Morphometry and subpopulation structure of Holstein bull spermatozoa： variations in ejaculates and cryopreservation straws

Sperm quality is evaluated for the calculation of sperm dosage in artificial reproductive programs. The most common parameter used is motility, but morphology has a higher potential as a predictor of genetic quality. Morphometry calculations from CASA-Morph technology improve morphological evaluation and allow mathematical approaches to the problem. Semen from 28 Holstein bulls was collected by artificial vagina, and several ejaculates were studied. After general evaluation, samples were diluted, packaged in 0.25 ml straws, and stored in liquid nitrogen. Two straws per sample were thawed, and slides were processed and stained with Diff-Quik. Samples were analyzed by a CASA-Morph system for eight morphometric parameters. In addition to the ＂classical＂ statistical approach, based on variance analysis （revealing differences between animals, ejaculates, and straws）, principal component （PC） analysis showed that the variables were grouped into PC1, related to size, and PC2 to shape. Subpopulation structure analysis showed four groups, namely, big, small, short, and narrow from their dominant characteristics, representing 31.0%, 27.3%, 24.1%, and 17.7% of the total population, respectively. The distributions varied between animals and ejaculates, but between straws, there were no differences in only four animals. This modern approach of considering an ejaculate sperm population as divided into subpopulations reflecting quantifiable parameters generated by CASA-Morph systems technology opens a new view on sperm function. This is the first study applying this approach to evaluate different ejaculates and straws from the same individual. More work must be done to improve seminal dose calculations in assisted reproductive programs.

The cumulative effects of periodic redox cycling on the mobility of As,Fe,and S from alluvial sediment to groundwater were investigated in bioreactor *** particular sediments from the alluvial floodplain of the Mekong Delta River were investigated: Matrix A (14 m deep) had a higher pyrite concentration than matrix B (7 m deep) *** present in matrix B but absent in matrix *** the reactors,the sediment suspensions were supplemented with As(III) and SO4^2-,and were subjected to three full-redox cycles entailing phases of nitrogen/CO2,compressed air sparging,and cellobiose *** differences in As concentration and speciation were observed upon redox *** support the fact that initial sediment composition is the main factor controlling arsenic release and its speciation during the redox ***,a high pyrite content associated with a low SO4^2- content resulted in an increase in dissolved As concentrations,mainly in the form of As(III),after anoxic half-cycles;whereas a decrease in As concentrations mainly in the form of As(V),was instead observed after oxic *** addition,oxic conditions were found to be responsible for pyrite and arsenian pyrite oxidation,increasing the As pool available for *** same processes seem to occur in sediment with the presence of gypsum,but,in this case,dissolved As were sequestered by biotic or abiotic redox reactions occurring in the FeeS system,and by specific physico-chemical condition (***).The contrasting results obtained for two sediments sampled from the same core show that many complexes and entangled factors are at work,and further refinement is needed to explain the spatial and temporal variability of As release to groundwater of the Mekong River Delta (Vietnam).

Influence of Induction Motor Design Data on Effects of Cage Asymmetry

The purpose of this paper is to analyze influence of design data on a component （1 - 2s）~ in stator currents of induction motors, mainly used for cage fault diagnosis. This paper shows that such an approach does not always lead to a correct outcome. The considerations are based on a ＂classical＂ model of induction motors extended to cage asymmetry by introducing cage asymmetry factors ko~ and ka. It has been found that in order to estimate the level of the component （1 - 2s）7~, it is enough to know the pole-pair number ＂p＂ and the number of rotor slots ＂N＂. The main objective of the paper is to provide engineers with simple qualitative prediction of effects due to cage faults for various motors when information on design data is very limited.

