Culture conditions for equine bone marrow mesenchymal stem cells and expression of key transcription factors during their differentiation into osteoblasts

Background： The use of equine bone marrow mesenchymal stem cells （BMSC） is a novel method to improve fracture healing in horses. However, additional research is needed to identify optimal culture conditions and to determine the mechanisms involved in regulating BMSC differentiation into osteoblasts. The objectives of the experiments were to determine： 1） if autologous or commercial serum is better for proliferation and differentiation of equine BMSC into osteoblasts, and 2） the expression of key transcription factors during the differentiation of equine BMSC into osteoblasts. Equine BMSC were isolated from the sterna of 3 horses, treated with purchased fetal bovine serum （FBS） or autologous horse serum （HS）, and cell proliferation determined. To induce osteoblast differentiation, cells were incubated with L-ascorbic acid-2-phosphate and glycerol-2-phosphate in the presence or absence of human bone morphogenetic protein2 （BMP2）, dexamethasone （DEX）, or combination of the two. Alkaline phosphatase （ALP） activity, a marker of osteoblast differentiation, was determined by ELISA. Total RNA was isolated from differentiating BMSC between d 0 to 18 to determine expression of runt-reloted tronscrJption foctor2 （Runx2）, osterix （Osx）, and T-box3 （Tbx3）. Data were analyzed by ANOVA. Results： Relative to control, FBS and HS increased cell number （133 ± 5 and 116 ± 5%, respectively; P 〈 0.001） and 5-bromo- 2＇-deoxyuridine （BrdU） incorporation （167 ± 6 and 120 ± 6%, respectively; P 〈 0.001）. Treatment with DEX increased ALP activity compared with control （1,638 ± 38%; P 〈 0.001）. In the absence and presence of Dex, BMP-2 did not alter ALP activity （P 〉 0.8）. Runt-reloted transcription foctor2 expression increased 3-fold （P 〈 0.001） by d 6 of culture. Osterix expression increased 94old （P 〈 0.05） by d 18 of culture. Expression of Tbx3 increased 1.8-fold at d 3 （P 〈 0.01）; however expression was reduced 4-fold at d 18 （P 〈 0.01）. Declining metabolic scaling parallels an ontogenetic change from elongate to deep-bodied shapes in juvenile Brown trout

Body shape and metabolic rate can be important determinants of animal performance,yet often their effects on influential traits are evaluated in a non-integrated way.This creates an important gap because the integration between shape and metabolism may be crucial to evaluate metabolic scaling theories.Here,we measured standard metabolic rate in 1-and 2-years old juvenile brown trout Salmo trutta,and used a geometric morphometrics approach to extricate the effects of ontogeny and size on the link between shape and metabolic scaling.We evidenced near-isometric ontogenetic scaling of metabolic rate with size,but also a biphasic pattern driven by a significant change in metabolic scaling,from positive to negative allometry.Moreover,the change in metabolic allometry parallels an ontogenetic change from elongate to deep-bodied shapes.This is consistent with the dynamic energy budget(DEB)and surface area(SA)theories,but not with the resource transport network theory which predicts increasing allometric exponents for trends towards more robust,three-dimensional bodies.In addition,we found a relationship between body shape and size independent metabolic rate,with a positive correlation between robustness and metabolic rate,which fits well within the view of Pace-of-Life Syndromes(POLS).Finally,our results align with previous studies that question the universality of metabolic scaling exponents and propose other mechanistic models explaining the diversity of metabolic scaling relationships or emphasizing the potential contribution of ecological factors.

A perceptual quality metric for 3D triangle meshes based on spatial pooling

In computer graphics, various processing operations are applied to 3D triangle meshes and these processes often involve distortions, which affect the visual quality of surface geometry. In this context, perceptual quality assessment of 3D triangle meshes has become a crucial issue. In this paper, we propose a new objective quality metric for assessing the visual difference between a reference mesh and a corresponding distorted mesh. Our analysis indicates that the overall quality of a distorted mesh is sensitive to the distortion distribution. The proposed metric is based on a spatial pooling strategy and statistical descriptors of the dis- tortion distribution. We generate a perceptual distortion map for vertices in the reference mesh while taking into account the visual masking effect of the human visual system. The proposed metric extracts statistical descriptors from the dis- tortion map as the feature vector to represent the overall mesh quality. With the feature vector as input, we adopt a support vector regression model to predict the mesh quality score. We validate the performance of our method with three publicly available databases, and the comparison with state-of-the-art metrics demonstrates the superiority of our method. Experimental results show that our proposed method achieves a high correlation between objective assessment and subjective scores.

Aplastic anemia and severe pancytopenia during treatment with peg-interferon,ribavirin and telaprevir for chronic hepatitis C

Telaprevir and Boceprevir are the first direct acting antivirals approved for chronic hepatitis C in combination with peg-interferon alfa and ribavirin.Pancytopenia due to myelotoxicity caused by these drugs may occur,but severe hematological abnormalities or aplastic anemia(AA) have not been described.We collected all cases of severe pancytopenia observed during triple therapy with telaprevir in four Spanish centers since approval of the drug in 2011.Among 142 cirrhotic patients receiving treatment,7 cases of severe pancytopenia(5%) were identified and three were consistent with the diagnosis of AA.Mean age was 59 years,five patients had compensated cirrhosis and two patients had severe hepatitis C recurrence after liver transplantation.Severe pancytopenia was diagnosed a median of 10 wk after the initiation of therapy.Three patients had pre-treatment hematological abnormalities related to splenomegaly.In six patients,antiviral treatment was interrupted at the onset of hematological abnormalities.Two patients died due to septic complications and one patient due to acute alveolar hemorrhage.The remaining patients recovered.Severe pancytopenia and especially AA,are not rare during triple therapy with telaprevir in patients with advanced liver disease.Close monitoring is imperative in this setting to promptly detect serious hematological disorders and to prevent further complications.

