Interstitial lung disease in rheumatoid arthritis:Current concepts in pathogenesis,diagnosis and therapeutics

Rheumatoid arthritis(RA) is the most common chronic autoimmune inflammatory joint disease. RA-associated interstitial lung disease(RA-ILd) is a major extraarticular complication and causes symptoms that lead to a deterioration in the quality of life, high utilization of health resources, and an increased risk of earlier mortality. Early in the course of RA-ILd, symptoms are highly variable, making the diagnosis difficult. Therefore, a rational diagnostic strategy that combines an adequate clinical assessment with the appropriate use of clinical tests, including pulmonary function tests and high-resolution computed tomography, should be used. In special cases, lung biopsy or bronchioalveolar lavage should be performed to achieve an early diagnosis. Several distinct histopathological subtypes of RA-ILd are currently recognized. These subtypes also have different clinical presentations, which vary in therapeutic response and prognosis. This article reviews current evidence about the epidemiology of RA-ILd and discusses the varying prevalence rates observed in different studies. Additionally, aspects of RA-ILd pathogenesis, including the role of cytokines and other molecules such as autoantibodies, as well as the evidence linking several drugs used to treat RA with lung damage will be discussed. Some aspects of the clinical characteristics of RA-ILd are noted, and diagnostic strategies are reviewed. Finally, this article analyzes current treatments for RA-ILd, including immunosuppressive therapies and biologic agents, as well as other therapeutic modalities. The prognosis of this severe complication of RA is discussed. Additionally, this paper examines updated evidence from studies identifying an association between drugs used for the treatment of RA and the development of ILd.

Background:Thrombocytopenia is a possible risk factor for bleeding after band ligation of esophageal ***,elevated von Willebrand factor(VWF)in cirrhosis improves platelet function and could decrease this *** objective was to assess platelet function in patients with cirrhosis undergoing esophageal variceal ligation(EVL).Methods:The assessment consisted of platelet count,antigen and activity of VWF and VWF-cleaving protease AdAMTS-13 activity,and a platelet adhesion and aggregation test simulating vascular flow in vivo(Impact-RR)prior to ***:Totally 111 patients were divided into three groups according to platelet count:(1)d(3)>100×109/L(n=26,23.4%).No statistically significant difference was found in the aggregate size of platelets[group 1:41.0(31.8–67.3)μm 2;group 2:47.0(33.8–71.3)μm 2;and group 3:47.0(34.0–66.0)μm 2;P=0.60]and no significant correlation was found between aggregate size and platelet count(Spearman r=0.07;P=0.47).Surface coverage was 4.1%(2.8%–6.7%),8.5%(4.0%–10.0%),and 9.0%(7.1%–12.0%)(Pd 3,respectively and correlated with platelet count(Spearman r=0.39;Pdifference between groups in VWF or ***-EVL bleeding occurred in six(5.4%)patients(n=2 in group 1,n=1 in group 2,and n=3 in group 3;P=0.32).Patients with bleeding had higher MELd scores[15.0(11.3–20.3)versus 12.0(10.0–15.0);P=0.025],but no difference was demonstrated for platelet function ***:Platelet function is preserved even in the presence of thrombocytopenia,including in the patients with post-EVL bleeding.

Non-contact,self-referenced and near-infrared luminescent nanothermometers have been recognized as emerging tools in the fields of nanomedicine and nanotechnology due to their great capability of precise temperature readout at the nanoscale and real-time deep-tissue ***,the development of multifunctional and biocompatible luminescent nanothermometers operating within the optically transparent biological windows with high thermal sensitivity(>2.0%/K)remains ***,we present(Gd0.98Nd0.02)2O3 nanothermometers operated effectively within the first and second biological windows upon continuous-wave laser diode excitation at 808 *** thermometric parameters are defined by the relative changes in the emission intensities originating from the two Stark components of the 4 F3/2 level(R2 and R1)to the 4 I9/2(900-1000 nm),4 I11/2(1035-1155 nm)and 4 I13/2(1300-1450 nm)*** thermo metric parameters are evaluated for colloidal samples in a cell culture medium and powder samples,and the highest thermal sensitivity(2.18%/K at 298 K)is attained for the former in the first biological window(both the excitation and emission in the 800-965 nm range).The repeatability and temperature uncertainty are 99%and 1.2 K,*** nanothermometers are biocompatible with human MNT-1 melanoma and HaCaT cells for 24 h of exposure and nanoparticle concentration up to 0.400 mg/mL,showing their potential for applications in nanomedicine,e.g.,intracellular imaging and temperature mapping.

