Complement-dependent neuroinflammation in spinal cord injury:from pathology to therapeutic implications

Spinal cord injury remains a major cause of disability in young adults,and beyond acute decompression and rehabilitation,there are no pharmacological treatments to limit the progression of injury and optimize recovery in this *** the thorough investigation of the complement system in triggering and propagating cerebral neuroinflammation,a similar role for complement in spinal neuroinflammation is a focus of ongoing *** this work,we survey the current literature investigating the role of complement in spinal cord injury including the sources of complement proteins,triggers of complement activation,and role of effector functions in the *** study relevant data demonstrating the different triggers of complement activation after spinal cord injury including direct binding to cellular debris,and or activation via antibody binding to damage-associated molecular *** effector functions of complement have been implicated in spinal cord injury,and we critically evaluate recent studies on the dual role of complement anaphylatoxins in spinal cord injury while emphasizing the lack of pathophysiological understanding of the role of opsonins in spinal cord *** this pathophysiological review,we systematically review the different translational approaches used in preclinical models of spinal cord injury and discuss the challenges for future translation into human *** review emphasizes the need for future studies to dissect the roles of different complement pathways in the pathology of spinal cord injury,to evaluate the phases of involvement of opsonins and anaphylatoxins,and to study the role of complement in white matter degeneration and regeneration using translational strategies to supplement genetic models.

The diversity of vegetation configuration is the key to ecological restoration in open-pit coal mine ***,the recovery outcomes of different areas with the same vegetation assemblage pattern are completely different after long-term ***,understanding the causes of differential vegetation recovery and the mechanism of plant succession is of great significance to the ecological restoration of *** Pinus tabulaeformis plantations with similar initial site conditions and restoration measures but with different secondary succession processes were selected from the open-pit coal mine dump that has been restored for 30 *** physicochemical properties,enzyme activities,vegetation and microbial features were investigated,while the structural equation models were established to explore the interactions between plants,soil and *** results showed that original vegetation configuration and soil nutrient conditions were altered due to secondary *** the advancement of the secondary succession process,the coverage of plants increased from 34.8%to 95.5%(P<0.05),soil organic matter increased from 9.30 g kg^(-1)to 21.13 g kg^(-1)(P<0.05),and total nitrogen increased from 0.38 g kg^(-1)to 1.01 g kg^(-1)(P<0.05).The activities of soil urease and p-glucosidase were increased by 1.7-fold and 53.26%,***,the secondary succession also changed the soil microbial community structure and *** relative abundance of Nitrospira genus which dominates the nitrification increased *** results showed that urease andβ-glucosidase promoted the increase of vegetation diversity and biomass by promoting the accumulation of soil organic matter and nitrate nitrogen,which promoted the ecological restoration of mine dumps.

Epidemiological Investigation and Genetic Characterization of Type 2 PCV2 （Type 2 Porcine Circovirus） in Mexican Commercial Herds

PCV2 （Porcine circovirus type 2） is considered as the essential infectious agent of PMWS （post weaning multisystemic wasting syndrome） in pigs. Serological studies have shown that the virus is ubiquitous. Currently, there are many reports about the epidemiology and genetic characteristics of this virus around the world, but in Mexico it has not been studied. More than 3,500 samples of serum, rectal swabs, tissues and semen of 34 Mexican porcine farms from 10 important producer regions were analyzed by PCR. Results show that 97% of the farms were positive. Both genotypes, PCV2a and PCV2b were detected. It also found that the most prevalent genotype in Mexico is PCV2b. Regarding to amino acid sequence; three major heterogenic regions were present in the positions 59-91,123-136 and 185-210.

