MXenes,a newly emerging class of layered two dimensional(2D)materials,are promising solid lubricants due to their 2D structure consisting of weakly-bonded layers with a low shear strength and ability to form beneficial *** work aims at evaluating for the first time MXenes lubrication performance and tribofilm formation ability on different metallic substrates(mirror-lapped Fe and Cu discs).After depositing MXenes via ethanol(1 wt%)on the substrates,pronounced differences in the resulting substrate-dependent frictional evolution are *** MXenes are capable to reduce friction for both substrates after the full evaporation of ethanol,MXenes lubricating effect on Cu is long-lasting,with a 35-fold increased lifetime compared to *** spectra acquired in the wear-tracks of the substrates and counter-bodies reveal notable differences in the friction-induced chemical changes depending on the substrate *** case of Fe,the progressive failure of MXenes lubrication generates different Fe oxides on both the substrate and the ball,resulting in continuously increasing friction and a poor lubrication *** Cu,sliding induces the formation of a Ti_(3)C_(2)-based tribofilm on both rubbing surfaces,enabling a long-lasting lubricating *** work boosts further experimental and theoretical work on MXenes involved tribo-chemical processes.

Subcellular localization of alpha-synuclein aggregates and their interaction with membranes

For more than a decade numerous evidence has been reported on the mechanisms of toxicity of α-synuclein（αS） oligomers and aggregates in α-*** species were thought to form freely in the cytoplasm but recent reports of αS multimer conformations when bound to synaptic vesicles in physiological conditions,have raised the question about where αS aggregation *** this review we focus on recent literature regarding the impact on membrane binding and subcellular localization of αS toxic species to understand how regular cellular function of αS contributes to *** αS has been reported to mainly associate with specific membranes in neurons such as those of synaptic vesicles,ER/Golgi and the mitochondria,while toxic species of αS have been shown to inhibit,among others,neurotransmission,protein trafficking and mitochondrial *** interfering with αS membrane binding have shown to improve αS-driven toxicity in worms and in ***,a selective membrane binding that would result in a specific subcellular localization could be the key to understand how aggregation and pathology evolves,pointing out to αS functions that are primarily affected before onset of irreversible damage.

Thyroid carcinoma showing thymus-like differentiation: Case presentation of a young man

Ectopic thymic tissue can be present in the thyroid gland and a carcinoma showing thymus-like differentiation(CASTLE) may arise from such tissue. We are reported the case of a 26-year-old man with CASTLE, with cervical subcutaneous nodules relapse, who showed a good response to treatment with surgery, chemotherapy and radiotherapy. The problematic aspect of this case was the diagnosis; only on review were we able to make a final diagnosis. CASTLE is a very rare neoplasm. It is important to differentiate this cancer from others tumors such as primary or metastatic squamous cell carcinoma of the head and neck or squamous cell thyroid carcinoma, because the therapy and prognosis are *** is complicated and requires careful histological analysis(CD5- and P63-positive with presence of Hassall's corpuscles); unfortunately there is no gold standard treatment so, in this case, we administered a sandwich of chemotherapy and radiotherapy.

Evaluation of dietary addition of 2 essential oils from Achillea moschata,or their components (bornyl acetate,camphor,and eucalyptol) on in vitro ruminal fermentation and microbial community composition

This study investigated the effects of 2 Achillea moschata essential oils extracted from plants collected in 2 different valleys of the Italian Alps and 3 pure compounds of oils—bornyl acetate(BOR),camphor(CAM),and eucalyptol(EUCA)—on in vitro ruminal fermentation and *** in vitro batch fermentation experiment(Exp.1)tested the addition of all of the substances(2 essential oils and 3 compounds)in fermentation bottles(120 mL)at 48 h of incubation,whereas a subsequent in vitro continuous culture experiment(Exp.2)evaluated the pure compounds added to the fermenters(2 L)for a longer incubation period(9 d).In both experiments,total mixed rations were incubated with the additives,and samples without additives were included as the control(CTR).Each treatment was tested in duplicate and was repeated in 3 and 2 fermentation runs in Exp.1 and 2,*** production(GP)in Exp.1 was similar for all of the treatments,and short chain volatile fatty acid(SCFA)production was similar in both experiments except for a decrease of SCFA produced(P=0.029)due to EUCA addition in *** to CTR,BOR and CAM reduced the valerate proportion(P=0.04)in Exp.1,and increased(P<0.01)the acetate proportion in *** treatments increased(P<0.01)total protozoa counts(+36.7%and+48.4%compared to CTR on average for Exp.1 and 2,respectively).In Exp.1,all of the treatments lowered the Bacteroidetes and Firmicutes and increased the Proteobacteria relative abundances(P<0.05),whereas in Exp.2,the EUCA addition increased(P=0.012)the *** Exp.1,methane(CH4)as a proportion of the GP was lowered(P=0.004)by the addition of CAM and EUCA compared to CTR,whereas in Exp.2,EUCA reduced the amount of stoichiometrically calculated CH4 compared to ***,essential oils extracted from *** and the pure compounds did not depress in vitro rumen fermentation,except for EUCA in *** both experiments,an increase of the protozoal population occurred for all the additives.

