Approaches for representing hydro-mechanical coupling between sub-surface excavations and argillaceous porous media at the ventilation experiment, Mont Terri

At the Mont Terri Underground Research Laboratory （Switzerland）, a field-scale investigation has been conducted in order to investigate the hydro-mechanical and chemical perturbations induced in the argilla- ceous formation by forced ventilation through a tunnel. This experiment has been selected to be used for processing model development and validation in the international project DECOVALEX-2011. The con- ceptual and mathematical representation of the engineered void, which itself forms a major part of the experiment and is not simply a boundary condition, is the subject of this paper. A variety of approaches have been examined by the contributors to DECOVALEX and a summary of their findings is presented here. Two major aspects are discussed. Firstly, the approaches for the treatment of the surface condition at the porous media/tunnel interface are examined, with two equivalent but differing formulations successfully demonstrated. Secondly, approaches for representing the tunnel with associated air and water vapour movement, when coupled with the hydro-mechanical （HM） representation of the porous medium, are also examined. It is clearly demonstrated that, for the experimental conditions of the ventilation experiment （VE） that abstracted physical and empirical models of the tunnel, can be used successfully to represent the hydraulic behaviour of the tunnel and the hydraulic interaction between the tunnel and the surrounding rock mass.

Accounting for anisotropic effects in the prediction of the hydro-mechanical response of a ventilated tunnel in an argillaceous rock

In order to investigate the hydro-mechanical （HM） and chemical perturbations induced in an argillaceous formation by forced ventilation during the operational period of a nuclear waste repository, a specific experiment has been performed in a tunnel, at Mont Terri Underground Research Laboratory （URL） in Switzerland. This experiment has been selected in the international project DECOVALEX for model vali- dation and the numerical simulation of this ventilation experiment （VE） is the object of the present paper. Since the argillaceous rock exhibits anisotropic properties, particular attention is given to the evaluation of the effects of various anisotropic features on the predicted results. In situ measurements such as relative humidity （RH）, global water mass extracted, pore water pressure, water content, and relative displace- ments are compared to predictions using both isotropic and anisotropic parameters. Water permeability anisotropy is shown to be the most influencing parameter by far, whereas in situ stress anisotropy has an effect only during the excavation phase. The anisotropy for mechanical parameterization has also some influence, in particular through HM couplings. These HM couplings have the potential to be very significant in terms of providing confidence in describing the experimental observation, and should be considered for further investigation.

At Mont Terri Underground Research Laboratory （URL） Switzerland, a specific experiment has been per- formed in a tunnel, in order to investigate the hydro-mechano-chemical （HMC） perturbations induced in the argillaceous formation by forced ventilation. This experiment has been selected in the international project DECOVALEX to be used for process model development and validation. The numerical simula- tion of the geochemical response to the ventilation experiment （VE） is the object of the present paper, focusing on the transport of chloride as a conservative species and sulphate as a reactive species. Uti- lising the validated hydro-mechanical （HM） results from earlier steps of the DECOVALEX task, reactive and non-reactive transport models, incorporating the current understanding of the geochemistry at the site, were successfully constructed for the whole experimental period. The associated parametric and process uncertainty analyses clearly demonstrate that the basic HM understanding must be sound. How- ever, to demonstrate this degree of robustness, the explicit inclusion of process representations of water desaturation, liquid vaporisation, species exclusion porosity, and redox processes, is required.

Bacterial plant pathogens affect the locomotor behavior of the insect vector:a case study of Citrus volkameriana-Triozae erytreae-Candidatus Liberibacter asiaticus system

Plant pathogens can alter the behavior of their insect vectors as well as their survival and reproduction.The African psyllid,Trioza erytreae,is one of the vectors of Huanglongbing,a citrus disease caused mainly by"Candidatus Liberibacter asiaticus"(CLas).The purpose of this study was to characterize the effects of CLas on the psyllid,T.erytreae using Citrus volkamerina plants as the study system.The study focused more specifically on the CLas effects prior to and after its acquisition by the psyllid T.erytreae.Our results did not support the hypothesis that CLas effects psyllid probing behavior prior to acquisition;few differences were observed between uninfected T.erytrea feeding on CLas-infected versus control plants.On the other hand,compared to psyllids that had completed their development on control plants,the ones that had completed their development on a CLas-infected plant exhibited changes in their behavior(greater velocity),physiology(smaller mass)and biochemistry(lower water and lipid content).Altogether,our results confirm the existence of a marked postacquisition effect on the vector locomotor behavior and a minor preacquisition effect of CLas on the vector behavior,which can be partially explained by physiological and biochemical changes.