Assessment of k–ε models using tetrahedral grids to describe the turbulent flow field of a PBT impeller and validation through the PIV technique

In turbulence modeling, the RNG and Realizable models have important improvements in the turbulent production and dissipation terms in comparison to the Standard. The selection of the appropriate turbulence model has an impact on the convergence and solution in STRs, and they are used in mixing, multiphase modeling or as starting solution of transient models as DES and LES. Although there are several studies with the pitched blade turbine(PBT) impeller, most of them used the Standard model as representative of all k–ε models, using structured hexahedral grids composed of low number of cells, and in some cases under axial symmetry ***, in this work the assessment of the Standard, RNG and Realizable models to describe the turbulent flow field of this impeller, using the Multiple Reference Frame(MRF) and Sliding Mesh(SM) approaches with tetrahedral domains in dense grids, is presented. This kind of cell elements is especially suitable to reproduce complex geometries. Flow velocities and turbulent parameters were verified experimentally by PIV and torque measurements. The three models were capable of predicting fairly the pumping number, the power number based on torque, and velocities. Although the RNG improved the predictions of the turbulent kinetic energy and dissipation rate, the Realizable model presented better performance for both approaches. All models failed in the prediction of the total dissipation rate, and a dependence of its value on the number of cells for the MRF was found.

Extreme drought triggers parallel shifts in wood anatomical and physiological traits in upper treeline of the Mediterranean Andes

Background Treeline ecotones of Mediterranean ecoregions have been affected by the increasing intensity and severity of droughts. Even though the effect of droughts on forest dynamics has been widely documented, knowledge is relatively scarce of how extreme climate episodes affect the hydraulic structure and, therefore, the phys-iology of woody plants. The Mediterranean Andes have experienced an uninterrupted period of drought since 2010, including an extremely dry year in 2019 with approximately 80% rainfall deficit. Here, we investigated shifts in wood anatomical and physiological traits of Kageneckia angustifolia, an endemic treeline species, in response to this drought *** We evaluated the xylem plasticity of three K. angustifolia populations across their natural distribution(31–35° SL) based on anatomical(vessel structure and distribution) and physiological(intrinsic water-use efficiency) variables in the tree rings. We focused on the period 2000–2020 that corresponds to before the megadrought(2000–2007),(ii) megadrought(2008–2018) and(iii) hyperdrought(2019–2020). The variables were annualized and analyzed by linear mixed-effects *** Our results provide insights to the anatomical and physiological mechanisms underlying the resilience of treeline forests to persistent droughts in central Chile. We found that the extreme drought in 2019–2020 triggered shifts in vessel size and frequency that increased hydraulic safety. These significant shifts in vessel traits occurred in parallel with a decrease in pit aperture area and an increase in water-use efficiency, further increasing the resilience of K. angustifolia to extreme drought *** Our results revealed coordinated shifts in vessel size and frequency and water-use efficiency in response to the megadrought, thereby reducing vulnerability to hydraulic failure. Energy Signaling in the Regulation of Gene Expression during Stress

Maintenance of homeostasis is pivotal to all forms of life. In the case of plants, homeostasis is constantly threatened by the inability to escape environmental fluctuations, and therefore sensitive mechanisms must have evolved to allow rapid perception of environmental cues and concomitant modification of growth and developmental patterns for adaptation and survival. Re-establishment of homeostasis in response to environmental perturbations requires reprog- ramming of metabolism and gene expression to shunt energy sources from growth-related biosynthetic processes to defense, acclimation, and, ultimately, adaptation. Failure to mount an initial ＇emergency＇ response may result in nutrient deprivation and irreversible senescence and cell death. Early signaling events largely determine the capacity of plants to orchestrate a successful adaptive response. Early events, on the other hand, are likely to be shared by different conditions through the generation of similar signals and before more specific responses are elaborated. Recent studies lend credence to this hypothesis, underpinning the importance of a shared energy signal in the transcriptional response to various types of stress. Energy deficiency is associated with most environmental perturbations due to their direct or indirect deleterious impact on photosynthesis and/or respiration. Several systems are known to have evolved for monitoring the available resources and triggering metabolic, growth, and developmental decisions accordingly. In doing so, energy-sensing systems regulate gene expression at multiple levels to allow flexibility in the diversity and the kinetics of the stress response.

Computing the topology of an arrangement of implicitly defined real algebraic plane curves~§

We introduce a new algebraic approach dealing with the problem of computing the topology of an arrangement of a finite set of real algebraic plane curves presented implicitly. The main achievement of the presented method is a complete avoidance of irrational numbers that appear when using the sweeping method in the classical way for solving the problem at hand. Therefore, it is worth mentioning that the efficiency of the proposed method is only assured for low-degree curves.
