AIM:To compare the performance of three commercially available anti-human epidermalgrowth factor receptor 2(HER2)antibodies in whole-tissue sections and tissue microarrays(TMAs)of a series of gastric ***:We present a comparative analysis of three anti-HER2 antibodies(HercepTest,4B5 and SP3)using TMA and whole-tissue sections prepared from the same paraffin blocks of 199 gastric adenocarcinomas operated upon between January 2004 and December2008 at a Brazilian cancer *** data on the patients’age,sex,the anatomical location of the tumor and the Lauren’s histological classification were collected from clinical and pathological *** immunohistochemical(IHC)results were examined by two pathologists and the cases were classified as positive(3+),equivocal(2+)and negative(0 or 1+),according to the criteria of the IHC scoring system of gastric *** and whole-tissue sections were evaluated separately and *** cases yielding discordant IHC results and/or scored as 2+were subjected to dual-color in situ hybridization in order to determine the final HER2 *** determining the sensitivity and predictive value for HER2-positive status,we measured the accuracy of each antibody by calculating the area under the receiver operating characteristic(ROC)*** agreement between the results obtained using the TMAs and those obtained using the whole-tissue sections was assessed by means of Kappa ***:Intratumoral heterogeneity of HER2 expression was observed with all ***2-positive expression(3+)in the whole-tissue sections was observed in 23 cases(11.6%)using the 4B5 antibody,in 18 cases(9.1%)using the SP3 antibody and in 10 cases(5.1%)using the HercepTest *** the TMAs,11 positive cases(5.6%)were identified using SP3 antibody,9(4.6%)using the 4B5 antibody and 6(3%)using the HercepTest *** sensitivity using whole-tissue sections and TMA,respectively,was 95.2%and 42.9%with 4B5,90.5%and 66.7%

Thermal Modeling of a Novel Heated Tip Injector for Otto Cycle Engines Powered by Ethanol

This work presents a thermal modeling of a new cold-start system technology designed for Otto cycle combustion based on the electromagnetic heating principle. Firstly, the paper presents a state-of-the-art review and presents the context of automobile industry where heated injectors are necessary. The novel method of electromagnetic heating principle to solve the cold-start problem is still in the development phase and it enables engine starting at low temperatures in vehicles powered by ethanol or flex-fuel vehicles (FFV). This new system technology should be available as an alternative to replace the existing system. Currently, the cold-start system uses an auxiliary gasoline tank, which brings some inconvenience for the user. Secondly, the aim was also to create a physical model that takes into consideration all the parameters involved on the heating process such as power heating and average heat transfer coefficient. The study is based on the lumped system theory to model the ethanol heating process. From the analysis, two ordinary differential equations arise, which allowed an analytical solution. Particularly, an ethanol heating curve inside the injector was obtained, an important parameter in the process. Comparison with experimental data from other authors is also provided. Finally, a sensitivity analysis of controlling parameters such as heating power and heat transfer coefficient variation. The paper is concluded with suggestions for further studies.

In the past century, there have been many attempts to treat cancer with low levels of electric and magnetic fields. We have developed noninvasive biofeedback examination devices and techniques and discovered that patients with the same tumor type exhibit biofeedback responses to the same, precise frequencies. Intrabuccal administration of 27.12 MHz radiofrequency(RF) electromagnetic fields(EMF), which are amplitude-modulated at tumor-specific frequencies, results in long-term objective responses in patients with cancer and is not associated with any significant adverse effects. Intrabuccal administration allows for therapeutic delivery of very low and safe levels of EMF throughout the body as exemplified by responses observed in the femur, liver, adrenal glands, and lungs. In vitro studies have demonstrated that tumor-specific frequencies identified in patients with various forms of cancer are capable of blocking the growth of tumor cells in a tissue- and tumor-specific fashion. Current experimental evidence suggests that tumor-specific modulation frequencies regulate the expression of genes involved in migration and invasion and disrupt the mitotic spindle. This novel targeted treatment approach is emerging as an appealing therapeutic option for patients with advanced cancer given its excellent tolerability. Dissection of the molecular mechanisms accounting for the anti-cancer effects of tumor-specific modulation frequencies is likely to lead to the discovery of novel pathways in cancer.

In Brazil,gastric cancer is the third most common type of cancer among men and fifth among women,with an estimated 13360 new cases among men and 7870 among women each year during the 2020-2022 *** study presents reflections and attempts to add knowledge to the theme of quality of life(QoL)in patients with gastric adenocarcinoma and describes some of its characteristics in three regions of Brazil,with an evaluation of the disease’s impacts in various dimensions of life,as reported by the patients *** performed a narrative review of the literature and a data analysis of studies on QoL in Brazilian patients treated for gastric adenocarcinoma from three different cities in three geographic regions:Brasília(the midwest),Jaú(the southeast),and Macapá(the north).

