Mesenchymal Stem Cells Express Low Levels of Cardiomyogenic Genes and Show Limited Plasticity towards Cardiomyogenic Phenotype

Mesenchymal stem cell differentiation towards osteogenic, chondrogenic and adipogenic lineages have been extensively described and reproduced in the literature. In contrast, cardiomyogenic differentiation still remains largely controversial. In this study the authors aim to shed new light into this unclear phenomenon and test whether BMMSC （bone marrow mesenchymal stem cells） and ATMSC （adipose tissue derived mesenchymal stem cells） are able to differentiate into functional cardiomyocytes, investigating two differentiation protocols. AT and BMMSC behaved differently when cultured in differentiation media and presented lower levels of proliferation and alkaline phosphatase production, expression of cardiomyocyte-specific transcription factors such as GATA-4, Nkx2-5 and proteins such as ct and 13 Myosin Heavy Chains. Furthermore, MSC started to express higher levels of Connexin-43 and c~ sarcomeric actinin protein. Unfortunately, though, MSC did not present cardiomyocyte-like electrophysiological properties. In order to analyze a possible explanation for such limited plasticity, the authors decided to address the issue using a quantitative approach. Gene expression was quantified by Real time PCR, and, for the first time, the authors show that a possible explanation for limited plasticity of MSC is that even though differentiated cells presented differential gene expression, the levels of key cardiomyogenic genes did not reach expression levels presented by adult cardiomyocytes, nor were maintained along differentiation, reaching peaks at 4 days of stimulation, and decaying thereafter.

Immunization with rPb27 Protects Mice from the Disruption of VEGF Signaling in Paracoccidioides brasiliensis Infection

VEGF （Vascular endothelial growth factor） signaling is critical for endothelial cell survival and maintenance of the vasculature. deregulation of VEGF signaling contributes to the physiopathology of many diseases. However, the ways in which infection with Paracoccidioides brasiliensis affects VEGF signaling and the influence of immunization with rPb27 （recombinant protein Pb27） on VEGF signaling have not yet been studied. Animals were immunized with rPb27 and subsequently infected with a virulent strain of P. brasiliensis. The fungal load was evaluated by measuring CFU （colony-forming unit） and histology was performed to evaluate the inflammatory reaction. At the two time points analyzed, the PC （positive control） and TREAT （treated） animals had decreased levels of pulmonary VEGF compared to basal levels. However, in the immunized （Pb27） and treated mice （Pb27 ＋ TREAT）, VEGF expression remained unchanged after infection. In the case of VEGFR-2, the Pb27 and Pb27 ＋ TREAT groups showed increased levels of expression. Regarding the levels of the eNOS enzyme, only the Pb27 group did not reduce the expression levels relative to baseline. The immunization with rPb27 kept VEGF signaling, NO production and increased VEGFR-2 expression, after infection with P. brasiliensis. Thus, the authors infer that immunization with rPb27 protects mice from the disruption of VEGF signaling in Paracoccidioides brasiliensis infection.

Risk Models for the Prize Collecting Steiner Tree Problems with Interval Data

Cytomegalovirus is the most frequent agent of congenital viral infections,affecting approximately 0.2%-2.4%of live births in different countries[1-7].Universal screening for congenital cytomegalovirus infection in newborns is not a standard practice considering cost-benefit issues and viabil-ity of diagnostic ***,most newborns are asympto-matic and there is no effective treatment for this population[2-5].

Size Distribution of Particulate Matter of Metals in Urban Aerosols, Comarca Lagunera, Mexico

This paper has demonstrated that there is great variation in the size range and chemical composition of metalliferous PM （particulate matter） present within Comarca Lagunera, M6xico due to the physiography of the terrain together the intense vehicular traffic, unpaved roads, cement and lime plants, marble quarries, brick plants, and the largest smelter of non-ferrous material in Latin America. Cascade impactor PM samples from six size ranges：〈 0.49 μm, 0.49-0.9 μm, 0.9-1.5μm, 1.5-3.0 p.m, 3.0-7.2 μm and 7.2-10 μm were collected from inside two stacks within G6mez Palacios and Torreon cities. ICP-AES （inductively coupled plasma-atomic emission spectrometry） analysis on collected Whatman fiberglass filters demonstrates that the PM are matrices mostly constituted of carbonates and silicates with abundant fine metalliferous particles. The metals with the most concentrations averaged over all size ranges were Cu, Fe, Mn, Ni, Pb and Ti. The size distributions of metals detected in this work have been plotted as normalized histogram concentration of each size fraction, the total concentration, and the aerodynamic particle diameter, which is a useful method for comparing the contributions of coarse and fine particles to pollutant concentrations.

