While hypoxic signaling has been shown to play a role in many cellular processes,its role in metabolism-linked extracellular matrix(ECM)organization and downstream processes of cell fate after musculoskeletal injury remains to be ***(HO)is a debilitating condition where abnormal bone formation occurs within extra-skeletal *** andhypoxia-inducible factor 1α(HIF-1α)activation have been shown to promote ***,the underlying molecular mechanisms bywhich the HIF-1αpathway in mesenchymal progenitor cells(MPCs)contributes to pathologic bone formation remain to ***,we used a proven mouse injury-induced HO model to investigate the role of HIF-1αon aberrant cell ***-cell RNA sequencing(scRNA-seq)and spatial transcriptomics analyses of the HO site,we found that collagen ECM organizationis the most highly up-regulated biological process in *** mesenchymal cell-specific deletion of Hif1α(Hoxa11-CreER^(T2);Hif1a^(fl/fl))significantly mitigated HO in ***-seq analysis of these Hoxa11-CreER^(T2);Hif1a^(fl/fl)mice identified the PlOD2/lOXpathway for collagen cross-linking as downstream of the HIF-1αregulation of ***,our scRNA-seq data and mechanisticstudies further uncovered that glucose metabolism in MPCs is most highly impacted by HIF-1α*** a translational aspect,a pan-lOX inhibitor significantly decreased HO.A newly screened compound revealed that the inhibition of PlOD2 activity in MPCssignificantly decreased osteogenic differentiation and glycolytic *** suggests that the HIF-1α/PlOD2/lOX axis linked tometabolism regulates HO-forming MPC *** results suggest that the HIF-1α/PlOD2/lOX pathway represents a promisingstrategy to mitigate HO formation.

BACKGROUND Percutaneous coronary intervention(PCI)of diseased saphenous vein grafts(SVG)continues to pose a clinical *** PCI guidelines give a class III recommendation against performing PCI on chronically occluded ***,contemporary outcomes after SVG intervention have incrementally improved with distal protection devices,intracoronary vasodilators,drug-eluting stents,and prolonged dual antiplatelet *** To reassess the procedural and long-term outcomes of PCI for totally occluded SVG with contemporary *** This was a retrospective observational study conducted at a single university *** study population consisted of 35 consecutive patients undergoing PCI of totally occluded ***-procedure dual antiplatelet therapy was continued for a minimum of one year and aspirin was continued *** outcomes were assessed at a mean follow-up of 1221±1038 *** primary outcome was freedom from a major adverse cardiac event(MACE)defined as the occurrence of any of the following:death,myocardial infarction,stroke,repeat bypass surgery,repeat PCI,or graft *** The study group included 29 men and 6 women with a mean age of 69±12 *** was present in 14(40%)*** patients had Canadian Heart Classification class III or IV *** presentation was an acute coronary syndrome in 34(97%)*** SVG age was 12±5 *** duration of occlusion was acute(24 h to 30 d)in 26%,and late(>30 d)in 40%.PCI was initially successful in 29/35 SVG occlusions(83%).Total stent length was 52±35 *** complications of distal embolization or no-reflow occurred in 6(17%)*** longer term follow-up,MACE-free survival was only 30%at 3 years and 17%at 5 *** PCI of totally occluded SVG can be performed with a high procedural success ***,its clinical utility remains limited by poor follow-up outcomes.

Cyclic polyolefin (COP) is an inexpensive hydrophobic material with low auto-fluorescence, high light transmittance and thermal stability, broad chemical resistance and no non-specific protein binding. Here, the hydrophobic alkane COP was modified to have carbonyl functionalities through oxygen plasma and chemical etching treatments to increase usefulness for chemical and biochemical applications. Then, biotin-hydrazide was used to create biotinylated surfaces that bound streptavidin. A biotinylated target oligonucleotide was subsequently bound to the immobilized biotin-streptavidin and ligation mediated rolling circle amplification-based (l-RCA) SNP detection was demonstrated.

