Screening of.Environmental b.cterial Strains f.r Wild Aedes Mosquito Larval b.ocontrol

The ef.ectiveness of.current control measures against Aedes mosquitoes remains low, resulting in persistent epidemics in urb.n areas. The emergence of.resistant mosquito populations to chemical insecticides highlights the need f.r novel, environmentally f.iendly, cost-ef.ective control strategies. This study explored the potential of.environmental b.cterial isolates to b.ocontrol wild Aedes larvae. Initially, we collected b.cterial samples f.om inf.ctious masses of.Aedes f.uviatilis larvae. The isolated b.cteria were identif.ed using b.ochemical, enzymatic, and molecular methods, including 16S rRNA sequencing and MALDI-TOf. Previously, Aeromonas hydrophila and b.cillus thuringiensis isolated f.om these inf.ctious masses showed limited Aedes larval inhib.tion. Consequently, we screened additional environmental isolates f.om the b.cteriotheque. Six isolates previously identif.ed were tested: Chromob.cterium violaceum, Enterob.cter cloacae, b.cillus cereus, b.cillus sphaericus, and two strains of.b.cillus thuringiensis israelensis. Among these strains, b. thuringiensis and C. violaceum exhib.ted signif.cant inhib.tory activities against wild Aedes larvae. b.cillus thuringiensis cultures grown under daylight conditions showed a slight ab.lity to inhib.t Aedes larvae. The potential of.b. thuringiensis and C. violaceum strains studied, along with optimized culture growth conditions, will b. f.rther investigated to develop b.oinsecticide products to provide saf.r and more sustainab.e alternatives f.r controlling larvae of.Aedes mosquitoes.

Aminoglycosides (AGs) are a class of.antib.otics with a b.oad spectrum of.activity. However, their use is limited b. saf.ty concerns associated with nephrotoxicity and ototoxicity, as well as drug resistance. To address these issues, semi-synthetic approaches f.r modif.ing natural AGs have generated new generations of.AGs, however, with limited types of.modif.cation due to signif.cant challenges in synthesis. This study explores a novel approach that harness the b.cterial b.osynthetic machinery of.gentamicins and kanamycins to create hyb.id AGs. This was achieved b. glycodiversif.cation of.gentamicins via swapping the glycosyltransf.rase (GT) in their producer with the GT f.om kanamycins b.osynthetic pathway and resulted in the creation of.a series of.novel AGs, theref.re ref.rred to as genkamicins (GKs). The manipulation of.the hyb.id b.osynthetic pathway enab.ed the targeted accumulation of.dif.erent GK species and the isolation and characterization of.six GK components. These compounds display retained antimicrob.al activity against a panel of.World Health Organization (WHO) critical priority pathogens, and GK-C2a, in particular, demonstrates low ototoxicity compared to clinical drugs in zeb.af.sh emb.yos. This study provides a new strategy f.r diversif.ing the structure of.AGs and a potential avenue f.r developing less toxic AG drugs to comb.t inf.ctious diseases.

Electron Acceleration b. Microwave Radiation Inside a Rectangular Waveguide

Electron dynamics in the f.elds associated with a transverse magnetic （TM） wave propagating inside a rectangular waveguide is analytically studied. The relativistic momentum and energy equations f.r an electron are solved, which was injected initially along the propagation direction of.the microwave. Expressions of.the acceleration gradient and def.ection angle are ob.ained. In principle, it is shown that the electron can b. accelerated in this condition and there is no def.ection when the electron is injected f.om the centre of.the waveguide f.ont. However, it is f.und that the acceleration gradient and def.ection angle depend strongly on the parameters of.the microwave （intensity, f.equency, etc.） and the dimensions of.the waveguide.

