Hydrothermally induced diagenesis: Evidence from shallow marinedeltaic sediments, WilhelmØya, Svalbard

Sedimentary basins containing igneous intrusions within sedimentary reservoir units represent an important risk in petroleum exploration. The Upper Triassic to Lower Jurassic sediments at Wilhelm?ya(Svalbard) contains reservoir heterogeneity as a result of sill emplacement and represent a unique case study to better understand the effect of magmatic intrusions on the general burial diagenesis of siliciclastic sediments. Sills develop contact metamorphic aureoles by conduction as presented in many earlier studies. However, there is significant impact of localized hydrothermal circulation systems affecting reservoir sediments at considerable distance from the sill intrusions. Dolerite sill intrusions in the studied area are of limited vertical extent(～12 m thick), but created localized hydrothermal convection cells affecting sediments at considerable distance(more than five times the thickness of the sill)from the intrusions. We present evidence that the sedimentary sequence can be divided into two units:(1) the bulk poorly lithified sediment with a maximum burial temperature much lower than 60-70 ℃,and(2) thinner intervals outside the contact zone that have experienced hydrothermal temperatures(around 140 ℃). The main diagenetic alteration associated with normal burial diagenesis is minor mechanical plastic deformation of ductile grains such as mica. Mineral grain contacts show no evidence of pressure dissolution and the vitrinite reflectance suggests a maximum temperature of ～40 ℃. Contrary to this, part of the sediment, preferentially along calcite cemented flow baffles, show evidence of hydrothermal alteration. These hydrothermally altered sediment sections are characterized by recrystallized carbonate cemented intervals. Further, the hydrothermal solutions have resulted in localized sericitization(illitization) of feldspars, albitization of both K-feldspar and plagioclase and the formation of fibrous illite nucleated on kaolinite. Metabolic dysfunction-associated steatotic liver disease:Navigating terminological evolution,diagnostic frontiers and therapeutic horizon-an editorial exploration

Metabolic dysfunction-associated steatotic liver disease(MASLD),once known as non-alcoholic fatty liver disease(NAFLD),represents a spectrum of liver disorders characterized by lipid accumulation within hepatocytes.The redefinition of NAFLD in 2023 marked a significant reposition in terminology,emphasizing a broader understanding of liver steatosis and its associated risks.MASLD is now recognized as a major risk factor for liver cirrhosis,hepatocellular carcinoma,and systemic complications such as cardiovascular diseases or systemic inflammation.Diagnostic challenges arise,particularly in identifying MASLD in lean individuals,necessitating updated diagnostic protocols and investing in non-invasive diagnostic tools.Therapeutically,there is an urgent need for effective treatments targeting MASLD,with emerging pharmacological options focusing on,among others,carbohydrate and lipid metabolism.Additionally,understanding the roles of bile acid metabolism,the microbiome,and dietary interventions in MASLD pathogenesis and management holds promise for innovative therapeutic approaches.There is a strong need to emphasize the importance of collaborative efforts in understanding,diagnosing,and managing MASLD to improve physicians’approaches and patient outcomes.

The structural and associated molecules of the extracellular matrix(ECM)complex is an important component of the local milieu of cells,both for maintaining their functions and homeostasis.It is a dynamic structure that is finely tuned to changes in the microenvironment.One of these factors is hypoxia,which can arise in tissues due to physiological or pathological effects.As a result of the hypoxic effect,the properties of the ECM are significantly modified,stiffness increases,the balance between degradation and synthesis of structural proteins shifts,and the deposition of biologically active mediators’changes.Hypoxia-inducible factors(HIFs)contribute significantly to the modification of the properties of the ECM under hypoxia.Among the three HIF isoforms,HIF-1 is the most studied,with numerous identified target genes encoding proteins that participate in all matrisome compartments.Much less is known about HIF-2 and HIF-3.In the context of the effects of hypoxia on the matrisome,HIF-3 isoform is of particular interest.Unlike the first two isoforms,transiently expressed during the first hours of hypoxia,HIF-3 is activated after around 24 h of exposure to hypoxia and persists for a longer period.In addition,its transcription is more pronounced under hypoxia than are HIFs 1 and 2.HIF-3,rather than the first two isoforms,is possibly responsible for the changes that occur in matrisome during prolonged hypoxic conditions.This review attempts to summarize the available data on the involvement of HIF-3 in the matrisome modification under hypoxia.