Involvement of CRF2 signaling in enterocyte differentiation

To determine the role of corticotropin releasing factor receptor (CRF2) in epithelial permeability and enterocyte cell *** this purpose, we used rat Sprague Dawley and various colon carcinoma cell lines (SW620, HCT8R, HT-29 and Caco-2 cell lines). Expression of CRF2 protein was analyzed by fluorescent immunolabeling in normal rat colon and then by western blot in dissociated colonic epithelial cells and in the lysates of colon carcinoma cell lines or during the early differentiation of HT-29 cells (ten first days). To assess the impact of CRF2 signaling on colonic cell differentiation, HT-29 and Caco-2 cells were exposed to Urocortin 3 recombinant proteins (Ucn3, 100 nmol/L). In some experiments, cells were pre-exposed to the astressin 2b (A2b) a CRF2 antagonist in order to inhibit the action of Ucn3. Intestinal cell differentiation was first analyzed by functional assays: the trans-cellular permeability and the para-cellular permeability were determined by Dextran-FITC intake and measure of the transepithelial electrical resistance respectively. Morphological modifications associated to epithelial dysfunction were analyzed by confocal microscopy after fluorescent labeling of actin (phaloidin-TRITC) and intercellular adhesion proteins such as E-cadherin, p120ctn, occludin and ZO-1. The establishment of mature adherens junctions (AJ) was monitored by following the distribution of AJ proteins in lipid raft fractions, after separation of cell lysates on sucrose gradients. Finally, the mRNA and the protein expression levels of characteristic markers of intestinal epithelial cell (IEC) differentiation such as the transcriptional factor krüppel-like factor 4 (KLF4) or the dipeptidyl peptidase IV (DPPIV) were performed by RT-PCR and western blot respectively. Entrapment neuropathies in diabetes mellitus

Neuropathy is a common complication of diabetes mellitus(DM) with a wide clinical spectrum that encompasses generalized to focal and multifocal forms. Entrapment neuropathies(EN), which are focal forms, are so frequent at any stage of the diabetic disease, that they may be considered a neurophysiological hallmarkof peripheral nerve involvement in DM. Indeed, EN may be the earliest neurophysiological abnormalities in DM,particularly in the upper limbs, even in the absence of a generalized polyneuropathy, or it may be superimposed on a generalized diabetic neuropathy. This remarkable frequency of EN in diabetes is underlain by a peculiar pathophysiological background. Due to the metabolic alterations consequent to abnormal glucose metabolism,the peripheral nerves show both functional impairment and structural changes, even in the preclinical stage,making them more prone to entrapment in anatomically constrained channels. This review discusses the most common and relevant EN encountered in diabetic patient in their epidemiological, pathophysiological and diagnostic features.

Serious drug-induced liver disease secondary to ezetimibe

Ezetimibe is the f irst member of a new family of lipid- lowering drugs that inhibits uptake of dietary and bili- ary cholesterol. It was approved by the FDA in 2002 for hypercholesterolemia alone or in combination with statins. Its use has been spreading over the last years. Ezetimibe was considered a safe drug. We report a case of a woman who developed a serious hepatocellular drug-induced liver disease after 4 mo therapy with 10 mg daily of ezetimibe. After withdrawal of the drug, the patient recovered slowly. Ezetimibe may produce seri- ous toxic hepatitis and prompt withdrawal is mandatory in case of a signif icant abnormality in liver testing after beginning or during treatment with ezetimibe.