美国放射学院(American College of Radiology,ACR)偶发病变委员会(Incidental Findings Committee,IFC)发布了CT或MR偶发胰腺囊肿的管理建议。该建议是对美国放射学院杂志(JACR)2010年发布的肾上腺、肾脏、肝脏以及胰腺偶发病变管理白皮书中胰腺部分的更新。该管理方案由多位腹部影像医师、1位胃肠道学医师以及1位胰腺外科医师所组成的胰腺亚组委员会制定。该方案参考已发表的文献和专家意见,经反复协商,最终达成共识。该方案的分支成功地根据患者的临床特点及影像特征将胰腺囊肿进行分类,评估终点为良性和/或进展缓慢病变的诊断(足以结束随访)或具体的管理建议。该方案适用于大多数但非全部的病理及临床情况。该方案旨在通过提供胰腺偶发囊肿的管理指导,来提高患者的管理质量。

目的选择性活化巨噬细胞（Alternativelyactivated macrophages, M2）与胃部解痉多肽表达性化生（spasmolytic polypeptide-expressingmetaplasia, SPEM）过程相关。然而，诱导SPEM的确切机制及关键介质尚不明确。研究设计为确定这一过程中的关键基因，分离胃部SPEM小鼠（DMP-777处理）及高级胃部SPEM小鼠（l635处理）胃体的巨噬细胞进行RNA测序。评估白介素．33、白介素-33受体（ST2）及白介素-13基因敲除小鼠，经l635处理急性壁细胞缺失后对上皮细胞化生过程的影响。结果胃部上皮化生相关性巨噬细胞中可鉴定出M2极化亚群。高级胃部SPEM小鼠巨噬细胞中自介素-33的表达显著上调。l635可在野生型小鼠胃组织中诱导上皮化生，但在白介素-33及白介素．33受体敲除鼠中未观察到这一现象。尽管在诱导上皮化生过程中不需白介素．5及白介素-9，白介素-13基因敲除小鼠无法在l635诱导下发生上皮化生。对白介素-33受体基因敲除小鼠应用白介素-13，可重新诱导急性壁细胞缺失后的上皮化生。结论壁细胞缺失后上皮化生及巨噬细胞极化过程通过白介素．33及白介素-13的细胞信号网络协调进行，将损伤应答过程中固有黏膜机制及M2浸润联系起来。

2012年7月21日-2012年9月8日,在中国第五次北极科学考察期间布放并回收1套潜标观测系统,进行了楚科奇海中部(69o30′9″N,169o00′39″E)海洋多层温度、盐度、深度、海流等要素的定点观测,获得约50天的连续观测数据。数据集经过整理,最终成果由一个观测点地理位置数据和7个表格数据组成。这7个表格数据包括:(1)Nortek Aquad"小阔龙"海流计于14 m深处测量的海流数据,共2,431组。每组数据包含u和v两个变量,分别代表海流的东分量和北分量。(2)RBR CT于18 m深处测量的温度和盐度数据,共69,740组。每组包含测量时间、温度和盐度三个变量。(3)alec TD于19 m深处测量的温度数据,共139,000组。每组数据包含测量时间和温度两个变量。(4)alecCT于25m深处测量的温度和盐度数据,共139,575组。每组数据包含时间、温度和盐度三个变量。(5)alec CT于35 m深处测量的温度和盐度数据,共139,575组。每组数据包含时间、温度和盐度三个变量。(6)RBR CTD于40 m深处测量的温度和盐度数据,共279,140组。每组数据包含时间、温度和盐度三个变量。(7)TRDIADCP于41m深处向上观测的海流剖面数据。每组数据包含u和v两个变量,分别代表海流的东分量和北分量。每个变量分为17层,每层3,499组数据。数据集以.kmz,.shp和.xlsx数据格式存储,压缩后数据量为28.6 MB。