Physiological strategies in wild rodents:immune defenses of commensal rats

The importance of issues associated with urban/commensal rats and mice(property damage,management costs,and health risks)press upon research on these animals.While the demography of commensal rodents is mostly studied,the need for understanding factors influencing their natural morbidity/mortality is also stressed.In this respect,more attention is expected to be paid to immunity,the physiological mechanism of defense against host survival threats(pathogens,parasites,diseases).Commensal rats and mice carry numerous pathogens that evoke diverse immune responses.The state of immunity in commensal house mice is studied in great detail,owing to the use of laboratory strains in biomedical research.Because commensal rats are,compared to mice,carriers of more zoonotic agents,rats’immunity is studied mainly in that context.Some of these zoonotic agents cause chronic,asymptomatic infections,which justified studies of immunological mechanisms of pathogen tolerance versus clearance regulation in rats.Occurrence of some infections in specific tissues/organs pressed upon analysis of local/regional immune responses and/or immunopathology.A survey of immunological activity/responses in commensal rats is given in this review,with mention of existing data in commensal mice.It should throw some light on the factors relevant to their morbidity and lifespan,supplementing the knowledge of commensal rodent ecology.

Catalytic co-combustion of biomass and brown coal in a fluidized bed:Economic and environmental benefits

The work is devoted to the study of combustion of brown coal,pine sawdust,and their mixtures in a fluidized bed of catalyst at 600-750℃.It is shown that an increase in the content of sawdust in a mixture with brown coal leads to an increase in the burnout degree of solid fuel from 94.4%to 99.9%,while the emission of greenhouse gases in the form of CO_(2)CO and NOxis reduced(CO_(2)from the biomass is not included in the balance).The high content of alkaline earth metal oxides(CaO and MgO)in the mineral part of brown coal,sawdust,and their mixtures eliminates the emission of sulfur oxides and the slagging of heat-exchange surfaces during the combustion in a fluidized bed of catalyst.The optimal temperature,when the highest burnout degree of the above fuels is achieved in the combustion is 750℃.It is also shown that the increase in temperature and the content of sawdust in the composition of the fuel mixtures has a positive effect on the economic and environmental process indicators.

Development and characterization of 3D-printed electroconductive pHEMA-co-MAA NP-laden hydrogels for tissue engineering

Tissue engineering(TE)continues to be widely explored as a potential solution to meet critical clinical needs for diseased tissue replacement and tissue regeneration.In this study,we developed a poly(2-hydroxyethyl methacrylate-co-methacrylic acid)(pHEMA-co-MAA)based hydrogel loaded with newly synthesized conductive poly(3,4-ethylene-dioxythiophene)(PEDOT)and polypyrrole(PPy)nanoparticles(NPs),and subsequently processed these hydrogels into tissue engineered constructs via three-dimensional(3D)printing.The presence of the NPs was critical as they altered the rheological properties during printing.However,all samples exhibited suitable shear thinning properties,allowing for the development of an optimized processing window for 3D printing.Samples were 3D printed into pre-determined disk-shaped configurations of 2 and 10 mm in height and diameter,respectively.We observed that the NPs disrupted the gel crosslinking efficiencies,leading to shorter degradation times and compressive mechanical properties ranging between 450 and 550 kPa.The conductivity of the printed hydrogels increased along with the NP concentration to(5.10±0.37)×10^(−7)S/cm.In vitro studies with cortical astrocyte cell cultures demonstrated that exposure to the pHEMA-co-MAA NP hydrogels yielded high cellular viability and proliferation rates.Finally,hydrogel antimicrobial studies with staphylococcus epidermidis bacteria revealed that the developed hydrogels affected bacterial growth.Taken together,these materials show promise for various TE strategies.

Polyelectrolyte Complexes between Hyperbranched and Linear Polysaccharides: Fucoidan/Chitosan

Polyelectrolyte complexes(PECs)of hyperbranched(HB)and linear polysaccharides are promising as more effective encapsulation agents compared to PECs formed by linear polysaccharides.We investigated the PECs between the HB anionic polysaccharide fucoidan(FUC)and the cationic linear polysaccharide chitosan(CS).The FUC had a molecular weight(MW)of 30×106.The PECs were prepared in three solvents(water,0.01 and 0.1 mol/L acetic acid)with CS of MW of 15,110 and 170 kDa,and deacetylation degrees(DDA)of 70%and 97%.The structures of the PECs and the initial FUC were investigated by multi-angle static and dynamic light scattering.As the FUC contained 18 wt%of—OSO3 groups and 5 wt%of uronic acid units,it was a“strong-weak”copolyanion,so the HB macromolecules of the FUC formed nanogel particles in 0.1 mol/L AcOH and open branched structures in water,as confirmed by the Kratky plots.After mixing the solutions of original components,the PEC structures underwent an equilibration period,the duration of which increased with the MW of CS.As the charge stoichiometry was approached,the PECs shrank;the fractal dimension approached unity,indicating the side-by-side packing of adjacent FUC branches with the help of CS.Secondary aggregation in the vicinity of the charge compensation was hardly observed,as it occurred in a very narrow region.The PEC content at theζ-potential inversion depended on solvents’pH and the DDA of CS.In the extreme case of core-shell PECs in 0.1 mol/L AcOH,obtained by mixing FUC nanogels with the solutions of high MW CS of 97%DDA,the protruding tails of CS formed a positively charged shell in the whole range of FUC content(10 wt%