Unipolar depressive disorder(UDD)affects more than 264 million people worldwide and was projected well before the severe acute respiratory syndrome coronavirus 2 pandemic to be the leading cause of disability-adjusted life years lost in 2030.It is imperative for leading economies to implement preventive strategies targeted towards UDD,given consistent policies are currently lacking.Recently established similarities between the aetiological hypotheses of depression and cardiometabolic diseases are shifting paradigms within this field.It is believed that dietary practices could potentially reduce the incidence of depression;similar to their effects on metabolism.Thus,the aim of this review was to compile current evidence on healthy dietary patterns as suitable contributors towards primary prevention strategies against UDD.Most of the well-known biological mechanisms behind depression have been positively associated with healthful diets and dietary patterns to varying degrees.Interestingly,a common factor of UDD is the production and overall effects of inflammatory cytokines,such as interleukin-6,tumor necrosis factor-α,and Creactive protein.These compounds have been associated with depressive symptoms,disturbances in neuroendocrine function,leaky gut,monoamine activity and brain function,while also being key factors in the development of cardiometabolic diseases.The Mediterranean diet(MD)in particular,is well supported by first-level evidence regarding its preventive qualities against metabolic and cardiovascular diseases and thus considered a model for healthy eating by various organizations.In one of the few clinical trials investigating these associations,the PREDIMED trial,individuals with diabetes assigned to a MD supplemented with mixed tree nuts experienced a 41%relative risk reduction for developing depression.Lastly,there is a need to include health related quality of life as an indicator of physical and mental well-being,considering its putative associations with depression and suicide risk.Going forward,

Biofunctionalized all-polymer photonic lab on a chip with integrated solid-state light emitter

A photonic lab on a chip(PhLOC),comprising a solid-state light emitter(SSLE)aligned with a biofunctionalized optofluidic multiple internal reflection(MIR)system,is presented.The SSLE is obtained by filling a microfluidic structure with a phenyltrimethoxysilane(PhTMOS)aqueous sol solution containing a fluorophore organic dye.After curing,the resulting xerogel solid structure retains the emitting properties of the fluorophore,which is evenly distributed in the xerogel matrix.Photostability studies demonstrate that after a total dose(at λ5365 nm)greater than 24 J cm^(-2),the xerogel emission decay is only 4.1%.To re-direct the emitted light,the SSLE includes two sets of air mirrors that surround the xerogel.Emission mapping of the SSLE demonstrates that alignment variations of 150 mm(between the SSLE and the external pumping light source)provide fluctuations in emitted light smaller than 5%.After this verification,the SSLE is monolithically implemented with a MIR,forming the PhLOC.Its performance is assessed by measuring quinolone yellow,obtaining a limit of detection(LOD)of(0.6060.01)mM.Finally,the MIR is selectively biofunctionalized with horseradish peroxidase(HRP)for the detection of hydrogen peroxide(H_(2)O_(2))target analyte,obtaining a LOD of(0.760.1)μM for H_(2)O_(2),confirming,for the first time,that solid-state xerogel-based emitters can be massively implemented in biofunctionalized PhLOCs.

Use of Propolis for Topical Treatment of Dermatophytosis in Dog

Here we present three clinical cases of canine dermatophytosis resolved with topical propolis treatment that involved alopecia and well-demarcated erythematous lesions. These cases were positively identified by direct observation of samples from the affected zones with 10% KOH. Each sample was cultured, leading to the isolation of Microsporum gypseum in one case and Microsporum canis in the other two cases. The animals’ subsequent treatment included bathing using a commercial soap with propolis every seven days for 3 to 8 weeks, as well as the use of a propolis-containing ointment elaborated in our laboratory, which was applied to the lesions once a day for three weeks. From the second week of treatment, all cultures were negative. At the end of treatment, all cases displayed full recovery of the injuries and hair growth in these areas. In these clin-ical cases, treatment with propolis was effective, supporting the use of propolis as a promising natural alternative with no known collateral effects.

Kohler’s disease is an osteochondrosis of the tarsal navicular bone. The exact causes of osteochondrosis remain unknown. We report a 4-year-old girl with a haematogenous tarsal arthritis who developed a K?hler’s disease immediately after the infection. It is hypothesised that avascular necrosis of the navicular bone was developed secondary to pyogenic arthritis.

Optimum Power in a Multi-Span DWDM System Limited by Non-Linear Effects

Limitations imposed by Four-Wave Mixing (FWM), Amplified Spontaneous Emission (ASE), dispersion and Stimulated Raman Scattering (SRS) on a multi-spam DWDM system are theoretically studied. In this work, expression for the linear dispersion parameter D has been defined as a function of number of channels in order to separate FWM and SRS effects and calculates both maximum fibre length and optimum power. Additionally, our simulation results consider the effect of ASE noise from Erbium Doped Fibre Amplifiers (EDFAs). This theoretical analysis yields a set of design criteria from optimized multi-span DWDM systems.
