Analysis of Short-Term Metabolic Alterations in Arabidopsis Following Changes in the Prevailing Environmental Conditions

Although a considerable increase in our knowledge concerning the importance of metabolic adjustments to unfavorable growth conditions has been recently provided, relatively little is known about the adjustments which occur in response to fluctuation in environmental factors. Evaluating the metabolic adjustments occurring under changing environmental conditions thus offers a good opportunity to increase our current understanding of the crosstalk between the major pathways which are affected by such conditions. To this end, plants growing under normal conditions were transferred to different light and temperature conditions which were anticipated to affect （amongst other processes） the rates of photosynthesis and photorespiration and characterized at the physiological, molecular, and metabolic levels following this transition. Our results revealed similar behavior in response to both treatments and imply a tight connec- tivity of photorespiration with the major pathways of plant metabolism. They further highlight that the majority of the regulation of these pathways is not mediated at the level of transcription but that leaf metabolism is rather pre-poised to adapt to changes in these input parameters.

Rotavirus induces intercellular calcium waves through ADP signaling

We found that rotavirus-infected cells produce paracrine signals that manifested as intercellular calcium waves(ICWs),observed in cell lines and human intestinal *** ICWs were caused by the release of extracellular adenosine 5'-diphosphate(ADP)that activated P2Y1 purinergic receptors on neighboring ***,rotavirus exploited paracrine purinergic signaling to generate ICWs that amplified the dysregulation of host cells and altered gastrointestinal physiology to cause diarrhea.

Sociability and pair-bonding in gerbils: a comparative experimental study

In a study of gerbils with contrasting social and mating systems (group-living monogamous Mongolian gerbil Meriones unguiculatus, solitary nonterritorial promiscuous midday jird M. meridianus, and solitary territorial promiscuous pale gerbil Gerbillus perpallidus), we employed part ner pref ere nee tests (PPTs) to assess among-species variation in sociability and pair-bonding patterns and tested whether the nature of contact between individuals: direct contact (DC) versus nondirect contact (NDC) affected our results. We measured male pref ere nces as the time: 1) spe nt alone, 2) with familiar (partner), and 3) unfamiliar (stranger) female in the 3-chambered apparatus. Gerbil species differed strongly in sociability and male partner preferences. The time spent alone was a reliable indicator of species sociability indepe ndent of the nature of con tact, whereas the pattern and level of betwee n-species differe nces in male partner preferences depended on con tact type: DC PPTs, un like NDC-tests, discriminated well betwee n monogamous and promiscuous species. In the DC-tests, stranger-directed aggression and stranger avoidanee were observed both in the highly social monogamous M. unguiculatus and the solitary territorial promiscuous G. perpallidus, but not in the non territorial promiscuous M. meridianus. In M. unguiculatus, stranger avoidanee in the DC-tests increased the time spent with the partner, thus providing evidence of a partner preference that was not found in the NDC-tests, whereas in G. perpallidus, stran ger avoidance in creased the time spe nt alone. This first comparative experimental study of partner prefere nces in gerbils provides new in sights into the in terspecific variation in gerbil sociality and mating systems and sheds light on behavioral mechanisms un derlyi ng social fidelity and pair-b on ding.

A plethora of studies investigating the origin and evolution of diverse mountain taxa has assumed a causal link between geological processes(orogenesis) and a biological response(diversification). Yet, a substantial delay(up to 30 Myr) between the start of orogenesis and diversification is often *** biologists should therefore identify alternative drivers of diversification and maintenance of biodiversity in mountain systems. Using phylogenetic, biogeographic, and diversification rate analyses,we could identify two independent processes that most likely explain the diversity of the widespread genus Allium in the Qinghaie-Tibet Plateau(QTP) region:(1) While the QTP-related taxa of the subgenus Melanocrommyum diversified in situ,(2) QTP-related taxa of other subgenera migrated into the QTP from multiple source areas. Furthermore, shifts in diversification rates within Allium could not be attributed spatially and temporally to the uplift history of the QTP region. Instead, global cooling and climate oscillations in the Quaternary were major contributors to increased speciation rates in three clades of Allium. Our study therefore adds to the growing evidence supporting the "mountain-geo-biodiversity hypothesis", which highlights the role of climate oscillations for the diversification of mountain organisms.

A Small Multifunctional Pentatricopeptide Repeat Protein in the Chloroplast of Chlamydomonas reinhardtii

Organellar biogenesis is mainly regulated by nucleus-encoded factors, which act on various steps of gene expression including RNA editing, processing, splicing, stabilization, and translation initiation. Among these regulatory factors, pentatricopeptide repeat （PPR） proteins form the largest family of RNA binding proteins, with hundreds of members in flowering plants. In striking contrast, the genome of the unicellular green alga Chlamydomonas reinhardtii encodes only 14 such proteins. In this study, we analyzed PPR7, the smallest and most highly expressed PPR protein in C. reinhardtii. Green fluorescent protein-based localization and gel-filtration analysis revealed that PPR7 forms a part of a high-molecular-weight ribonu- cleoprotein complex in the chloroplast stroma. RIP-chip analysis of PPRT-bound RNAs demonstrated that the protein associates with a diverse set of chloroplast transcripts in vivo, i.e. rrnS, psbH, rpoC2, rbcl, atpA, cemA-atpH, tscA, and atpl-psaJ. Furthermore, the investigation of PPR7 RNAi strains revealed that deple- tion of PPR7 results in a light-sensitive phenotype, accompanied by altered levels of its target RNAs that are compatible with the defects in their maturation or stabilization. PPR7 is thus an unusual type of small multi- functional PPR protein, which interacts, probably in conjunction with other RNA binding proteins, with numerous target RNAs to promote a variety of post-transcriptional events.

Primary gastric choriocarcinoma-a rare and aggressive tumor with multilineage differentiation: A case report

BACKGROUND Primary choriocarcinoma of the stomach (PCCS) is a rare tumor, with fewer than 60 cases published in the English-language literature up to December 2018. In this paper, we present the complex immunoprofile of one PCCS and a hypothesis regarding its histogenesis. CASE SUMMARY A 66-year-old previously healthy male underwent an emergency palliative gastrectomy for a gastric obstructive tumor. The histologic examination and immunoprofile of tumor cells showed a mixed tumor that consisted of choriocarcinoma (90%) and moderate differentiated adenocarcinoma (10%), with hepatic metastases (Stage pT2NxM1l1V1R0) and microsatelite stable status. The patient died one month after surgery. The tumor cells showed focal positivity for CDX2 (adenocarcinoma component), HCG (choriocarcinoma) and CD138 (plasmacytoid carcinoma component) and were negative for HER-2,α- fetoprotein, VEGF, maspin and markers of epithelial-mesenchymal transition. The gastric mucosa cells displayed positivity for CDX2, Hepar A and CD138. The complex immunoprofile and literature data synthesis prove that the gastric mucosa cells can present a multilineage differentiation. CONClUSION PCCS should be considered as an aggressive variant of microsatellite stable gastric adenocarcinoma of an epithelial type, and not a germ cell tumor.
