Quantification of corneal neovascularization after ex vivo limbal epithelial stem cell therapy

AIM: To assess cultured limbal epithelial stem cell transplantation in patients with limbal stem cell deficiency by analyzing and quantifying corneal ***: This retrospective, interventional case series included eight eyes with total limbal stem cell deficiency. Ex vivo limbal epithelial stem cells were cultured on human amniotic membrane using an animalfree culture method. The clinical parameters of limbal stem cell deficiency, impression cytology, and quantification of corneal neovascularization were evaluated before and after cultured limbal stem cell transplantation. The area of corneal neovascularization,vessel caliber(VC), and invasive area(IA) were analyzed before and after stem cell transplantation by image analysis software. Best-corrected visual acuity(BCVA),epithelial transparency, and impression cytology were also ***: One year after surgery, successful cases showed a reduction(improvement) of all three parameters of corneal neovascularization [neovascular area(NA), VC, IA], while failed cases did not. NA decreased a mean of 32.31%(P =0.035), invasion area29.37%(P =0.018) and VC 14.29%(P =0.072). BCVA improved in all eyes(mean follow-up, 76 ±21mo).Epithelial transparency improved significantly from 2.00 ±0.93 to 0.88±1.25(P =0.014). Impression cytology showed that three cases failed after limbal epithelial stem cell therapy before 1y of ***: This method of analyzing andmonitoring surface vessels is useful for evaluating the epithelial status during follow-up, as successful cases showed a bigger reduction in corneal neovascularization parameters than failed cases. Using this method,successful cases could be differentiated from failed cases.

AIM:To compare the mydriatic efficacy of an ophthalmic insert(Mydriasert,MY)versus phenylephrine and tropicamide(PT)eye ***:Two controlled,prospective,randomized,single-blind studies were *** the first study,a total of 80 eyes from 40 outpatient-clinic patients were *** drops were applied to the right eye,and a MY device was inserted in the left eye for *** until maximal pupil dilation for each eye was then *** the second study,80 eyes from 80 patients undergoing cataract surgery were *** dilation was achieved using either PT drops three-times for one hour prior to surgery(40 patients),or a MY device was inserted one hour prior to surgery(40 patients).RESUlTS:In the first study,MY achieved superior mydriasis compared to PT eye drops at 90min(9.04±1.33mm vs 8.78±1.37mm,P=0.012).However MY took longer than PT drops to achieve maximal dilation,and mydriasis was inferior in eyes with MY compared to PT drops at 30min(7.21±1.73mm vs 8.22±1.43mm,P<0.001),the two groups only becoming similar by 60min(8.85±1.44mm vs 8.71±1.27mm,P=0.236).In the second study,both MY and PT achieved similar levels of mydriasis at the beginning of surgery(8.75±0.76mm with MY vs 8.77±0.63mm with PT),and also at the end of surgery(7.96±1.06mm with MY vs 8.32±0.72mm with PT),with no significant difference between groups(P=0.08).MY was well tolerated and cardiovascular effects were not influenced by dilation ***:MY could be a safe and efficacious alternative for *** mydriatic effect of MY is as good as conventional PT eye drops after 60min,and is superior after *** also maintains good pupil dilation during cataract surgery.

Analysis of heterogeneous characteristics in a geothermal area with low permeability and high temperature

An analytical methodology for reservoir characterization was applied in the central and southwestern zones of los Humeros geothermal field(lHGF). This study involves analysis of temperature, pressure,enthalpy and permeability in wells and their distribution along the area. The wells located in the central western side of the geothermal field are productive, whereas those located at the central-eastern side are non-productive. Through temperature profiles, determined at steady state in the analyzed wells, it was observed that at bottom conditions(approximately 2300 m depth), temperatures vary between 280 and360℃. The temperatures are higher at the eastern side of central zone of lHGF. A review of transient pressure tests, laboratory measurements of core samples, and correlation of circulation losses during drilling suggest that permeability of the formation is low. The enthalpy behavior in productive wells shows a tendency of increase in the steam fraction. It was found that productivity behavior has inverse relation with permeability of rock formation. Further, it is observed that an imbalance exists between exploitation and recharge. It is concluded from the results that the wells located at central-eastern area have low permeability and high temperature, which indicates possibility of heat storage.