Production Optimization of Biodiesel from Frying Oil Waste to Reduce the Environmental Impacts

The objective of this study was to reduce the environmental impacts of used frying oil waste through the production of biodiesel. A 22 factorial planning has been used to evaluate the influences of alcohol/oil and reaction time on the biodiesel production yield. The optimal condition to produce the biodiesel has been found by use of the response surface methodology and analysis of variance to obtain the fitting model. This study was conducted in Campinas city, Brazil, where were collected the waste oil. An analysis of ecological cost also has been developed. Cooking oils collected from Campinas homes were mixed with ethanol in planned proportions （1：9, 1：7 and 1：5） and were transesterified at 60 ℃ and planned reaction times （30, 60 or 90 min）, in order to obtain biodiesel, using 0.1% NaOH as a catalyst. The results of the physical-chemical analyses demonstrated that the biodiesels obtained possessed characteristics close to those required by Brazilian standards. This fuel could be used in fleets of buses, trucks and machines, or even sold to fuel distributors, which results in a solving between US$0.8 and US$4.5 millions. Thus, Campinas would gain environmental credits and become a sustainable city.

Application of Automatic Mass Balance in Metal Accounting of Iron Ore Plant

The purpose of this work is to present state of the art mass balances closing strategies in ore treatment plants to report metal accounting in a reliable manner. The current methodology is based on usage of an expert system which seizes and filters the plant data, analyses situations of equipment stops and mostly instruments failures. It is still possible to internally create a strategy to check the data of an instrument failure. The system is coupled to software with a Lagrangian model of mass balance closure that receives the pre-treated data. Thus, the data goes through filtering and reconciling processes to generate a reliable mass balance of the ore treatment plant. This made it possible to obtain a system that is able to tell the real productivity of an ore plant through reliable mass balances. Residence times on equipment are critical to the balance, thus, they require good logic to eliminate errors due to the characteristics of non-steady state. Among the next challenges is the inclusion of a soft sensor as a replacement of an instrument required for mass balance closure.

Clostridium difficile is a nosocomial enteric pathogen, considered the main etiological agent of antibiotic-associated diarrhea in hospitals. C. difficile is resistant to several antimicrobial agents used in empiric treatment protocols, which confers selective advantages to this species as compared with other members of the intestinal microbiota. The aim of this study was to evaluate and identify possible alterations of the intestinal microbiota population resulting from the use of antimicrobials associated with infections by C. difficile (CDI). denaturing Gradient Gel Electrophoresis (DGGE) was used to evaluate fecal samples from two groups: healthy subjects and patients with C. difficile-associated diarrhea (CDAD). A decrease in bacterial diversity was highlighted by the low number of bands in samples from CDAD patients, compared with healthy subjects. This may indicate that antibiotic treatment would affect bacterial diversity, leading to a significant difference between the intestinal microbiota of these two groups, but further studies are still needed. Firmicutes and Verrucomicrobia phyla were detected mainly in healthy individuals, and these could be related to protection factors against the CDI. Klebsiella variicola/K. pneumoniae were found mostly in samples from CDI patients. This study shows the effects of antimicrobials and the CDI itself on human intestinal microbiota.

Coral reefs have largely declined across multiple spatial scales due to a combination of local-scale anthropogenic impacts, and due to regional-global climate change. This has resulted in a significant loss of entire coral functional groups, including western Atlantic Staghorn coral (Acropora cervicornis) biotopes, and in a net decline of coral reef ecosystem resilience, ecological functions, services and benefits. Low-tech coral farming has become one of the most important tools to help restore depleted coral reefs across the Wider Caribbean Region. We tested a community-based, low-tech coral farming approach in Culebra Island, Puerto Rico, aimed at adapting to climate change-related impacts through a two-year project to propagate A. cervicornis under two contrasting fishing management conditions, in coastal areas experimenting significant land use changes. Extreme rainfall events and recurrent tropical storms and hurricanes had major site-and method-specific impacts on project outcome, particularly in areas adjacent to deforested lands and subjected to recurrent impacts from land-based source pollution (LBSP) and runoff. Overall, coral survival rate in “A frame” units improved from 73% during 2011-2012 to 81% during 2012-2013. Coral survival rate improved to 97% in horizontal line nurseries (HLN) incorporated during 2012-2013. Percent tissue cover ranged from 86% to 91% in “A frames”, but reached 98% in HLN. Mean coral skeletal extension was 27 cm/y in “A frames” and 40 cm/y in HLN. These growth rates were up to 545% to 857% faster than previous reports from coral farms from other parts of the Caribbean, and up to 438% faster than wild colonies. Branch production and branchiness index (no. harvestable branches > 6 cm) increased by several orders of magnitude in comparison to the original colonies at the beginning of the project.