Rare extraintestinal manifestations of ulcerative colitis treated with dual biologic therapy:A case report

BACKGROUND Ulcerative colitis(UC)is an idiopathic,chronic inflammatory bowel disease(IBD)most often located in the rectum,but may involve the entire colon.Extra intestinal manifestations(EIMs)occur with varying frequency depending on the affected organ.The most common ones are musculoskeletal EIMs,affecting up to 33%-40%of IBD patients.These include,among others,inflammatory back pain,tendinitis,plantar fasciitis and arthritis.Only a few case reports in literature discuss Achilles tendinitis.CASE SUMMARY This report describes a patient with UC and Achilles tendinitis in whom after many unsuccessful attempts of treatment with sulfasalazine,mesalazine,glucocorticosteroids,infliximab and tofacitinib,a complete UC remission and resolution of Achilles tendinitis were achieved with the use of dual biologic therapy(DBT)-ustekinumab and adalimumab(ADA).CONCLUSION This case mentions rare EIMs of UC and suggests that DBT may be an alternative for patient with ulcerative colitis and EIMs.

Inﬂammatory myoﬁbroblastic tumor(IMT)is a rare neoplasm with intermediate malignancy characterized by a propensity for recurrence but a low metastatic rate.Diagnostic challenges arise from the diverse pathological presentation,variable symptomatology,and lack of different imaging features.However,IMT is identiﬁed by the fusion of the anaplastic lymphoma kinase(ALK)gene,which is present in approximately 70%of cases,with various fusion partners,including ran-binding protein 2(RANBP2),which allows conﬁrmation of the diagnosis.While surgery is the preferred approach for localized tumors,the optimal long-term treatment for advanced or metastatic disease is difﬁcult to deﬁne.Targeted therapies are crucial for achieving sustained response to treatment within the context of genetic alteration in IMT.Crizotinib,an ALK tyrosine kinase inhibitor(TKI),was ofﬁcially approved by the US Food and Drug Administration(FDA)in 2020 to treat IMT with ALK rearrangement.However,most patients face resistance and disease progression,requiring consideration of sequential treatments.Combining radiotherapy with targeted therapy appears to be beneﬁcial in this indication.Early promising results have also been achieved with immunotherapy,indicating potential for combined therapy approaches.However,deﬁned recommendations are still lacking.This review analyzes the available research on IMT,including genetic disorders and their impact on the course of the disease,data on the latest targeted therapy regimens and the possibility of developing immunotherapy in this indication,as well as summarizing general knowledge about prognostic and predictive factors,also in terms of resistance to systemic therapy.

Brain-derived neurotrophic factor signaling in the neuromuscular junction during developmental axonal competition and synapse elimination

During the development of the nervous system,there is an overproduction of neurons and synapses.Hebbian competition between neighboring nerve endings and synapses performing different activity levels leads to their elimination or strengthening.We have extensively studied the involvement of the brain-derived neurotrophic factor-Tropomyosin-related kinase B receptor neurotrophic retrograde pathway,at the neuromuscular junction,in the axonal development and synapse elimination process versus the synapse consolidation.The purpose of this review is to describe the neurotrophic influence on developmental synapse elimination,in relation to other molecular pathways that we and others have found to regulate this process.In particular,we summarize our published results based on transmitter release analysis and axonal counts to show the different involvement of the presynaptic acetylcholine muscarinic autoreceptors,coupled to downstream serine-threonine protein kinases A and C(PKA and PKC)and voltage-gated calcium channels,at different nerve endings in developmental competition.The dynamic changes that occur simultaneously in several nerve terminals and synapses converge across a postsynaptic site,influence each other,and require careful studies to individualize the mechanisms of specific endings.We describe an activity-dependent balance(related to the extent of transmitter release)between the presynaptic muscarinic subtypes and the neurotrophin-mediated TrkB/p75NTR pathways that can influence the timing and fate of the competitive interactions between the different axon terminals.The downstream displacement of the PKA/PKC activity ratio to lower values,both in competing nerve terminals and at postsynaptic sites,plays a relevant role in controlling the elimination of supernumerary synapses.Finally,calcium entry through L-and P/Q-subtypes of voltage-gated calcium channels(both channels are present,together with the N-type channel in developing nerve terminals)contributes to reduce transmitter release and promote withdrawal o