Topotecan Use for Second-Line Treatment in Patients with Recurrent or Metastatic Cervical Cancer at Brazilian National Cancer Institute (INCA)

Objective: Cervical cancer represents the third most commonly diagnosed cancer and is an important cause of death for women suffering with malignancies. Patients who are refractory or progressed after first-line palliative treatment have a dismal prognosis and no second-line chemotherapy is considered standard so far. Several agents have been investigated in this setting and topotecan is one of the most characterized. The objective of this study was to evaluate response rate (RR), progression-free survival (PFS), overall survival (OS) and toxicity of topotecan in second palliative line for cervical cancer. Methods: An analysis was performed of all patients with recurrent or metastatic cervical cancer treated with topotecan in second palliative line at Brazilian National Cancer Institute, between 2008 and 2010. Results: A total of 73 courses of topotecan were given in the current study (median: 3.5 cycles;range 1 - 6). Anemia was the most frequent adverse event (grade 2:35%;grade 3:30%). Of the 20 patients evaluable, there were 2 partial responders to the treatment. The overall response rate (ORR) was 10%;3 patients (15%) had stable disease as maximum response. The median PFS for the entire group was 2.93 months (95% CI 2.41 - 3.45) and OS was 4.66 months (95% CI 1.21 - 8.11). Conclusion: The limited activity of topotecan schemas in second-line treatment of cervical cancer and the associated overall toxicity may not justify their use in this setting. Patients who progress after first-line treatment may be offered participation in clinical trials, other second-line agents or best supportive care measures.

Sample Dimension for Evaluating Characters of Yellow Mombin

The objective of this study was to determine the sample size needed to evaluate characters of mature fruits of yellow mombin (Spondias mombin L.). 100 fruits were evaluated from plants grown under a shading system. In the laboratory, these fruits were measured by the characters: longitudinal length (mm);equatorial diameter (mm);fruit mass (g);yield of pulp (%);total soluble solids—TSS (°Brix);titratable acidity—TA (% citric acid) and ratio (TSS/TA), being calculated the measures of central tendency and dispersion, and verified normality. Next, the sample size for each character was determined. For experimental evaluation of cajámirim fruits, considering an accuracy of 10% around the mean, 40 fruits per treatment should be evaluated, considering a completely randomized experiment.

Detailing the Structure of Liquid THF Based on an EPSR Study

The structure of pure liquid tetrahydrofuran (THF) has been investigated via Monte Carlo simulations in the wake of a previous Empirical Potential Structure Refinement (EPSR) study on that liquid (Bowron, D.T.;Finney, J.L.;Soper, A.K. (2006) J. Am. Chem. Soc., 128, 5119). The molecules are allatoms rigid structures and the intermolecular potential used is described for the classical 6-12 Lennard-Jones plus Coulomb in the NPT ensemble at 1 atm and 25°C. THF is a poorly structured liquid. Typical preferred orientation of molecules has been explored and calculations shown different types of molecular pairs exist concurrently in the liquid. The geometry of those pairs was deeply investigated and its influence in the liquid structure discussed. The lack of molecular organization in the liquid is closely related to the existence of that diversity of molecular pairs. Its geometry changes from antiparallel up to T-like depending on the distance between the molecules in the pairs.

Healthcare Professionals’ Adherence to Contact Precautions at a Maternal and Child Teaching Hospital

Background: Healthcare-associated infections affect hundreds of millions of patients worldwide. Children have greater susceptibility to healthcare-associated infections due to the immaturity of their immune system. Contact precautions aim to promote safety, protection and prevention of contamination. Thus, the objective of this study was to verify adherence to contact precaution measures, as well as compliance to the use of personal protective equipment. Methods: This cross-sectional observational study was carried out from July to October 2019 using a checklist to evaluate health professionals’ adherence to hand hygiene procedures and the use of gloves and surgical gowns when assisting children on contact precautions. Results: A total of 941 observations were carried out in a total of 300.532 hours. Hand hygiene was performed before and after contact with the patient in 58.84% and 75.09% of the cases, respectively and a surgical gown was used in 86.40% of the cases. The use of gloves was the variable most adhered to by professionals (87.57%). Intensive care unit professionals were the care workers who most complied with the regulation regarding hand hygiene after contact with the patient (p = 0.009) and the use of the surgical gown (p < 0.001). The correct hand hygiene technique was the recommendation with least adherence. Non-compliance to the hand hygiene technique was statistically significant among intensive care unit professionals (p = 0.002). Conclusions: Adherence to hand hygiene before contact with the patient and compliance with the hand hygiene technique were neglected by most professionals. However, there was good adherence to the use of surgical gloves and gowns, as well as high compliance to the techniques of removing these items. .