Affective responses to supervised 10-week programs of resistance exercise in older adults

Background:Resistance exercise has numerous,well-documented benefits on the health and functional performance of older ***,little information exists on the affective responses to resistance exercise in this *** affective responses can predict continued exercise behavior,examining if and how they differ between resistance exercise intensities and frequencies in older adults may provide important data to improve resistance exercise ***:We monitored the affective responses of older adults when performing high-velocity,low-load(HVll)or low-velocity,high-load(lVHl)resistance exercise once or twice weekly.A total of 40 moderately to highly active,but resistance-exercise naïve,older adults(60-79 years)completed 1 of 4 randomly assigned progressive resistance exercise conditions:HVll once weekly(HVll1:n=10),HVll twice weekly(HVll2:n=10),lVHl once weekly(lVHl1:n=10),or lVHl twice weekly(lVHl2:n=10).The Physical Activity Affect Scale,Felt Arousal Scale,Feeling Scale,rating of perceived exertion,Visual Analog Scale,and the Physical Activity Enjoyment Scale were used to assess enjoyment and affective ***:The results from Physical Activity Enjoyment Scale(out of 35)suggest that all exercise conditions were largely and similarly enjoyable to older adults(HVll1:30.9±2.4;lVHl1:31.9±3.6;HVll2:30.9±3.5;lVHl2:30.2±3.7)despite greater fatigue(p=0.033;ηp^2p=0.22)and perceived workload(p=0.042;ηp^2=0.20)in lVHl(results from Visual Analog Scale).Conclusion:Moderately to highly physically active older adults may tolerate higher intensities of resistance exercise performed once or twice weekly without experiencing a negative impact on ***,the role that supervision and social interaction played in shaping the participants’responses is unclear.

The ionic liquid(Il) tributylmethylammonium bis(trifluoromethylsulfonyl)amide([N4441][NTf2]) was used as neat lubricant and as an additive(1.5 wt%) in a polar oil to study its friction and wear reducing *** tests were completed for 90 minutes at room temperature and 100 °C in a reciprocating configuration at loads of 30 and 70 N,10 Hz-frequency,and 4 mm stroke *** volume was measured by confocal microscopy and the surface-Il interaction determined by *** main findings were that neat Il showed the best tribological behavior;the Il-containing mixture behaved similar to the base oil regarding friction,however outperformed the antiwear behavior of the base oil under higher temperature;surface-Il chemical interaction was found mainly at 100 °C.

Maternal reproductive health: Expression patterns of antioxidant enzyme selenoproteins of post-implantation embryos conceived by ethanol-treated murine mothers supplemented with α-tocopherol

Objective:To investigate if the protective effect ofα-tocopherol against the impact of ethanol on brain morphogenesis involved the activity of the selenoproteins phospholipid hydroperoxide glutathione peroxidase (PHGPx;GPx4) and selenoprotein P (SelPP) that have roles against oxidative ***:Forty female mice were randomly assigned into natural control (CON), positive control (ETOH), low-, medium-, and high-α-tocopherol-supplemented-ethanol groups (lTOC, MTOC, HTOC, respectively). CON received drinking water without ethanol while ETOH, lTOC, MTOC and HTOC groups received 20% ethanol in drinking water. The supplemented groups were given respective dosages ofα-tocopherol, 0.410, 0.819, and 1.640 mg/g body weight, at day 14 before mating onwards to the day 9 of gestation. At 10.5 ED of gestation (1100 h), the pregnant females were sacrificed by cervical dislocation and the embryos were harvested. Total RNA were extracted, cDNA synthesis and qRT- PCR analyses were carried ***: The level of expression of PHGPx in the positive control was significantly lower than that of the natural control. Among the threeα-tocopherol-supplemented groups, only the medium dose- group was significantly higher than the positive control. The level of expression of SelPP in the positive control was significantly lower than those of the natural control, the low- and medium- dose -tocopherol supplemented groups. In the high dose-α-tocopherol supplemented group, the level of expression was not significantly different from the positive control but significantly lower than the natural ***: The activity of the selenoproteins PHGPx and SelPP are involved in the internetwork of antioxidative enzymes with vitamin E when given up to a medium dose only and is one of the possible pathways of shielding embryonic development against the impact of ethanol on brain morphogenesis. Non-redundant Contribution of the Plastidial FAD8 ω-3 Desatumse to Glycerolipid Unsatumtion at Different Temperatures in Arabidopsis

Plastidial ω-3 desaturase FAD7 is a major contributor to trienoic fatty acid biosynthesis in the leaves of Arabidopsis plants. However, the precise contribution of the other plastidial ω-3 desaturase, FAD8, is poorly understood. Fatty acid and lipid analysis of several ω-3 desaturase mutants, including two insertion lines of AtFAD7 and AtFAD8, showed that FAD8 partially compensated the disruption of the AtFAD7 gene at 22℃, indicating that FAD8 was active at this growth temperature, contrasting to previous observations that circumscribed the FAD8 activity at low temperatures. Our data revealed that FAD8 had a higher selectivity for 18：2 acyl-lipid substrates and a higher preference for lipids other than galactolipids, particularly phosphatidylglycerol, at any of the temperatures studied. Differences in the mechanism controlling AtFAD7 and AtFAD8 gene expression at different temperatures were also detected. Confocal microscopy and biochemical analysis of FAD8-YFP over-expressing lines confirmed the chloroplast envelope localization of FAD8. Co-localization experiments suggested that FAD8 and FAD7 might be located in close vicinity in the envelope membrane. FAD8-YFP over-expressing lines showed a specific increase in 18：3 fatty acids at 22℃. Together, these results indicate that the function of both plastidial ω-3 desaturases is coordinated in a non-redundant manner.
