]thiazol-2-yl)-3-(2,4-dihydroxy-3-methylphenyl)-1H-pyrazole-5-carboxylicacid4-(2-benz othiazolyl)-5-(2,4-dihydroxy-3-methylphenyl),to which we will refer as ThiAKS Green(Thiazole akyol shifting green),that is pH sensitive.The results presented here show that the probe can penetrate the celi membrane in iess than 30minutes and does not show any detectable toxicity.The measured color shifts up on pH change are linear and most significant around physiological pH(pKa=7.45),thus making this probe suitable for live-cell imaging and intracellular pH measurements.During the long-incubation periods following the application of the probe and the fluorescent microscopy measurements,it shows stable properties and is easy to detect in live cells.In conclusion,the results suggest that ThiAKS Green can be used to obtain precise information on the H+distribution at various compartments of the live cells.

Cervical adenocarcinoma and adenomatoid tumor of the uterus;A rare coexisting finding and review of the literature

Genital origin adenocarcinomas with coexisting uterine adenomatoid tumors are very rare with only four published cases in the literature. We report a case of adenomatoid tumor of the uterus that was found incidentally during treatment of cervical adenocarcinoma. Although adenomatoid tumors may cause a differential diagnostic problem, in our case the hematoxylin-eosin histological and immunhistochemical studies confirmed mesothelial phenotype of tumor and these also helped in differentiating an adenomatoid tumor from the carcinomatous myometrial invasion. Adenomatoid tumors are easily recognizable benign pathologies. An incidental finding of adenomatoid tumors accompanied by adenocarcinoma of the endometrium or cervix should always be considered.

A rare cause for gestational trophoblastic neoplasia: Placental site trophoblastic tumor

Placental site trophoblastic tumor is a rare sub-group of gestational trophoblastic neoplasia. There is a wide clinical spectrum of presentation and behaviour ranging from a benign condition to an aggressive disease with a fatal outcome. We report a case of placental site trophoblastic tumor in 23-year-old women with irregular vaginal bleeding during postpartum lactation period. In addition to persistent low level β-hCG titers, ultrasound examination revealed a suspicious low-echoic area in the myometrium consistent with gestational trophoblastic disease. After histopathological examination of the specimen achieved by ultrasound-guided dilatation and curettage of the uterus, the placental site trophoblastic tumor diagnosis was made and subsequently total abdominal hysterectomy was performed. The patient had an uneventful recovery, and no recurrence was detected for 40 months in the follow-up period.