developing and validating a nestling photographic aging guide for cavity-nesting birds: an example with the European Bee-eater(Merops apiaster)

Background: Accurate estimation of nestlings’ age is essential in avian demography studies as well as in population ecology and conservation. For example, it can be useful for synchronizing nest visits with events of particular interest, such as the age at which young can be safely ringed, or in choosing the best period to attain the most accurate calculation of laying or hatching ***: We constructed a photographic guide for aging European Bee-eaters(Merops apiaster) nestlings to 3-day age classes and evaluated the aging method by performing a validation exercise with several observers with no previous experience in aging bee-eater ***: The aging guide for bee-eater nestlings allowed estimating age to within 3 days with an average accuracy of 0.85. We found the optimal period for aging nestlings was between days 13-18(with accuracy between 0.94 and 0.99), during which the status of feather development was more easily distinguishable from the preceding and subsequent age classes. during the first 3 days after hatching, nestlings could also be aged with high accuracy(0.93). The small size of the nestling in relation to the eggs and the nestling’s inability to raise its head during these first days allowed for good discrimination from the subsequent age class. Between days 25 and 28, nestlings were correctly aged in only half of assignments(0.55 sensitivity) and nestlings belonging to class 7(days 7-9) were the least correctly identified(0.38 sensitivity). However, by visiting the nests at 12 days intervals it is possible to achieve the highest accuracy in age estimation with the smallest disturbance and logistic ***: This study highlighted how indirect methods and a simple protocol can be established and employed to quickly estimate nestling age in cases where handling nestlings is challenging or impossible, while minimizing disturbance in and around the nest.

Pentoxifylline enhances the protective effects of hypertonic saline solution on liver ischemia reperfusion injury through inhibition of oxidative stress

BACKGROUNd：Liver ischemia reperfusion（IR）injury triggers a systemic inflammatory response and is the main cause of organ dysfunction and adverse postoperative outcomes after liver ***（PTX）and hypertonic saline solution（HTS）have been identified to have beneficial effects against IR *** study aimed to investigate if the addition of PTX to HTS is superior to HTS alone for the prevention of liver IR ***： Male Wistar rats were allocated into three groups. Control rats underwent 60 minutes of partial liver ischemia, HTS rats were treated with 0.4 mL/kg of intravenous 7.5% NaCl 15 minutes before reperfusion, and HPTX group were treated with 7.5% NaC1 plus 25 mg/kg of PTX 15 minutes before reperfusion. Samples were collected after reperfusion for determination of ALT, AST, TNF-α, IL-6, IL-10, mitochondrial respiration, lipid peroxidation, pulmonary permeability and myeloperoxidase. RESULTS： HPTX significantly decreased TNF-α 30 minutes after reperfusion. HPTX and HTS significantly decreased ALT,AST, IL-6, mitochondrial dysfunction and pulmonary myelo- peroxidase 4 hours after reperfusion. Compared with HTS only, HPTX significantly decreased hepatic oxidative stress 4 hours after reperfusion and pulmonary permeability 4 and 12 hours after reperfusion. CONCLUSION： This study showed that PTX added the beneficial effects of HTS on liver IR injury through decreases of hepatic oxidative stress and pulmonary permeability.

