SP100 is an antiviral protein that restricts the productive stage of human papillomavirus(HPV)and multiple other viruses,and viruses in turn block sUMo-1-mediated stabilization of SP100 and promotes its degradation(Table S1).Interferon(IFN)signaling could still produce more SP100 through transcription to counteract viruses.1 Viruses also disable the transcriptional up-regulation of SP100 to achieve persistent infection in hosts.

Flavonoids, including fisetin, have been linked to a reduced risk of colorectal cancer(CrC) and have potential therapeutic applications for the condition. Fisetin, a natural flavonoid found in various fruits and vegetables, has shown promise in managing CrC due to its diverse biological activities. It has been found to influence key cell signaling pathways related to inflammation, angiogenesis, apoptosis, and transcription *** results of this study demonstrate that fisetin induces colon cancer cell apoptosis through multiple mechanisms. It impacts the p53 pathway, leading to increased levels of p53 and decreased levels of murine double minute 2, contributing to apoptosis induction. Fisetin also triggers the release of important components in the apoptotic process, such as second mitochondria-derived activator of caspase/direct inhibitor of apoptosis-binding protein with low pI and cytochrome c. Furthermore, fisetin inhibits the cyclooxygenase-2 and wingless-related integration site(Wnt)/epidermal growth factor receptor/nuclear factor kappa B signaling pathways, reducing Wnt target gene expression and hindering colony formation. It achieves this by regulating the activities of cyclin-dependent kinase 2 and cyclin-dependent kinase 4, reducing retinoblastoma protein phosphorylation, decreasing cyclin E levels, and increasing p21 levels, ultimately influencing E2 promoter binding factor 1 and cell division cycle 2(CDC2) protein levels. Additionally, fisetin exhibits various effects on CrC cells, including inhibiting the phosphorylation of Y-box binding protein 1 and ribosomal S6 kinase, promoting the phosphorylation of extracellular signal-regulated kinase 1/2, and disrupting the repair process of DNA double-strand breaks. Moreover, fisetin serves as an adjunct therapy for the prevention and treatment of phosphatidylinositol-4,5-bisphosphate 3-kinase catalytic subunit α(PIK3ca)-mutant CrC, resulting in a reduction in phosphatidylinositol-3 kinase(PI3K) expressio

The prostate is an accessory sex gland that develops under precise androgenic control. It is known that hormonal imbalance may disrupt its development predisposing this gland to develop diseases during aging. Although the hypothesis regarding earlier origins of prostate diseases was proposed many years ago, the mechanisms underlying this complex phenomenon are poorly understood. Therefore, the aim of this study was to evaluate the prostates of old male gerbils exposed to testosterone during intrauterine and postnatal life using morphological, biometrical, stereological, Kariometric, immunohistochemical, and immunofluorescence analyses. our findings demonstrate that prenatal and pubertal exposure to testosterone increases the susceptibility to the development of prostate diseases during aging. The presence of a more proliferative gland associated with foci of adenomatous hyperplasia in animals exposed to testosterone during the prenatal and pubertal phase show that the utero life and the pubertal period are important phases for prostatic morphophysiology establishment, which is a determinant for the health of the gland during aging. Therefore, these findings reinforce the idea that prostate disease may result from hormonal disruptions in early events during prostate development, which imprint permanently on the gland predisposing it to develop lesions in later stages of life.

