BACKGROUND Adipose-derived stem cells(ASCs)have been increasingly explored for cell-based medicine because of their numerous advantages in terms of easy availability,high proliferation rate,multipotent differentiation ability and low immunogenicity.In this respect,they have been widely investigated in the last two decades to develop therapeutic strategies for a variety of human pathologies including eye disease.In ocular diseases involving the retina,various cell types may be affected,such as Müller cells,astrocytes,photoreceptors and retinal pigment epithelium(RPE),which plays a fundamental role in the homeostasis of retinal tissue,by secreting a variety of growth factors that support retinal cells.AIM To test ASC neural differentiation using conditioned medium(CM)from an RPE cell line(ARPE-19).METHODS ASCs were isolated from adipose tissue,harvested from the subcutaneous region of healthy donors undergoing liposuction procedures.Four ASC culture conditions were investigated:ASCs cultured in basal Dulbecco's Modified Eagle Medium(DMEM);ASCs cultured in serum-free DMEM;ASCs cultured in serumfree DMEM/F12;and ASCs cultured in a CM from ARPE-19,a spontaneously arising cell line with a normal karyotype derived from a human RPE.Cell proliferation rate and viability were assessed by crystal violet and MTT assays at 1,4and 8 d of culture.At the same time points,ASC neural differentiation was evaluated by immunocytochemistry and western blot analysis for typical neuronal and glial markers:Nestin,neuronal specific enolase(NSE),protein gene product(PGP)9.5,and glial fibrillary acidic protein(GFAP).RESULTS Depending on the culture medium,ASC proliferation rate and viability showed some significant differences.Overall,less dense populations were observed in serum-free cultures,except for ASCs cultured in ARPE-19 serum-free CM.Moreover,a different cell morphology was seen in these cultures after 8 d of treatment,with more elongated cells,often showing cytoplasmic ramifications.Immunofluorescence results and western blot anal

Background: Currently, surgical resection represents the only curative treatment for pancreatic cancer(PC), however, the majority of tumors are no longer resectable by the time of diagnosis. The aim of this study was to describe time trends and distribution of pancreaticoduodenectomies(PDs) performed for treating PC in Brazil in recent years. Methods: Data were retrospectively obtained from Brazilian Health Public System(namely DATASUS) regarding hospitalizations for PC and PD in Brazil from January 2008 to December 2015. PC and PD rates and their mortalities were estimated from DATASUS hospitalizations and analyzed for age, gender and demographic characteristics. Results: A total of 2364 PDs were retrieved. Albeit PC incidence more than doubled, the number of PDs increased only 37%. Most PDs were performed in men(52.2%) and patients between 50 and 69 years old(59.5%). Patients not surgically treated and those 70 years or older had the highest in-hospital mortality rates. The most developed regions(Southeast and South) as well as large metropolitan integrated municipalities registered 76.2% and 54.8% of the procedures, respectively. LMIM PD mortality fluctuated, ranging from 13.6% in 2008 to 11.8% in 2015. Conclusions: This study suggests a trend towards regionalization and volume-outcome relationships for PD due to PC, as large metropolitan integrated municipalities registered most of the PDs and more stable mortality rates. The substantial differences between PD and PC increasing rates reveals a limiting step on the health system resoluteness. Reduction in the number of hospital beds and late access to hospitalization, despite improvement in diagnostic methods, could at least in part explain these findings.

The object of this study is the impact of technology in four exhibitions held to celebrate the 500 years of the death of Leonardo da Vinci on 2019.Two exhibitions took place in Milan,Italy,one took place in Rio de Janeiro,Brazil and the last one took place in São Paulo,also in Brazil.This study aims to explore the empirical evidences found during the field research in three of the four exhibitions object of this study(one of the exhibitions could not be observed to the museum being closed as a result of the pandemic caused by the coronavirus).This study is based on references and recent secondary researches that analyzed the aspects of quantity and quality in exhibitions sites,and also on other data available on websites and in the media.This study aims to question the importance of the original work in digital exhibitions and a possibly a further extension on the word“experience”not specifically through technological immersion.Lastly,these subjects are revisited once we look at the museums amid the world pandemic.

