The global expansion of cultivation areas of Jatropha has contributed to the emergence of various diseases.Currently in Brazil,the occurrence of a new disease has been reported that not only reduces the productivity but also causes the death of Jatropha.This disease is associated with collar and root rot of plants.From morphological and phylogenetic studies(based on Internal Transcribed Spacers,β-tubulin and Translation Elongation Factor 1-αsequences),nine species of Botryosphaeriaceae were identified.These species include Lasiodiplodia egyptiacae,L.pseudotheobromae,L.theobromae,Macrophomina phaseolina,Neoscytalidium hyalinum and four Lasiodiplodia spp.that are proposed as new species(L.euphorbicola,L.jatrophicola,L.macrospora and L.subglobosa).All the species in this study,except M.phaseolina,are pathogenic.The results show that root rot of physic nut plants is caused by complex pathogens.This study provides new information for future studies of disease management,quarantine programs and,especially,the development of resistant varieties for collar and root rot disease in J.curcas.

Liver transplantation as an alternative for the treatment of perihilar cholangiocarcinoma: A critical review

Background:Perihilar cholangiocarcinoma(phCCC)is a dismal malignancy.There is no consensus regard-ing the best treatment for patients with unresectable phCCC.The present review aimed to gather the current pieces of evidence for liver transplantation and liver resection as a treatment for phCCC and to build better guidance for clinical practice.Data sources:The search was conducted in PubMed,Embase,Cochrane,and LILACS.The related references were searched manually.Inclusion criteria were:reports in English or Portuguese literature that a)patients with confirmed diagnosis of phCCC;b)patients treated with a curative intent;c)patients with the outcomes of liver resection and liver transplantation.Case reports,reviews,letters,editorials,conference abstracts and papers with full-text unavailability were excluded from the analysis.Results:Most of the current literature is based on observational retrospective studies with low grades of evidence.Liver resection has better long-term outcomes than systemic chemotherapy or palliation ther-apy and liver transplantation is a good alternative for selected patients with unresectable phCCC.All candidates for resection or transplantation should be medically fit and free of intrahepatic or extrahep-atic diseases.As a general rule,patients presenting with a tumor having a longitudinal size>3 cm or extending below the cystic duct,lymph node disease,confirmed extrahepatic dissemination;intraoper-atively diagnosed metastatic disease;a history of other malignancies within the last five years,and did not complete chemoradiation regimen and were medically unfit should not be considered for transplan-tation.Some of these criteria should be individually assessed.Liver transplantation or resection should only be considered in highly experienced hepatobiliary centers,and any decision-making must be based on a multidisciplinary evaluation.Conclusions:phCCC is a complex condition with high morbidity.Surgical therapies,including hepatec-tomy and liver transplantation,are the best option for bette

BACKGROUND Hepatocellular carcinoma(HCC)is an aggressive malignant neoplasm that requires liver transplantation(LT).Despite patients with HCC being prioritized by most organ allocation systems worldwide,they still have to wait for long periods.Locoregional therapies(LRTs)are employed as bridging therapies in patients with HCC awaiting LT.Although largely used in the past,transarterial embolization(TAE)has been replaced by transarterial chemoembolization(TACE).However,the superiority of TACE over TAE has not been consistently shown in the literature.AIM To compare the outcomes of TACE and TAE in patients with HCC awaiting LT.METHODS All consecutive patients with HCC awaiting LT between 2011 and 2020 at a single center were included.All patients underwent LRT with either TACE or TAE.Some patients also underwent percutaneous ethanol injection(PEI),concom-itantly or in different treatment sessions.The choice of LRT for each HCC nodule was determined by a multidisciplinary consensus.The primary outcome was waitlist dropout due to tumor progression,and the secondary outcome was the occurrence of adverse events.In the subset of patients who underwent LT,complete pathological response and post-transplant recurrence-free survival were also assessed.RESULTS Twelve(18.5%)patients in the TACE group(only TACE and TACE+PEI;n=65)and 3(7.9%)patients in the TAE group(only TAE and TAE+PEI;n=38)dropped out of the waitlist due to tumor progression(P log-rank test=0.29).Adverse events occurred in 8(12.3%)and 2(5.3%)patients in the TACE and TAE groups,respectively(P=0.316).Forty-eight(73.8%)of the 65 patients in the TACE group and 29(76.3%)of the 38 patients in the TAE group underwent LT(P=0.818).Among these patients,complete pathological response was detected in 7(14.6%)and 9(31%)patients in the TACE and TAE groups,respectively(P=0.145).Post-LT,HCC recurred in 9(18.8%)and 4(13.8%)patients in the TACE and TAE groups,respectively(P=0.756).Posttransplant recurrence-free survival was similar between the groups(P log-rank test=0.71).CONCL

