The neutrophil–osteogenic cell axis promotes bone destruction in periodontitis

The immune-stromal cell interactions play a key role in health and diseases. In periodontitis, the most prevalent infectious disease in humans, immune cells accumulate in the oral mucosa and promote bone destruction by inducing receptor activator of nuclear factor-κB ligand (RANKL) expression in osteogenic cells such as osteoblasts and periodontal ligament cells. However, the detailed mechanism underlying immune–bone cell interactions in periodontitis is not fully understood. Here, we performed single-cell RNAsequencing analysis on mouse periodontal lesions and showed that neutrophil–osteogenic cell crosstalk is involved in periodontitis-induced bone loss. The periodontal lesions displayed marked infiltration of neutrophils, and in silico analyses suggested that the neutrophils interacted with osteogenic cells through cytokine production. Among the cytokines expressed in the periodontal neutrophils, oncostatin M (OSM) potently induced RANKL expression in the primary osteoblasts, and deletion of the OSM receptor in osteogenic cells significantly ameliorated periodontitis-induced bone loss. Epigenomic data analyses identified the OSM-regulated RANKL enhancer region in osteogenic cells, and mice lacking this enhancer showed decreased periodontal bone loss while maintaining physiological bone metabolism. These findings shed light on the role of neutrophils in bone regulation during bacterial infection, highlighting the novel mechanism underlying osteoimmune crosstalk.

Inflammatory pseudotumor of the liver and peripheral eosinophilia in autoimmune pancreatitis

AIM: Inflammatory pseudotumor (IPT) of the liver is a rare benign lesion, the etiology of which remains *** is not associated with any particular diseases apart from phlebitis and Crohn's ***: A middle-aged male with hepatic IPT and peripheral eosinophilia associated with autoimmune pancreatitis (AIP) was selected for this study and review of ***: A 59-year-old male was admitted with obstructive jaundice, marked eosinophilia (1 343/mm3)and hypergammaglobulinemia (4 145 mg/dL). Imaging techniques revealed dilatation of the intrahepatic bile duct,stenosis of the common bile duct with diffuse wall thickening, gallbladder wall thickening, irregular narrowing of the pancreatic duct, and swelling of the pancreatic parenchyma. Multiple liver masses were also demonstrated and diagnosed as IPT by biopsy specimens. Six months later, the abnormal features of the biliary tree remarkably improved by the oral administration of prednisolone, and the liver masses disappeared. The swelling of the pancreatic head also improved. The peripheral eosinophil count normalized. IPT associated with AIP, as we know,has not been reported in the literature. The clinical features of the present case mimicked those of pancreatic cancer with liver metastasis. This case deserves to be documented to prevent misdiagnosis of similar cases.

The ferrous [Fe（II）] and α-ketoglutarate（α-KG）-dependent dioxygenases can catalyze hydroxylation in proteins, nucleic acids, lipids and small molecules. These enzymes participate in a vast array of protein side-chain modifications, repairing of alkylated DNA/RNA, biosynthesis of antibiotics and plant products, and so on. Recently, the studies on dioxygenases for asymmetric synthesis are being increasingly targeted for therapeutic technologies. A branched-chain amino acid dioxygenase from Burkholderia ambifaria AMMD（SadA, 30.7 kDa） catalyzes the（S）-selective hydroxylation at C-3 position of N-substituted branched-chain amino acids to produce a chiral molecule, coupled with the conversion of α-ketoglutarate to succinate and CO2. To elucidate the structural basis of the stereoselective hydroxylation and substrate specificity, we determined the crystal structure of ***（II） and ***（II）.α-KG complex at 2.5 A and 2.0 A resolution, respectively. SadA was crystallized as a homodimer by a disulfide bond of Cys101-Cys101 and two salt bridges of Lys171-Asp87 and Asp105-Arg102. It possessed a double-stranded β-helix（DSBH） fold at the core of the structure. In addition, HXD/EXnH motif in the active site coordinated a Zn（II） molecule as an Fe（II） substitute. The α-KG molecule bounded to Zn（II） in a bidentate manner via its 1-carboxylate and 2-oxo groups. Based on the ***（II）.α-KG structure and mutation analyses, we constructed substrate-binding models with N-succinyl-L-leucine and N-succinyl-L-phenylalanine, which provided new insight into the substrate specificity. The results will be useful for the rational design of SadA variants aimed at the recognition of various N-succinyl L-amino acids.

发现一种简便快捷的从头发中提取蛋白质的方法,可以更方便地检测头发蛋白的生物化学性能。这种提取过程原理是在还原剂存在的条件下,硫脲和尿素的结合能有效地提取出头发皮层部分的蛋白质。提取出的蛋白质主要是由分子量为40～60 kDa的硬α-角蛋白、分子量为12～18 kDa的基质蛋白、以及少量的分子量为110～115 kDa和125～135 kDa的蛋白质。此法对羊毛、鸡毛、鼠毛和人类指甲的开发利用也非常有效。

Design Study of Top Lid with Clamp Structure in JT-60SA Cryostat

JT-60SA is a fully superconducting coil tokamak upgraded from the JT-60U. This paper focused on the integrity of the top lid of cryostat in JT-60SA. The design requirement for the cryostat in normal operations is to achieve vacuum insulation of 10-1 Pa, and the top flange of the top lid is lightly welded onto its body flange. The weld is tensile-loaded by bending deformation of the top lid due to vacuum pressure of external 0.1 MPa. This weld integrity is evaluated with tensile-load reduction, which results in clamp reinforcement. The structural integrity of the top lid is validated.

