Identification of common and distinct origins of human serum and breastmilk IgA1 by mass spectrometry-based clonal profiling

The most abundant immunoglobulin present in the human body is IgA. It has the highest concentrations at the mucosal lining and in biofluids such as milk and is the second most abundant class of antibodies in serum. We assessed the structural diversity and clonal repertoire of IgA1-containing molecular assemblies longitudinally in human serum and milk from three donors using a mass spectrometry-based approach. IgA-containing molecules purified from serum or milk were assessed by the release and subsequent analysis of their Fab fragments. Our data revealed that serum IgA1 consists of two distinct structural populations, namely monomeric IgA1 (∼80%) and dimeric joining (j-) chain coupled IgA1 (∼20%). Also, we confirmed that IgA1 in milk is present solely as secretory (S)IgA, consisting of two (∼50%), three (∼33%) or four (∼17%) IgA1 molecules assembled with a j-chain and secretory component (SC). Interestingly, the serum and milk IgA1-Fab repertoires were distinct between monomeric, and j-chain coupled dimeric IgA1. The serum dimeric j-chain coupled IgA1 repertoire contained several abundant clones also observed in the milk IgA1 repertoire. The latter repertoire had little to no overlap with the serum monomeric IgA1 repertoire. This suggests that human IgA1s have (at least) two distinct origins;one of these produces dimeric j-chain coupled IgA1 molecules, shared in human serum and milk, and another produces monomeric IgA1 ending up exclusively in serum.

Determining minimum sample size for the conditioned Latin hypercube sampling algorithm

In digital soil mapping(DSM),a fundamental assumption is that the spatial variability of the target variable can be explained by the predictors or environmental *** to adequately sample the predictors have been well documented,with the conditioned Latin hypercube sampling(cLHS)algorithm receiving the most attention in the DSM *** advances in sampling design,a critical gap remains in determining the number of samples required for DSM *** propose a simple workflow and function coded in r language to determine the minimum sample size for the cLHS algorithm based on histograms of the predictor variables using the Freedman-Diaconis rule for determining optimal bin *** preprocessing was included to correct for multimodal and non-normally distributed data,as these can affect sample size determination from the *** on a user-selected quantile range(Qr)for the sample plan,the densities of the histogram bins at the upper and lower bounds of the Qr were used as a scaling factor to determine minimum sample *** technique was applied to a field-scale set of environmental covariates for a well-sampled agricultural study site near Guelph,Ontario,Canada,and tested across a range of *** results showed increasing minimum sample size with an increase in the Qr *** sample size increased from 44 to 83 when the Qr increased from 50% to 95% and then increased exponentially to 194 for the 99%*** technique provides an estimate of minimum sample size that can be used as an input to the cLHS algorithm.

Evaluating Methods of Visual Assistance for Workers to Improve Quality and Usability in Individualized Kitchen Cabinet Assembly

In times of digitalisation, visual assistance systems in assembly are increasingly important. The design of these assembly systems needs to be highly complex to meet the requirements. Due to the increasing number of variants in production processes, as well as shorter innovation and product life cycles, assistance systems should improve quality and reduce complexity of assembly processes. However, many large kitchen manufacturers still assemble kitchen cabinets manually, due to the high variety of components, such as rails and fittings. This paper focuses on the analysis and evaluation of virtual assistance systems to improve quality and usability in individualised kitchen cabinet assembly processes at a large German manufacturer. A solution is identified and detailed.

Academic productivity correlates with industry earnings in foot and ankle fellowship programs in the United States:A retrospective analysis

BACKGrOUND The study investigates the connection between academic productivity and industry earnings in foot and ankle orthopedic surgery *** metrics like the H-index and Open Payments Database(OPD)data,it addresses a gap in understanding the relationship between scholarly achievements and financial outcomes,providing a basis for further exploration in this specialized medical *** To elucidate the trends between academic productivity and industry earnings across foot and ankle orthopedic surgery fellowship programs in the United *** This study is a retrospective analysis of the relationship between academic productivity and industry earnings of foot and ankle orthopedic surgery fellowships at an individual faculty and fellowship *** productivity was defined via H-index and recorded from the Scopus *** earnings were recorded from the *** Forty-eight foot and ankle orthopedic surgery fellowships(100%of fellowships)in the United States with a combined total of 165 physicians(95.9%of physicians)were *** individual physician(n=165)total life-time earnings reported on the OPD website was United States Dollar(USD)451430.30±1851084.89(range:USD 25.16-21269249.85;median:USD 27839.80).Mean physician(n=165)H-index as reported on Scopus is 14.24±12.39(range:0-63;median:11).There was a significant but weak correlation between individual physician H-index and individual physician total life-time earnings(Prman’s rho=0.334)and a significant and moderate positive correlation between combined fellowship H-index and total life-time earnings per fellowship(P=0.004,Spearman’s rho=0.409).CONCLUSION There is a significant and positive correlation between academic productivity and industry earnings at foot and ankle orthopedic surgery fellowships in the United *** observation is true on an individual physician level as well as on a fellowship level.