Psoriasis is a prolonged inflammation of the skin. The causes of psoriasis are still unclear. The treatment options depend on the severity of the disease and may include topical agents (such as topically-applied drugs, sulfur spa therapy, phototherapy) and systemic agents (orally or percutaneously administered). The aim of this study was to investigate the mechanical properties of the skin in psoriatic plaques before and after treatment with sulfur therapy (Thermae Luigiane, Guardia Piemontese—Acquappesa, Italy), in comparison with the skin of healthy subjects. The study has been performed on 20 psoriatic plaques (10 on upper arm, 10 on upper back) and 10 control subjects (healthy males aged 47 ± 15). The efficacy of sulfur therapy was observed through the evaluation of the biomechanical properties of the skin. Investigation was performed with a Skin meter, an instrument useful to determine the physiological parameters by the means of a suction method, a non-invasive in vivo suction. The apparatus was equipped with 2-mm measuring probe. Unlike the optical method, in our test the color and/or distribution of the skin micro-circulation didn’t interfere with the measurement. The evaluation of mechanical properties of the psoriatic plaques after treatment showed a significative recovery of the parameters analysed, with an increase of the elasticity parameters (Ur, Ua, Ua/Uf, Ur/Uf) and a decrease of the viscoelasticity module (Uv/Ue).

Protective Effect of <i>Aloe vera (Aloe barbadensis</i>Miller) on Erythrocytes Anion Transporter and Oxidative Change

The purpose of this study was to evaluate the ability of aqueous extract of Aloe barbadensis Miller (Aloe vera) on oxidative damage and Anion Exchanger 1 (AE1, also known as Band 3) expression in human erythrocytes exposed to the water soluble free radical initiator 2.2’-azobis-2-amidinopropano dihydrochloride (AAPH). In addition, total phenolic compounds in the extracts were determined as catechin equivalent and the various antioxidant activities were compared to natural and synthetic standard antioxidants such as BHA and ascorbic acid. Since Aloe vera extract did not cause a consumption of the cytosolic antioxidant, glutathione (GSH) when it was direct incubated with GSH in basic aerated aqueous solution, this indicates that Aloe vera extract does not proceed auto oxidation at this experimental condition. Furthermore, Aloe vera extract prevent the consumption of GSH, in radical treated RBCs. It also inhibit consumption of GSH when it was direct incubated with AAPH. Aloe vera gel extract inhibits the generation of diphenyl-2-picrylhy-drazyl (DPPH) and the scavenging activity was increased in a dose dependent manner. Aloe vera extract was shown the similar reducing power than standards BHT and ascorbic acid. Biochemical analysis by SDS-PAGE and western blotting showed that AAPH-induced oxidative stress increased the susceptibility of AE1 to proteolytic degradation. Of note, our data evidenced that Aloe vera treatment was able to partially restore the normal RBC membrane protein profiles in a dose-dependent manner. These results clearly demonstrate the antioxidative activity of Aloe vera gel extract that might be ascribed to a synergistic action of the bioactive compounds contained therein.

Rheumatoid arthritis is a chronic autoimmune inflammatory disease associated with increased cardiovascular risk and higher mortality in respect to general population.Beyond joint disease,inflammation is the major determinant of accelerated atherosclerosis observed in rheumatoid arthritis.We review the relationship between inflammation,atherosclerosis and cardiovascular risk in rheumatoid arthritis,focusing on the assessment of subclinical atherosclerosis by functional and morphological methods.These tools include flow mediated dilatation,carotid intima-media thickness,ankle/brachial index,coronary calcium content,pulse wave analysis and serum biomarker of subclinical atherosclerosis.

The prognosis for children with recurrent or refractory neuroblastoma remains a significant clinical challenge,and currently there are no known curative salvage regimens.In this paper we investigated the effect of imatinib with rapamycin and the chemotherapeutic agents temozolomide and irinotecan.We treated two children with recurrent neuroblastoma with this so called RIST protocol.Both patients,off therapy for 15 and 31 months,respectively are well,and developing normally,without any complications.These findings suggest that a combination regimen of RIST may provide a therapeutic benefit with a favorable toxicity profile to a unfortunate subset of patients with neuroblastoma.