Glass melt inclusion in clinopyroxene from Linqu Cenozoic basalt,Shandong Province,China

Cenozoic basalts from the Linqu County, Shandong Province, China entrain some clinopy- roxene crystals, of which many contain abundant glass melt inclusions. These melt inclusions are ex- tremely irregular in shape with most grain sizes in a range of 10-50 μm and coexist with low-Mg# olivines, labradorites and Ca-rich potassium feldspars. In-situ major and trace element analyses show that the glass melt inclusions are high in alkalis (Na2O+K2O > 10 wt%), SiO2 (>54 wt%), CaO and FeO (>4 wt%), but low in MgO (Mg# 2), thus having phonoli- tic compositions. The compositional features of cli- nopyroxene crystals, glass melt inclusions and their coexistent minerals suggest that these melt inclu- sions were exotic melts in clinopyroxenes trapped prior to their entrainment in the host basalt. The dis- covery of these melt inclusions provides a new ap- proach to further investigating the evolution of Meso- zoic lithospheric mantle beneath the southeastern North China Craton.

Two-year clinical outcome of denosumab treatment alone and in combination with teriparatide in Japanese treatment-naive postmenopausal osteoporotic women

This randomized prospective study aimed to evaluate the clinical outcome of denosumab treatment alone and in combination with teriparatide in treatment-naive postmenopausal Japanese female patients with osteoporosis. Thirty patients were randomly assigned to two groups:(1) denosumab group(denosumab alone, n=13); and(2) combination group(denosumab+teriparatide, n=17). Serum bone-specific alkaline phosphatase(BAP), serum tartrate-resistant acid phosphatase(TRACP)-5b, urinary cross-linked N-terminal telopeptides of type I collagen(NTX), and bone mineral density(BMD) of L1–4 lumbar vertebrae(L-BMD)and bilateral total hips(H-BMD) were determined at the first visit and at various time points up to 24 months post-treatment to determine percentage changes. Serum TRACP-5b and urinary NTX were equally suppressed in both groups and maintained at low levels, with slight increases at 12, 18 and 24 months. BAP was significantly decreased in both groups from 4 to 24 months, with significant differences between the groups at 4, 8 and 15 months(P<0.05). L-BMD was significantly increased at most time points in both groups, with a significant difference between the combination group and denosumab group at 24 months(17.2% increase versus 9.6% increase; P<0.05). There was no significant difference in H-BMD between the two groups, although the levels tended to be higher in the combination group than in the denosumab group(9.5% increase versus 5.6% increase). These findings suggest that denosumab+teriparatide combination therapy may represent an important treatment for primary osteoporotic patients at high risk of vertebral fracture.

Caterpillars(Lepidoptera and Symphyta larvae)employ diverse visual defensive tactics,and effectiveness of such tactics may be highly dynamic across time due to seasonal changes in the predator assemblages and their ***,this has rarely been studied especially in tropical *** we assessed temporal changes in the defensive value of caterpillar color and shape,using six types of plasticine dummy caterpillars:three colors(green,black,and white)x two shapes(curled and straight).These dummy caterpillars were deployed five times over different seasons in tropical forests of Xishuangbanna(China)and,as a comparison,twice in a temperate forest of Hirosaki(Japan).The colors and shapes of dummy caterpillars simulate visual traits of black sawfly larvae which take the curled resting posture in tropical rainforests of Xishuangbanna,apparently masquerading excrements commonly found on plants,while in Hirosaki there is no black-curled sawfly larvae and few excrements on *** found no significant effects of caterpillar colors or shapes on predation in *** contrast,black and curled caterpillars received significantly lower predation by birds in Xishuangbanna constantly across ***,we were unable to provide evidence that the black-curled sawfly larvae are masquerading as *** of the dummy caterpillars also affected the predation by ants and parasitoid wasps at certain *** is the first report on ecological function of the curled posture of sawfly larvae,and we demonstrated the importance to assess the temporal dynamics of predation and effectiveness of defensive tactics in tropical forests.

Presence of columnar-lined esophagus is negatively associated with the presence of esophageal varices in Japanese alcoholic men

AIM To determine whether the presence of columnar-lined esophagus(CLE) is associated with the presence of esophageal varices(EVs) in male Japanese alcoholics. METHODS The subjects were 1614 Japanese alcohol-dependent men(≥ 40 years of age) who had undergone upper gastrointestinal endoscopic screening. Digitalized records of high-quality endoscopic images that included the squamocolumnar junction and esophagogastric junction were retrospectively jointly reviewed by four expert endoscopists for the purpose of diagnosing CLE. The authors investigated whether and to what extent there were associations between the presence of CLE and the presence of EVs, especially in the group with liver cirrhosis(LC).RESULTS CLE ≥ 5 mm in length was found in 355 subjects(≥ 30 mm in 6 of them), LC without EVs in 152 subjects, LC with EVs in 174 subjects, and EVs without LC in 6 subjects. Advanced EVs, i.e., nodular, large or coiled forms, red color sign, or post-treatment, were found in 88 subjects. The incidence of CLE ≥ 5 mm decreased in the following order(P < 0.0001): 23.3% in the group without EVs, 17.4% in the group with small and straight EVs, and 5.7% in the group with advanced EVs. The multivariate ORs(95%CI) for EVs and advanced EVs in the group with LC were lower when CLE ≥ 5 mm was present [0.46(0.23-0.93) and 0.24(0.08-0.74), respectively, vs 0-4 mm CLE]. CONCLUSION The presence of CLE in male Japanese alcoholics was negatively associated with the presence of EVs.