recent Trends in Satellite-Derived Chlorophyll and Aerosol Optical Depth Together with Simulated Dimethylsulfide in the Eastern China Marginal Seas

The biogenic compound dimethylsulfide(DMS)produced by a range of marine biota is the major natural source of re-duced sulfur to the atmosphere and plays a major role in the formation and evolution of aerosols,potentially affecting *** spatio-temporal distribution of satellite-derived chlorophyll_a(CHL)and aerosol optical depth(AOD)for the recent years(2011-2019)in the Eastern China Marginal Seas(ECMS)(25°-40°N,120°-130°E)are *** seasonal CHL peaks occurred during late April and the CHL distribution displays a clear zonal *** CHL was also observed along the northern and western coastlines during summer and winter *** analysis shows that mean CHL decreases by about 10%over the 9-year study period,while AOD was higher in south and lower in north during summertime.A genetic algorithm technique is used to calibrate the key model parameters and simulations are carried out for 2015,a year when field data was *** simulation results show that DMS seawater concentration ranges from 1.56 to 5.88 nmol L^(−1) with a mean value of 2.76 nmol L^(−1).DMS sea-air flux ranges from 2.66 to 5.00mmol m^(−2) d^(−1) with mean of 3.80mmol m^(−2) d^(−1).Positive correlations of about 0.5 between CHL and AOD were found in the study region,with higher correlations along the coasts of jiangsu and Zhejiang *** elevated CHL concentration along the west coast is correlated with increased sea-water concentrations of DMS in the *** results suggest a possible influ-ence of DMS-derived aerosol in the local ECMS atmosphere,especially along the western coastline of ECMS.

We and others have published extensively on the benefits of physical activity(PA),exercise training,and cardiorespiratory fitness(CrF)to decrease the prevalence of cardiovascular diseases(CVDs)and CVD and all-cause morbidity and mortality.^(1-5)Both PA and CrF are important;however,CrF,which is mostly obtained through PA,although there is a genetic component,seems to be even more important than PA for predicting prognosis.^(1,4-6)Certainly,the wealth of data from across the world has indicated that increasing PA to enhance CrF while mitigating sedentary behavior would go a long way to lessen the world-wide obesity epidemic and reduce CVD and all-cause mortality.

Lithium-oxygen batteries are a promising technology because they can greatly surpass the energy density of lithium-ion ***,this theoretical characteristic has not yet been converted into a real device with high *** with air contamination,metallic lithium reactivity,and complex discharge and charge reactions are the main issues for this technology.A fast and reversible oxygen reduction reaction(Orr)is crucial for good performance of secondary batteries',but the partial knowledge of its mechanisms,especially when devices are concerned,hinders further *** this perspective,the present work uses operando raman experiments and electrochemical impedance spectroscopy(EIS)to assess the first stages of the discharge processes in porous carbon electrodes,following their changes cycle by cycle at initial operation.A growth kinetic formation of the discharge product signal(Li_(2)O_(2))was observed with operando raman,indicating a first-order reaction and enabling an analysis by a microkinetic *** solution mechanism in the evaluated system was ascribed for an equivalent circuit with three time *** the time constant for the anode interface reveals to remain relatively constant after the first discharge,its surface seemed to be more *** model indicated that the reaction occurs at the Li_(2)O_(2) surface,decreasing the associated resistance during the initial discharge ***,the growth of Li_(2)O_(2) forms a hetero-phase between Li_(2)O_(2)/electrolyte,while creating a more compact and homogeneous on the Li_(2)O_(2)/cathode *** methodology here described thus offers a way of directly probing changes in surface chemistry evolution during cycling from a device through EIS analysis.