We have recently proposed a new technique of.plasma tailoring b. laser-driven hydrodynamic shockwaves generated on b.th sides of.a gas jet[Marquès et al.,*** 28,023103(2021)].In a continuation of.this numerical work,we study experimentally the inf.uence of.the tailoring on proton acceleration driven b. a high-intensity picosecond laser in three cases:without tailoring,b. tailoring only the entrance side of.the picosecond laser,and b. tailoring b.th sides of.the gas *** tailoring,the acceleration is transverse to the laser axis,with a low-energy exponential spectrum,produced b. Coulomb.*** the f.ont side of.the gas jet is tailored,a f.rward acceleration appears,which is signif.cantly enhanced when b.th the f.ont and b.ck sides of.the plasma are *** f.rward acceleration produces higher-energy protons,with a peaked spectrum,and is in good agreement with the mechanism of.collisionless shock acceleration(CSA).The spatiotemporal evolution of.the plasma prof.le is characterized b. optical shadowgraphy of.a prob. *** ref.action and ab.orption of.this b.am are simulated b. post-processing 3D hydrodynamic simulations of.the plasma *** with the experimental results allows estimation of.the thickness and near-critical density of.the plasma slab.produced b. tailoring b.th sides of.the gas *** parameters are in good agreement with those required f.r CSA.

Exploring the role of.mononuclear phagocytes in the epididymis

The onslaught of.f.reign antigens carried b. spermatozoa into the epididymis, an organ that has not demonstrated immune privilege, a decade or more af.er the estab.ishment of.central immune tolerance presents a unique b.ological challenge. Historically, the physical conf.nement of.spermatozoa to the epididymal tub.le enf.rced b. a tightly interwoven wall of.epithelial cells was considered suf.icient enough to prevent cross talk b.tween gametes and the immune system and, ultimately, autoimmune destruction. The discovery of.an intricate arrangement of.mononuclear phagocytes （MPs） comprising dendritic cells and macrophages in the murine epididymis suggests that we may have underestimated the existence of.a sophisticated mucosal immune system in the posttesticular environment. This review consolidates our current knowledge of.the physiology of.MPs in the steady state epididymis and speculates on possib.e interactions b.tween auto-antigenic spermatozoa, pathogens and the immune system b. drawing on what is known ab.ut the immune system in the intestinal mucosa. Ultimately, f.rther investigation will provide valuab.e inf.rmation regarding the origins of.pathologies arising as a result of.autoimmune or inf.ammatory responses in the epididymis, including epididymitis and inf.rtility.

Zika virus (ZIKV), a mosquito-b.rne f.avivirus, has b.en associated with b.nign inf.ctions f.r decades. However, it has b.come a pub.ic health concern due to its association with severe f.tal and neurological complications. Although many ef.orts have b.en made to control ZIKV inf.ction, approved vaccines or antiviral drugs are still lacking. Consequently, the development of.new ef.ective anti-ZIKV agents is urgently needed. In this context, we investigated the antiviral potential of.pyrazolone derivatives against ZIKV replication using in silico and in vitro methods. The f.ur pyrazolone derivatives evaluated (1a, 1b. 1c, and 1d) inhib.ted over 50% of.ZIKV replication with low cytotoxicity. Among them, compound 1b.exhib.ted the most potent activity (EC50 = 4.3 μM) and the highest selectivity (SI = 342). Mechanism of.action studies indicated that these compounds act at early stages of.virus replication, and compound 1b.can also directly inactivate ZIKV particles. Molecular docking studies suggested that these compounds can b.nd to and b.ock the activity of.ZIKV NS5 methyltransf.rase. f.nally, pharmacokinetic and toxicological predictions have reinf.rced the saf.ty and drug-like prof.les of.these derivatives. In conclusion, the pyrazolone scaf.old proved to b. valuab.e f.r anti-ZIKV drug development, and the derivatives studied deserve f.rther investigation.

We use density f.nctional perturb.tion theory b.sed on the pseudo-potential to calculate the phonon spectrum, phonon density of.states, specif.c heat capacity and mechanical properties of.pristine and cob.lt doped （5,0） single wall carb.n nanotub. （CNT）. In the calculations, we consider one Co atom in the center of.the unit cell of.the tub. and it is shown that the pristine （5~0） CNT is nonmagnetic while the Co-doped tub. b.comes magnetic. Young＇s modulus f.r b.th systems is ab.ut 1 TPa （＇af.er Co-doping it goes slightly higher） and the Poisson ratio f.r the pristine tub. b.comes quite a b.t larger than the doped one. On the other hand, the calculated value of.radial b.eath mode f.r the pristine CNT is in good agreement with the experimental reports while af.er Co-doping it is increased. In addition, heat capacity of.the doped CNT is reduced, which leads to some important empirical applications.