The presence and outcome of biliary sphincter disorders in liver-transplant recipients according to the Rome IV classification

Background Biliary sphincter disorders after liver transplantation(LT)are poorly *** aim to describe the presence and outcome of patients with papillary stenosis(PS)and functional biliary sphincter disorders(FBSDs)after LT according to the updated Rome IV *** We reviewed all endoscopic retrograde cholangiopancreatographies(ERCPs)performed in LT recipients between January 2003 and December *** on clinical and endoscopic findings was obtained from electronic health records and endoscopy *** and clinical findings were collected at the time of ERCP and 1 month after *** Among the 1,307 LT recipients,336 underwent 849 ***(1.0%)patients met the updated Rome IV criteria for PS[former sphincter of Oddi dysfunction(SOD)type I]and 14 patients(1.0%)met the Rome IV criteria for FBSD(former SOD type II).Biliary sphincterotomy was performed in 13 PS and 10 FBSD *** month after sphincterotomy,bilirubin,gamma-glutamyl transferase and alkaline phosphatase levels decreased in 85%,61%,and 92%of those in the PS group(P¼0.019,0.087,and 0.003,respectively)and in 50%,70%,and 80%of those in the FBSD group(P¼0.721,0.013,and 0.093,respectively).All the 14 patients initially suspected of having a FBSD turned out to have a different diagnosis during the *** PS after LT is uncommon and occurs in only 1%of LT *** data do not support the presence of an FBSD after *** is a safe and effective procedure in LT recipients with PS.

Winter Wheat Crop Height Estimation Using Small Unmanned Aerial System (sUAS)

Deploying the small Unmanned Aerial System (sUAS) for data collection of high-resolution images is a big potential in determining crop physiological parameters. The advantage of using sUAS technology is the ability to acquire a high-resolution orthophoto and a 3D Model which is highly suitable for plant height monitoring. Plant height estimation has a big impact in the growth and development of wheat because it is essential for obtaining biomass, which is a factor for higher crop yield. Plant height is an indicator of high yield estimation and it correlates to biomass, nitrogen content, and other plant growth parameters. The study is aimed to determine an accurate height of wheat using the sUAS generated Digital Surface Model (DSM). A high-resolution imagery between 1.0 - 1.2 cm/pixel was obtained from a 35 m altitude with area coverage of 1.01 hectares. The DSM and orthophoto were generated from the sUAS, and the computed wheat heights were derived from the difference of Digital Elevation Model (DEM) and DSM data. Field measurement using steel tape was done for ground truth. The sUAS-based wheat height data were evaluated using the ground truth of 66 wheat-rows by applying correlation and linear regression analysis. Datasets were collected from three different flight campaigns (March 2018-May 2018). The sUAS-based wheat height data were significantly correlated, obtaining the result of R2 = 0.988, R2 = 0.996 and R2 = 0.944 for the month of March, April and May 2018 respectively. The significance of linear regression results was also validated by computing for the p-value. The p-value results were 0.00064, 0.0000824 and 0.0058 respectively. The main concern is the lodging of winter wheat, especially during the month of April which affects the recording of the plant’s height. Olfactory ensheathing glia promote axonal regeneration in the mammalian central nervous system,including retinal ganglion cell axonal growth through the injured optic ***,it is unknown whether olfactory ensheathing glia also have neuroprotective *** ensheathing glia express brain-derived neurotrophic factor,one of the best neuroprotectants for axotomized retinal ganglion ***,we aimed to investigate the neuroprotective capacity of olfactory ensheating glia after optic nerve *** ensheathing glia cells from an established rat immortalized clonal cell line,TEG3,were intravitreally injected in intact and axotomized retinas in syngeneic and allogeneic mode with or without microglial inhibition or immunosuppressive *** and gene expression analyses were *** bulb-derived primary olfactory ensheathing glia and TEG3 express major histocompatibility complex classⅡ*** and syngenically transplanted TEG3 cells survived in the vitreous for up to 21 days,forming an *** axotomized retinas,only the allogeneic TEG3 transplant rescued retinal ganglion cells at 7 days but not at 21 *** these retinas,microglial anatomical activation was higher than after optic nerve crush *** intact retinas,both transplants activated microglial cells and caused retinal ganglion cell death at 21 days,a loss that was higher after allotransplantation,triggered by pyroptosis and partially rescued by microglial inhibition or ***,neuroprotection of axotomized retinal ganglion cells did not improve with these *** different neuroprotective properties,different toxic effects,and different responses to microglial inhibitory treatments of olfactory ensheathing glia in the retina depending on the type of transplant highlight the importance of thorough preclinical studies to explore these variables.