The analysis of bird ringing data often comes with some potential sources of error and bias,as ring wear and/or loss could affect mark-recapture analyses and produce erroneous estimates of survival.Furthermore,ring wear and loss rates may differ between and within species based on the habitat they use or the species’ life-history traits and behaviour as well as the type of the ring.In this study we use resighting data from a long-term double marking experiment to directly estimate the rate of colour-ring loss among different Dalmatian Pelican colonies over time,evaluate any possible factors that could contribute to differential ring loss and assess how it may bias the results of mark-resighting analyses.Based on 14,849 resightings from 1275 individuals and using multi-state continuous-time hidden Markov models(HMMs) we showed that probability of ring loss was markedly different among colonies,ranging from 0.10 to 0.42 within the first year of marking,whereas the cumulative probability of losing a ring after ten years ranged 0.64 to 0.99.These rates are among the highest estimated when compared to previous studies in waterbirds.Our approach assessing the intra-specific variance in ring loss provided several factors potentially involved,such as the use of glue and the fledgling age accuracy and we could further hypothesise the effect of environmental factors.Finally,our results showed that ring loss can be a significant challenge for the assessment of the species’ population dynamics using mark-recapture methods as survival was consistently underestimated when not accounting for ring loss and varied significantly among different colonies.

Efficient scar-free knock-ins of several kilobases in plants by engineered CRISPR-Cas endonucleases

In plants and mammals,non-homologous end-joining is the dominant pathway to repair DNA doublestrand breaks,making it challenging to generate knock-in events.In this study,we identified two groups of exonucleases from the herpes virus and the bacteriophage T7 families that conferred an up to 38-fold increase in homology-directed repair frequencies when fused to Cas9/Cas12a in a tobacco mosaic virus-based transient assay in Nicotiana benthamiana.We achieved precise and scar-free insertion of several kilobases of DNA both in transient and stable transformation systems.In Arabidopsis thaliana,fusion of Cas9 to a herpes virus family exonuclease led to 10-fold higher frequencies of knock-ins in the first generation of transformants.In addition,we demonstrated stable and heritable knock-ins in wheat in 1%of the primary transformants.Taken together,our results open perspectives for the routine production of heritable knock-in and gene replacement events in plants.

Mycorrhizal fungi mitigate nitrogen losses of an experimental grassland by facilitating plant uptake and soil microbial immobilization

Nitrogen(N)is one of the most limited nutrients of terrestrial ecosystems,whose losses are prevented in tightly coupled cycles in finely tuned systems.Global change-induced N enrichment through atmospheric deposition and application of vast amounts of fertilizer are now challenging the terrestrial N cycle.Arbuscular mycorrhizal fungi(AMF)are known drivers of plant-soil nutrient fluxes,but a comprehensive assessment of AMF involvement in N cycling under global change is still lacking.Here,we simulated N enrichment by fertilization(low/high)in experimental grassland microcosms under greenhouse conditions in the presence or absence of AMF and continuously monitored different N pathways over nine months.We found that high N enrichment by fertilization decreased the relative abundance of legumes and the plant species dominating the plant community changed from grasses to forbs in the presence of AMF,based on aboveground biomass.The presence of AMF always maintained plant N:phosphorus(P)ratios between 14 and 16,no matter how the soil N availability changed.Shifts in plant N:P ratios due to the increased plant N and P uptake might thus be a primary pathway of AMF altering plant community composition.Furthermore,we constructed a comprehensive picture of AMF’s role in N cycling,highlighting that AMF reduced N losses primarily by mitigating N leaching,while N_(2)O emissions played a marginal role.Arbuscular mycorrhizal fungi reduced N_(2)O emissions directly through the promotion of N_(2)O-consuming denitrifiers.The underlying mechanism for reducing N leaching is mainly the AMF-mediated improved nutrient uptake and AMF-associated microbial immobilization.Our results indicate that synergies between AMF and other soil microorganisms cannot be ignored in N cycling and that the integral role of AMF in N cycling terrestrial ecosystems can buffer the upcoming global changes.

