Optimization Techniques and Development of Neural Models Applied in Biosurfactant Production by <i>Bacillus subtilis</i>Using Alternative Substrates

Bacillus subtilis was investigated as production of biosurfactant using a combination based on waste of candy industry and glycerol from biodiesel production process as only substrate. The experimental design chosen for optimization by response surface methodology was a central composite rotatable design (CCRD) and dry weight (DW) and crude biosurfactant (CB) concentrations were selected as responses in analysis. Two techniques were implemented response surface methodology (RSM) and artificial neural network (ANN). First challenge of study was to assess the effects of the interactions between variables and reach optimum values. With the CCRD results, RSM and ANN models were developed, optimizing the production of biosurfactant. The correlation coefficients (R2) of RSM models explained 88% for DW and 73% for CB of the interactions among substrate concentrations, while ANN models explained 99% for DW and 98% for CB, demonstrating that developed ANN models were more accurate and consistent in predicting optimized conditions than RSM model. The maximum DW and CB produced in the optimum conditions were 25.60 ± 5.0 g/L and 668 ± 40 mg/L, respectively. The crude biosurfactant also showed applications in cases of oil spreading in water due to clear zone produced in Petri dishes assays.

Structural and optical properties of pillared Eu^(3+)-containing layered double hydroxides intercalated by 2-to 12-carbon aliphatic dicarboxylates

While the versatile composition of layered double hydroxides(LDHs)allows the introduction of almost any polyvalent cation in their structure,the exchangeable negatively charged ions intercalated between the plates increase their vast range of *** we report on the preparation and optical properties of pillared Eu^(3+)-substituted ZnAl LDHs intercalated by aliphatic dicarboxylates:-OOC-(CH_(2))_(n-2)-COO-.The basal distance in these materials is dependent on the size and packing of the intercalated *** varying the number(n)of carbon atoms in the aliphatic chain from 2 to 12,the interlayer gallery of these ZnAlEu LDHs is considerably expanded from 0.9 to 2.1 *** the interlayer gallery,the aliphatic dicarboxylates form a monolayer,with the aliphatic chain inclined by an angleα≈63°with the hydroxide *** carbon atom in the COO-group is 0.43 nm far from the metal plane,showing that these carboxylates are not grafted in the brucite-like *** LDHs are thermally stable up to around 150℃,after which dehydroxylation of the hydroxide layers is *** Judd-Ofelt intensity parameters were obtained and compared to the Eu^(3+)-containing complexes with the same ligand series indicating a more symmetrical and less polarizable chemical environment around the rare earth ion.

Rescuing agroforestry as strategy for agriculture in Southern Brazil

Knowledge about agricultural ecosystems, as well as about ecological and social interactions that occur in an agricultural context, is essential to promote a balanced environment for production. Incorporating the forest landscape into farming systems has promoted the development of integrated production systems such as agroforestry. However, many aspects of the relationship between farming and the forest landscape have changed since the modernization of agriculture introduced by the Green Revolution. The objectives of this review are：（1） to contribute to the understanding of the changes in cultivation strategies and in the dynamics of agricultural systems,particularly in the context of farming in Santa Catarina State, Brazil;（2） to contribute to the understanding of agroforestry systems as a strategy to reintroduce the element of forestry into agricultural production systems; and（3） to compile a list of forest plant species that could be used in agroforestry. The approach adopted in this manuscript seeks to highlight the potential of agroforestry systems to conserve natural resources and thereby to support agricultural development by connecting environmental conservation with income for farmers.

Combined Selection in Backcross Population of Papaya(Carica papaya L.)by the Mixed Model Methodology

The selection of superior genotypes based on the simultaneous response to several characteristics of agronomic importance is a key strategy to overcome the scarcity of available varieties of papaya. This study aimed to apply the combined selection by using distinct selection indexes based on both the genetic values obtained by the REML/BLUP methodology and the real measured values to select agronomically superior genotypes of papaya within backcross progenies. The combined selection was carried out based on genetic and phenotypic values, original and standardized, multiplied by the agronomic weights. The results of the analysis of genetic parameters indicate that the evaluated progenies have expressive genetic variability for the considered traits, and that there are real possibilities of genetic progress with the selection. Among the analyzed indexes, the one based on standardized genetic value presented greater consistency in the ranking of genetic material, demonstrating the advantage of data standardization. Five progenies belonging to the BC1 generation, and five to the BC3 generation were selected using this index. A total of 27 plants ag-ronomically superior were selected within the top five progenies and recommended for generation advance, 23 being selected by combined selection and 4 using the direct selection for the four mainly characters in papaya breeding program: production, pulp and fruit firmness and soluble solids. Beyond the selection of superior genotypes for the development of future inbred lines, this study also allowed defining the best strategy to apply the combined selection in papaya using pre-dicted breeding values obtained by BLUP. This strategy may allow higher accuracy in the selection process, thus increasing the chances of success of the breeding programs.