The cryptic lifestyle of most fungi necessitates molecular identification of the guild in environmental studies.Over the past decades,rapid development and affordability of molecular tools have tremendously improved insights of the fungal diversity in all ecosystems and habitats.Yet,in spite of the progress of molecular methods,knowledge about functional properties of the fungal taxa is vague and interpretation of environmental studies in an ecologically meaningful manner remains challenging.In order to facilitate functional assignments and ecological interpretation of environmental studies we introduce a user friendly traits and character database FungalTraits operating at genus and species hypothesis levels.Combining the information from previous efforts such as FUNGuild and FunFun together with involvement of expert knowledge,we reannotated 10,210 and 151 fungal and Stramenopila genera,respectively.This resulted in a stand-alone spreadsheet dataset covering 17 lifestyle related traits of fungal and Stramenopila genera,designed for rapid functional assignments of environmental stud-ies.In order to assign the trait states to fungal species hypotheses,the scientific community of experts manually categorised and assigned available trait information to 697,413 fungal ITS sequences.On the basis of those sequences we were able to summarise trait and host information into 92,623 fungal species hypotheses at 1%dissimilarity threshold.

Genetic diversity and structure of Drimys brasiliensis in southern Brazil:insights for conservation

Population genetics studies are widely recognized for generating useful knowledge for biodiversity conservation.To date,however,little is known about the levels and distribution of genetic diversity of Drymis brasiliensis(Miers LC),a tree species from the Atlantic Rainforest.Therefore,in this study,we investigated how genetic diversity is distributed within and among populations of D.brasiliensis from southern Brazil using allozyme markers to genotype reproductive trees(8 populations)and seedlings(3 populations).Furthermore,in two populations,we established two permanent plots(5.1 and 1 ha)to analyze fine-scale genetic structure(FSGS).Studied populations presented low levels of genetic diversity(reproductive=0.085;seedlings=0.054)and high fixation indexes(reproductive=0.396;seedlings=0.231).Genetic divergence among populations was equal to 0.05,which is significant,signaling that few populations can conserve large portions of the species total genetic diversity.FSGS was only detected for one population,when reproductive individuals were separated by less than40 m.Low genetic diversity combined with high fixation indexes clearly signal a risk of losing diversity.Therefore,conservation efforts should be aimed at enhancing gene flow within the studied populations.

Ureteral stents play a fundamental role in modern time urology. However, following the deployment, stent-related symptoms are frequent and affect patient health and quality of life. Using biodegradable metals as ureteral stent materials have emerged as a promising strategy, mainly due to the improved radial force and slower degradation rate expected. Therefore, this study aimed to characterize different biodegradable metals in urinary tract environment to understand their propensity for future utilization as base materials for ureteral stents. The corrosion of 5 Mg alloys - AZ31, Mg-1Zn, Mg-1Y, pure Mg, and Mg-4Ag - under simulated urinary tract conditions was accessed. The corrosion layer of the different alloys presented common elements, such as Mg(OH)_(2), MgO, and phosphate-containing products, but slight variations in their chemical compositions were detected. The corrosion rate of the different metals varied, which was expected given the differences in the corrosion layers. On top of this, the findings of this study highlighted the significant differences in the samples' corrosion and corrosion layers when in stagnant and flowing conditions. With the results of this study, we concluded that Mg-1Zn and Mg-4Ag presented a higher propensity for localized corrosion, probably due to a less protective corrosion layer;Mg-4Ag corroded faster than all the other four alloys,and Mg-1Y stood out due to its distinct corrosion pattern, that showed to be more homogeneous than all the other four samples, making this one more attractive for the future studies on biodegradable metals.

Esophagogastric anastomosis with invagination into stomach:New technique to reduce fistula formation

AIM: To present a new technique of cervical esophagogastric anastomosis to reduce the frequency of fistula formation. METHODS: A group of 31 patients with thoracic and abdominal esophageal cancer underwent cervical esophagogastric anastomosis with invagination of the proximal esophageal stump into the stomach tube. In the region elected for anastomosis, a transverse myotomy of the esophagus was carried out around the entire circumference of the esophagus. Afterwards, a 4-cm long segment of esophagus was invaginated into the stomach and anastomosed to the anterior and the posterior walls. RESULTS: Postoperative minor complications occurred in 22 (70.9%) patients. Four (12.9%) patients had serious complications that led to death. The discharge of saliva was at a lower region, while attempting to leave the anastomosis site out of the alimentary transit. Three (9.7%) patients had fistula at the esophagogastric anastomosis, with minimal leakage of air or saliva and with mild clinical repercussions. No patients had esophago- gastric fistula with intense saliva leakage from either the cervical incision or the thoracic drain. Fibrotic stenosis of anastomoses occurred in seven (22.6%) patients. All these patients obtained relief from their dysphagia with endoscopic dilatation of the anastomosis.CONCLUSION: Cervical esophagogastric anastomosis with invagination of the proximal esophageal stump into the stomach tube presented a low rate of esophagogastric fistula with mild clinical repercussions.

