This a.ticle p.esents a.synop.is of the current da.a.on the mecha.isms of blood-bra.n ba.rier (BBB) a.tera.ion a.d a.toimmune resp.nse in a.ute ischemic stroke. Most resea.chers confirm the rela.ionship.between the severity of immunobiochemica. cha.ges a.d clinica. outcome of a.ute ischemic stroke. Ischemic stroke is a.comp.0}ied by a.ep.ic infla.ma.ion, which a.ters the bra.n tissue a.d exp.ses the co-stimula.ory molecules of the immune system a.d the neurona. a.tigens. To da.e, BBB is not considered the border between the immune system a.d centra. nervous system, a.d the loca. immune subsystems a.e found within a.d behind the BBB. BBB disrup.ion contributes to the lea.a.e of bra.n a.toa.tigens a.d induction of seconda.y a.toimmune resp.nse to neurona. a.tigens a.d long-term infla.ma.ion. Glymp.a.ic system function is a.tered a.d jeop.0}dized both in hemorrha.ic a.d ischemic stroke typ.s. The recep.ors of inna.e immunity (toll-like recep.or-2 a.d toll-like recep.or-4) a.e a.so involved in a.ute ischemia.rep.rfusion injury. Immune resp.nse is rela.ed to the key p.ocesses of blood clotting a.d fibrinolysis. a. the sa.e time, the stroke-induced immune a.tiva.ion ma. p.omote rep.0}a.ion p.enomena.in the bra.n. Subsequent resea.ch on the reduction of the a.ute ischemic bra.n injury through the ta.get regula.ion of the immune resp.nse is p.omising.

Effects of two doses of the a.ti-dia.etic drug, metformin （MF）, on hormona. a.d meta.olic levels of serum of non-dia.etic ma.e Wista. ra.s with 1,2-dimethylhydra.ine （DMH）-induced colon tumor a.enoca.cinoma. were studied. Ca.cinogenesis in the a.ima.s wa. a.so observed. Ra.s with DMH-induced colon a.enoca.cinoma. ha. eleva.ed levels of serum glucose, insulin, insulin- like growth fa.tor-l, tota. cholesterol, triglycerides, ca.a.a.e, ma.onic dia.dehyde, glyca.ed hemoglobin, a.p.0}ta.e a.inotra.sfera.e, a.d a.a.ine a.inotra.sfera.e a.d decrea.ed hemoglobin. Trea.ment with two doses of MF norma.ized ma.ority of these cha.ges in DMH-trea.ed ra.s, wherea. the drug wa. ineffective in ra.s without DMH trea.ment. The only excep.ion wa. the decrea.ed triglyceride levels in MF-trea.ed ra.s. a.100 mg/kg dose of MF increa.ed DMH-induced exop.ytic colon ca.cinoma. a.d decrea.ed endop.ytic tumors comp.0}ed with untrea.ed ra.s. Moreover, both MF doses increa.ed DMH-induced a.d highly differentia.ed tumors a.d decrea.ed the inva.iveness of colon ca.cinoma. comp.0}ed with ra.s p.ovided with DMH a.d wa.er. Therefore, effects of MF on meta.olic homeosta.is a.e critica. for p.eventing colon ca.cer.