Mixed-Species Allometric Equations to Quantify Stem Volume and Tree Biomass in dry Afromontane Forest of Ethiopia

Volume and biomass equations are essential tools to determine forest productivity and enable forest managers to make informed decisions. However, volume and biomass estimation equations are scarce for Afromontane forests in Africa in general and Ethiopia in particular. This limits our knowledge of the standing volume of wood, biomass, and carbon stock of the forests therein. In this study, we developed a new mixed-species volume and biomass equations for Afromontane forests and compared them with generic pantropical and local models. A total of 193 sampled trees from seven dominant tree species were used to develop the equations. Various volume and biomass equations were fitted using robust linear and nonlinear regression. Model comparison indicated that the best model to estimate stem volume was ln(v)=-9.909+ 0.954*ln(d2h), whereas the best model to estimate biomass was ln(b)=-2.983+ 0.949*ln(ρd2h) . These equations explained over 85% of the variations in the stem volume and biomass measurements. The mean density and basal area of trees in the forest with d ≥ 2 cm was 631.5 stems·ha-1 and 24.4 m2·ha-1. Based on the newly developed equations, the forest has on average 303.0 m3·ha-1 standing volume of wood and 283.8 Mg·ha-1 biomass stock. The newly developed allometric equations derived from this study can be used to accurately determine the stem volume, biomass, and carbon storage in the Afromontane forests in Ethiopia and elsewhere with similar stand characteristics and ecological conditions. By contrast, the generic pan-tropical and other local models appear to provide biased estimates and are not suitable for dry Afromontane forests in Ethiopia. The estimated stem biomass and carbon stock in the Chilimo forest are comparable with the estimates from various tropical forests and woodlands elsewhere in Africa, indicating the importance of dry Afromontane fores

Molecular basis and management of gastrointestinal stromal tumors

Molecularly targeted agents have dramatically impacted the management of several cancers. Targeting KIT has led to a new treatment paradigm in gastrointestinal stromal tumors (GISTs). KIT is a cell surface receptor with tyrosine kinases that, upon binding of its ligand, stem cell factor, activates various signaling pathways. Imatinib and sunitinib, both tyrosine kinase inhibitors directed to KIT, were approved for firstand secondline treatment of metastatic and unresectable GISTs. In this article, we will review the molecular pathogenesis of GISTs followed by a discussion of imatinib and sunitinib’s role in the treatment of GISTs. Finally, we will introduce novel therapeutic options for imatiniband sunitinib-resistant GISTs.

Historical relationships of areas of endemism of the Brazilian Atlantic rain forest： a cladistic biogeographic analysis of harvestman taxa （Arachnida： Opiliones）

Based on a cladistic biogeographic analysis of 6 species-level phylogenies of harvestman taxa, wesearched for congruence in the historical relationships of 12 areas of endemism of the BrazilianAtlantic Rain Forest. We constructed general area cladograms using Primary Brooks ParsimonyAnaJysis （BPA）, BPA of nodes, and paralogy-free subtree analysis. These analyses resulted in 6general area cladograms, that allow to infer a general pattern of the relationships among areas ofendemism from the Brazilian Atlantic Rain Forest. Northern areas resulted related basally showingmain disjunctions at the doce River Valley and Todos os Santos Bay/S^o Francisco River *** remaining areas of endemism were included in a southern and a southeastern block, sepa-rated by the Ribeira do Iguape Valley. Incongruence Length differences tests showed no significantincongruence among the resulting cladograms and other matrix partitions. We concluded that tec-tonism and ancient marine transgressions were the probable processes responsible for the maindisjunctions, whereas Neogene refugia seem to have caused the more recent disjunctions. Thegeneral pattern and redundancy in area relationships suggest a model of main reiterative barriersin diversification at multiple times for the evolution of the Atlantic Rain Forest. The renewal of cla-distic biogeography and the search for common biogeographic patterns are discussed.

pysky: An Application for the Planning of multi-Target Astronomical Observations

To automate the process of planning and curating multi-target observation sessions, pysky application builds on the astroplan Python package to identify visible objects during an observation window and produce relevant information about those objects in visual and graphical form. The package calculates object visibility based on a provided time window and observing location as well as maximum airmass and limiting magnitude requested by the user. The pysky application images of the target objects with identifying and astrometric data to provide context for the images. In addition, pysky creates polar plots of each object’s horizontal coordinates, and the images and plots are designed to be shown side-by-side. The package also generates an HTML table of the selected target objects with their related data to relay the entire target list as one. The pysky application draws on a variety of Python packages to collect and process data from databases such as JPL Horizons and SIMBAd. Results for a test event were verified by hand using database web interfaces. The pysky application provides a platform for further integration of automated observation planning with websites and apps to enhance multi-target observation sessions.