The trade creation and conservation of wild species in Amazon, including collared peccaries (Pecari tajacu), may be favored by artificial insemination, but the time of semen storage may lead to reduced fertility in sperm of some wild animals. Thus, the aim of this research was to evaluate the influence of semen storage at 17 °C over time on seminal features of peccaries in captivity. Eight adult males were sedated and underwent electroejaculation. The ejaculates (n = 65) were evaluated for volume, concentration, pH, sperm motility, vigor, and cell with intact plasma membrane intact (IPM) and sperm morphology. Selected ejaculates (n = 21) were diluted (1:1) in Beltsville Thawing Solution and kept during 48 hours under controlled temperature (17 °C). Assessments were made after dilution (T0), after 24 hours (T24), and 48 hours after the onset of cooling (T48). The storage impacted on sperm survival (P < 0.05). Semen characteristics changed throughout the storage period studied and after 48 hours storage. The decline of sperm motility was of 55.2% for 10.9%, vigor was 2.3 for 0.5 and IPM cells were of 59.0% for 15.8%. Primary defects sperm increased of 19.8% for 32.2%, secondary defects of 9.8% for 10.4% and total defects of 29.4% for 42.7%. However, within 24 hours of preservation, chilled semen peccaries presented sperm motility average rate and IMP cells levels indicative to use in assisted reproductive events. These results indicate chilled semen for 24 hours could be used in experimentally artificial insemination of peccaries, technology that still has not been performed before.

Residual Effects of the Organic Amendments Poultry Litter,Farmyard Manure and Biochar on Soybean Crop

The use of organic wastes, as an alternative to inorganic fertilizer, can be an important strategy for Brazilian and tropical agriculture. Despite the importance, few field studies have been done for evaluating organic amendments on soybean crops in Brazil. The study aimed to evaluate the residual effects of the organic amendments poultry litter, farmyard manure and biochar combined with mineral fertilizer on some agronomic attributes of a soybean crop. A field experiment was carried out in a split-split-plot scheme, with three replicates in a randomized block experimental design. The organic sources (plots) at rates of 0, 3, 6 and 9 Mg·ha-1 (subplots) combined with 0, 100, 200, 300 and 400 kg·ha-1 (sub subplots) of a mineral fertilizer were applied in 2008. In 2009, only the mineral fertilizer was used on the soybean crop. As a?result, all evaluated attributes were influenced by the treatments, except for the number of grains per pod. The application of poultry litter provided the highest yield (3715 kg·ha-1 using 9 Mg·ha-1 of the source). A synergistic effect between organic amendments and mineral fertilizer was observed. It was found the possibility of decrease doses of mineral fertilizers by prior use of organic amendments. The most effective dose combination application is 5.5 Mg·ha-1 of organic amendments associated with 200 kg·ha-1 of mineral fertilizer to provide optimum yield. The use of organic amendments, rich in nutrients, is a technology to sustainably increase the soybean grain yield.

Various mouse models to study dengue have been described by different authors, some of them using immunodeficient or some using humanized mice. Our group reported previously a deadly murine model, which used the intracranial inoculum of highly virulent Dengue virus (DENV) in immune competent mouse. Here we present a model of immune competent mouse (C57BL/6), infected subcutaneously by the same highly virulent DENV (DENV3 genotype I). In this immunocompetent systemic mice model, the cytokine levels and hematological parameters such as total and differential leukocyte and platelets counts, together with weight loss, were considered important monitoring parameters, allowing a better understanding of the systemic human disease. Mice were inoculated subcutaneously and evaluated by the percentage weight variation as well as the clinical signs. Hematological parameters and cytokines levels were measured and viral titration in brain tissue or serum neutralization was performed to confirm mice infection. The subcutaneously DENV inoculated mice showed weight loss after infection, but they did not show any other clinical signs. The leukocytes and platelets decreased after subcutaneous inoculation. The cytokines TNF alpha and IFN gamma increased after infection in mice. The subcutaneous model provided scope for improved understanding of the dengue pathogenesis, as well as possible mechanism for protection to subsequent mouse infected by intracranial route in mice. This model could be used to study the vertebrate immune response and evaluation of drugs or vaccine against dengue virus.