Thoracic ultrasound has attracted much interest in detecting pleural effusion or pulmonary consolidation after cardiac surgery.In 2016,Trovato reported,in the World Journal of Cardiology,the interest of using,in addition to echocardiography,thoracic ultrasound.In this editorial,we highlight the value of assessing diaphragm function after cardiac surgery and interventional cardiology pro-cedures.Various factors are able to impair diaphragm function after such interventions.Diaphragm motion may be decreased by chest pain secondary to sternotomy,pleural effusion or impaired muscle function.Hemidiaphragmatic paralysis may be secondary to phrenic nerve damage complicating cardiac surgery or atrial fibrillation ablation.Diagnosis may be delayed.Indeed,respi-ratory troubles induced by diaphragm dysfunction are frequently attributed to pre-existing heart disease or pulmonary complications secondary to surgery.In addition,elevated hemidiaphragm secondary to diaphragm dysfunction is sometimes not observed on chest X-ray performed in supine position in the intensive care unit.Analysis of diaphragm function by ultrasound during the recovery period appears essential.Both hemidiaphragms can be studied by two complementary ultrasound methods.The mobility of each hemidiaphragms is measured by M-mode ultrasonography.In addition,recording the percentage of inspiratory thickening provides important information about the quality of muscle function.These two approaches make it possible to detect hemidiaphragm paralysis or dysfunction.Such a diagnosis is important because persistent diaphragm dysfunction after cardiac surgery has been shown to be associated with adverse respiratory outcome.Early respiratory physio-therapy is able to improve respiratory function through strengthening of the inspiratory muscles i.***.diaphragm and accessory inspiratory muscles.

The hidden struggle of overworked urology residents“the hour”:The overlooked burnout,a narrative review

Burnout is a growing concern across numerous professions and industries worldwide.It has increased for many people since the onset of COVID-19 due to the stress,uncertainty,and changes brought on by the pandemic.Burnout is also a significant issue in the field of medicine,particularly urology,where residents face long working hours and high workloads,leading to physical and mental health problems,as well as limited opportunities for rest and rejuvenation.This alarming trend highlights the need for more effective measures to address burnout and promote employee well-being and job satisfaction.A thorough search of electronic databases such as PubMed,Scopus,and Google Scholar was conducted to obtain pertinent articles using relevant keywords.Retrieved articles were screened for relevance,with duplicates and irrelevant articles excluded.Thematic and critical review analysis was conducted on chosen articles,identifying key themes and contributing factors related to burnout in urology residents.The hidden struggle of overworked urology residents and their overlooked burnout represents a complex issue that requires systemic change and individual-level interventions.Organizations must prioritize employee well-being by providing adequate support and resources for managing burnout.Additionally,promoting effective leadership practices that prioritize resident well-being over any personal or institution recognitions or gains is crucial.These steps will improve the quality of care while mitigating negative outcomes for residents.

A novel responsive stabilizing Janus nanosilica as a nanoplugging agent in water-based drilling fluids for exploiting hostile shale environments

Thermo-responsive nanocomposites have recently emerged as potential nanoplugging agents for shale stabilization in high-temperature water-based drilling fluids(WBDFs). However, their inhibitory properties have not been very effective in high-temperature drilling operations. Thermo-responsive Janus nanocomposites are expected to strongly interact with clay particles from the inward hemisphere of nanomaterials, which drive the establishment of a tighter hydrophobic membrane over the shale surface at the outward hemisphere under geothermal conditions for shale stabilization. This work combines the synergistic benefits of thermo-responsive and zwitterionic nanomaterials to synchronously enhance the chemical inhibitions and plugging performances in shale under harsh conditions. A novel thermoresponsive Janus nanosilica(TRJS) exhibiting zwitterionic character was synthesized, characterized,and assessed as shale stabilizer for WBDFs at high temperatures. Compared to pristine nanosilica(Si NP)and symmetrical thermo-responsive nanosilica(TRS), TRJS exhibited anti-polyelectrolyte behaviour, in which electrolyte ions screened the electrostatic attraction between the charged particles, potentially stabilizing nanomaterial in hostile shaly environments(i.e., up to saturated brine or API brine). Macroscopically, TRJS exhibited higher chemical inhibition than Si NP and TRS in brine, prompting a better capability to control pressure penetration. TRJS adsorbed onto the clay surface via chemisorption and hydrogen bonding, and the interactions became substantial in brine, according to the results of electrophoretic mobility, surface wettability, and X-ray diffraction. Thus, contributing to the firm trapping of TRJS into the nanopore structure of the shale, triggering the formation of a tight hydrophobic membrane over the shale surface from the outward hemisphere. The addition of TRJS into WBDF had no deleterious effect on fluid properties after hot-treatment at 190℃, implying that TRJS could find potential use as a shale stabilizer