Assessment of Acute Episodes in Chronic Stable Angina: A Clinical-Laboratory Approach to the Use of the Coronary Vasodilator Propatyl Nitrate

Background/Objectives: Propatyl nitrate is a coronary vasodilator with immediate and prolonged action, indicated in the treatment and prevention of acute angina pectoris episodes. Methods: This was an open, self-paired comparative study performed at UNIFESO Medical School evaluating the clinical and laboratory results of treatment with propatyl nitrate in patients with chronic stable angina pectoris. Subjects received 10 mg of propatyl nitrate, at the dose of three sublingual tablets per day, to be taken at 8:00 A.M., 2:00 P.M., and 8:00 P.M. Subjects returned to the study center after 15 days of treatment for Visit 2 assessments, and at the end of the 30-day treatment period (Visit 3). Results: A total of 200 subjects were included in the study. There was a statistically significant reduction in blood pressure (p < 0.0001) and heart rate (p = 0.0001), but no change in respiratory rate (p = 0.23). Laboratory results did not vary throughout the treatment period. There was no significant change from pretreatment in the SAQ Physical Limitation scale (p = 0.7415). The Angina Stability, Angina Frequency, and Treatment Satisfaction, and Quality of Life scales showed a significant improvement from pretreatment (p < 0.0001). Adverse events were observed among 41 subjects at Visit 2 and 35 subjects at Visit 3. Conclusions: Propatyl nitrate was safe and effective in treating chronic stable angina pectoris over the course of the 30-day treatment period. Treatment with propatyl nitrate increased angina stability and reduced angina frequency while increasing treatment satisfaction and quality of life in the patient population evaluated.

B-1 cells modulate the murine macrophage response to Leishmania major infection

AIM To investigate the modulatory effect of B-1 cells on murine peritoneal macrophages infected with Leishmania major(L. major) in *** Peritoneal macrophages obtained from BALB/c andBALB/c XID mice were infected with L. major and cultured in the presence or absence of B-1 cells obtained from wild-type BALB/c mice. Intracellular amastigotes were counted, and interleukin-10(IL-10) production was quantified in the cellular supernatants using an enzymelinked immunosorbent assay. The levels of the lipid mediator prostaglandin E2(PGE2) were determined using a PGE2 enzyme immunoassay kit(Cayman Chemical, Ann Arbor, MI), and the number of lipid bodies was quantified in the cytoplasm of infected macrophages in the presence and absence of B-1 cells. Culturing the cells with selective PGE2-neutralizing drugs inhibited PGE2 production and confirmed the role of this lipid mediator in IL-10 production. In contrast, we demonstrated that B-1 cells derived from IL-10 KO mice did not favor the intracellular growth of L. *** We report that B-1 cells promote the growth of L. major amastigotes inside peritoneal murine macrophages. We demonstrated that the modulatory effect was independent of physical contact between the cells, suggesting that soluble factor(s) were released into the cultures. We demonstrated in our co-culture system that B-1 cells trigger IL-10 production by L. major-infected macrophages. Furthermore, the increased secretion of IL-10 was attributed to the presence of the lipid mediator PGE2 in supernatants of L. major-infected macrophages. The presence of B-1 cells also favors the production of lipid bodies by infected macrophages. In contrast, we failed to obtain the same effect on parasite replication inside L. major-infected macrophages when the B-1 cells were isolated from IL-10 knockout mice. CONCLUSION Our results show that elevated levels of PGE2 and IL-10 produced by B-1 cells increase L. major growth, as indicated by the number of parasites

New Sulfonamides Derived from Carvacrol: Compounds with High Antibacterial Activity against Resistant Staphylococcus aureus Strains

Nine new sulfonamides derived from carvacrol were prepared through a reaction between 4-hydroxy-2-isopropyl-5-methyl benzene sulfonyl chloride with various amines in excellent yields (76% - 92%). The sulfonamides were characterized using spectrometric and spectroscopic methods. Among these compounds, three derivatives showed excellent results in antibacterial activity against resistant S. aureus strains, with MIC values ranging from 3.9 to 62.50 ppm. The sulfonamide derivative of 4-methylaniline (SULF-1) had the best performance for all tested strains of bacteria (MIC = 3.9 to 15.62 ppm). Furthermore, the sulfonamide derivative of 4-fluoro aniline (SULF-3), which also presented promising results, was found to have a synergistic effect when combined with tetracycline and partial synergistic effect when combined with ampicillin, exhibiting an FIC index between 0.50 and 0.75. The sulfonamide derivative of 4-methylaniline had a synergistic effect in combination with erythromycin exhibiting an FIC index of 0.37. Carvacrol in association with the antibiotics tested did not have a synergistic effect.

Complex abdominal wall defects might be challenging for human and veterinary surgeons worldwide. Defects from trauma or congenital causes may lead to hernias development. The introduction of meshes to reinforce hernia repairs has improved surgical outcomes and several synthetic and biologic materials have been used. In this context, biomaterial prosthesis seems to be a satisfactory solution when managing great abdominal wall defects. The aim of the current study is to evaluate the bovine tunica albuginea (BTA) preserved in honey as graft material for rats’ abdominal wall reinforcement in incisional herniorrhaphy surgery as well as its viability, cicatrization and integration into the host tissue. Wistar rats were assigned to two main groups: 1) animals (n = 20) underwent median longitudinal laparotomy followed by laparorraphy with suture of the bovine tunica albuginea (BTA) graft as abdominal wall reinforcement;and 2) animals (n = 20) underwent only laparotomy and subsequent laparorrhaphy. Rats were clinically evaluated until euthanasia at post-surgical day 7, 14, 21 and 28. Necropsy and histopathological analysis of abdominal wall fragments were performed to compare groups and subgroups findings. BTA promoted abundant fibrosis, providing resistance and low postoperative complication rates. Besides, animals did not show rejection signs to the implant. In conclusion, BTA preserved in honey is an affordable, easy collection and handling biomaterial for graft, demanding simple surgical implantation technique for abdominal wall repair in rats.