目的基于血液筛查核酸检测反应性献血者的HBV感染的确认,探讨核酸检测反应性献血者的归队策略。方法联合应用自建的高灵敏度核酸检测体系、血液核酸筛查等多种核酸检测(NAT)方法,并结合血清学检测、献血者随访,对核酸检测反应性(NAT-yield)献血者中的HBV感染进行确认和感染状态识别。依据确认的HBV感染血浆样本,比较不同确认方法、确认指标或指标组合对HBV感染确认的效果。结果2010年11月—2021年2月,在血液筛查检出的876位NAT-yield献血者中共确认HBV感染者511人(oBI 451人,急性早期HBV感染者27人,不能确认感染者33人,无感染者30人,不能确认HBV感染者335人)。采用单检系统对混检系统检出的HBV感染血浆进行复测的检出率为96.6%,明显高于混检系统对单检系统检出的HBV DNA反应性(HBV DNA r)组和鉴别试验无反应性(NDr)组的复测检出率(76.4%和55.7%)(Pr样本在模式2(ID×5+鉴别×2)下复测检出率(65.2%)高于模式1(ID×2+鉴别×1)(39.2%)(Pr样本复测检出率无明显差异(P>0.05),但均明显高于NDr样本(PoBI献血者既往NAT数据,有46%经历多次NAT检测而未能检出。有59.1%oBI献血者随访检不出HBV DNA。oBI献血者中抗-HBc+占比为90.2%,单独抗-HBc+为49.2%,远高于不能确认感染组(PoBI和不能确认感染组中的比例极低且无差异(P>0.05)。结论近60%的NAT-yield献血者可以确认HBV感染。为保证献血者归队的安全性,需要更高灵敏度的HBV DNA确证技术提高HBV感染的确认率。抗-HBc是NAT-yield献血者oBI风险排查和归队评估最重要的血清学指标。

Background Extracellular vesicles(EVs) present in oviductal(oF) and uterine fluid(UF) have been shown to enhance bovine embryo quality during in vitro culture by reducing lipid contents and modulating lipid metabolism-related genes(LMGs), while also influencing cell proliferation, suggesting their involvement on the regulation of different biological pathways. The regulation of signaling pathways related to cell differentiation, proliferation, and metabo-lism is crucial for early embryo development and can determine the success or failure of the pregnancy. Bioactive molecules within EVs in maternal reproductive fluids, such as micro rNAs(mirNAs), may contribute to this regulatory process as they modulate gene expression through post-transcriptional *** This study evaluated mirNA cargo in oF-EVs from the early luteal phase and UF-EVs from the mid-luteal phase, coinciding with embryo transit within oviduct and uterus in vivo, and its possible influence on LMGs and sign-aling pathways crucial for early embryo development. A total of 333 mirNAs were detected, with 11 exclusive to oF, 59 to UF, and 263 were common between both groups. From the 20 differentially expressed mirNAs, 19 up-regulated in UF-EVs(bta-mir-134, bta-mir-151-3p, bta-mir-155, bta-mir-188, bta-mir-181b, bta-mir-181d, bta-mir-224, bta-mir-23b-3p, bta-mir-24-3p, bta-mir-27a-3p, bta-mir-29a, bta-mir-324, bta-mir-326, bta-mir-345-3p, bta-mir-410, bta-mir-652, bta-mir-677, bta-mir-873 and bta-mir-708) and one(bta-mir-148b) in oF-EVs. These mirNAs were predicted to modulate several pathways such as Wnt, Hippo, MAPK, and lipid metabolism and degradation. Clinical and Demographic Profile of Women with Uterine Fibroids Living in Puerto rico

Introduction: Uterine leiomyomas, or fibroids, are prevalent benign tumors affecting up to 70% of reproductive-age women in the United States, significantly impacting productivity and quality of life. Despite their commonality, the epidemiology and clinical patterns of leiomyomas in Puerto rico remain inadequately researched. Methods: This study conducted a secondary data analysis using clinical, menstrual cycle characteristics, and demographic data from self-administered questionnaires of women undergoing diagnostic laparoscopy for endometriosis in Puerto rico. Chi-square tests and t-tests analyzed categorical and continuous variables, respectively (p results: Among 1,610 endometriosis cases, 312 included uterine fibroids. Prevalence estimates were 10.3% (40 years). Primiparous women averaged 22.14 years, with 40.71% having 2 - 3 pregnancies and 26.28% experiencing miscarriages. Symptoms included infertility (29.5%), dyspareunia (40.7%), severe pain (50%), and dysmenorrhea (75.6%). Comorbidities included ovarian cysts (52.2%) and abnormal uterine bleeding (37.8%). Conclusions: Advanced age, parity, and comorbidities like diabetes and hypertension were prevalent in our cohort. Understanding the epidemiological profile of women living in Puerto rico with uterine fibroids is crucial for improving diagnostic awareness and enhancing patient care.
