Virus-induced gene silencing and virusinduced flowering in strawberry(Fragaria×ananassa)using apple latent spherical virus vectors

Apple latent spherical virus(ALSV)vector is a convenient alternative to genetic transformation in horticultural plants,especially in species recalcitrant to genetic ***,an RNA virus,can infect a wide variety of plant species including major horticultural plants without inducing ***,methodologies were developed for infection of ALSV vectors to strawberry seedlings and plantlets cultured in vitro.A seed-propagated F1 hybrid strawberry cultivar'Yotsuboshi'was aseptically grown on half-strength Murashige–Skoog medium for 1 month and true leaves were inoculated with an ALSV RNA preparation by particle *** vector infection rates varied from 58 to 100%according to the insertion sequences,in‘Yotsuboshi’***(‘Dover’)propagated in vitro could also be infected with ALSV vector at a similar infection *** virus-induced gene silencing(VIGS),we prepared an ALSV vector carrying a 201 nucleotide segment of the strawberry phytoene desaturase gene.‘Yotsuboshi’and‘Dover’plants infected by this vector generated completely white leaves at fifth or sixth true leaves and *** virus-induced flowering(VIF),we used an ALSV vector expressing the Arabidopsis thaliana flowering locus T *** seedlings infected by this vector started to flower from about 2 months post inoculation and bore fruits with viable *** ALSV vector was no longer detected in any of the seedlings from early-flowered ***,the ALSV vector may be beneficial for examination of gene functions by VIGS in strawberry,and VIF using ALSV vector constitutes an effective new plant breeding technique for the promotion of cross-breeding in strawberry.

Sclerosing epithelioid fibrosarcoma of the liver infiltrating the inferior vena cava

Sclerosing epithelioid fibrosarcoma （SEF） is a rare and distinct variant of fibrosarcoma, composed of epithelioid tumor cells arranged in strands, nests, cords, or sheets embedded within a sclerotic collagenous matrix. We report a 39-year-old man with SEF of the liver, which infiltrated the inferior vena cava （IVC）. The SEF of the liver was successfully resected, and the infiltrated IVC was also removed together with the liver tumor. Histopathological examination of the tumor showed typical histopathology of SEE Immunohistochemically, the tumor was positive for vimentin. Recurrence was noted 7 mo after surgery. After chemotherapy, the recurrent tumor was resected surgically, and histopathological examination showed similar findings to those of the primary tumor. To our knowledge, this is the first report of SEF of the liver with tumor invasion of the IVC.

Eosinophilic cholecystitis is an infrequent form of cholecystitis. The etiology of eosinophilic cholecystitis is still obscure, and it is sometimes accompanied with several complications, but a simultaneous onset with pericarditis is very rare. Early apoptosis and cell death induced by ATX-S10Na(Ⅱ)-mediated photodynamic therapy are Bax- and p53-dependent in human colon cancer cells

AIM： To investigate the roles of Bax and p53 proteins in photosensitivity of human colon cancer cells by using lysosome-localizing photosensitizer, ATX-S10Na （Ⅱ）. METHODS： HCT116 human colon cancer cells and Bax-null or p53-null isogenic derivatives were irradiated with a diode laser. Early apoptosis and cell death in response to photodynamic therapy were determined by MTT assays, annexin Ⅴ assays, transmission electron microscopy assays, caspase assays and western blotting. RESULTS： Induction of early apoptosis and cell death was Bax- and p53-dependent. Bax and p53 were required for caspase-dependent apoptosis. The levels of anti-apoptotic Bcl-2 family proteins, Bcl-2 and Bcl-xL, were decreased in Bax- and p53-independent manner. CONCLUSION： Our results indicate that eady apoptosis and cell death of human colon cancer cells induced by photodynamic therapy with lysosome-localizing photosensitizer ATX-S10Na （Ⅱ） are mediated by p53- Bax network and low levels of Bcl-2 and Bcl-xL proteins. Our results might help in formulating new therapeutic approaches in photodynamic therapy.

Development of novel rare earth doped fluoride and oxide scintillators for two-dimensional imaging

Two topics were focused. The first one was about the gamma-ray scintillator, Pr^3＋：Lu3Al5O12 （LuAG）. The second one was about neutron scintillator, Ce^3＋：^6LiCaAlF6 and Eu^2＋：^6LiCaAlF6 （^6LiCAF）. Those scintillators have been developed very recently for modem imaging applications in the medical and homeland security fields. In both cases, the rare earth ions are playing the crucial role as emission centers. Pr^3＋ in LuAG provided fast 5d→4f transition providing noticeably shorter decay time than that of Ce^3＋. Among several candidate hosts, LuAG showed the best performance. Bulk crystal growth, basic scintillation properties, two-dimensional gamma-ray imaging and positron emission mammography （PEM） application were demonstrated. Due to the international situation, the homeland security was compromized by illicit traffic of explosives, drugs, nuclear materials, etc. and the ways to its improvement became an important R＆D topic. For this purpose the Ce and Eu doped LiCAF appeared competitive candidates. Especially, when substitution of 3He neutron detectors was considered, the discrimination ability of gamma-ray from alpha-ray was important. Bulk crystal growth, basic scintillation properties and two-dimensional neutron imaging were demonstrated.

The Usefulness of Ryusei<sup>®</sup>Perfusion Balloon for Treating Acute Coronary Syndrome with Vulnerable Plaque

Background: Intracoronary thrombus followed by a rupture of unstable vulnerable plaque is a well-known cause of acute coronary syndrome (ACS). The no reflow/slow flow phenomenon is sometimes observed during a primary percutaneous coronary intervention (PCI) against ACS. It has already been shown that long inflation using a perfusion balloon (PB) is useful to remediate a coronary perforation. Thus, we investigated the usefulness of a PB for treating ACS. Methods: This study was a retrospective, single-center, observational study. One hundred-seven patients with ACS underwent PCI from January 2015 to December 2017 in our hospital. Fifty patients were treated by PB directly (PB group) and the remaining 57 patients were treated by another conventional balloon (C group). We used the Ryuseiò balloon (Kaneka, Japan) as a PB. The clinical outcome was the incidence of the no reflow or slow flow phenomenon, the incidence of using IABP. Results: One patient in the PB group demonstrated slow flow phenomenon temporarily, and the coronary flow was quickly restored by thromboaspiration. In contrast, nine patients in the C group had occurrences of no reflow/slow flow phenomenon. Although all patients in the C group required stenting, some patients (24%) of the PB group did not require stenting. Conclusion: We found that the use of PB had a favorable effect on the treatment of ACS. Some patients completed PCI without a need for stenting.

Transforming growth factor-β(TGF-β)and bone morphogenetic protein(BMP)play important roles in bone *** ubiquitination regulatory factors(Smurfs)regulate TGF-β/BMP signaling via ubiquitination,resulting in degradation of signaling molecules to prevent excessive activation of TGF-β/BMP *** Smurf2 has been shown to negatively regulate TGF-β/Smad signaling,its involvement in BMP/Smad signaling in bone metabolism has not been thoroughly *** the present study,we sought to evaluate the role of Smurf2 in BMP/Smad signaling in bone *** collagen sponges containing 3μg of recombinant human BMP2(rhBMP2)were implanted in the dorsal muscle pouches of wild type(WT)and Smurf2−/−*** rhBMP2-induced ectopic bone in Smurf2−/−mice showed greater bone mass,higher mineral apposition and bone formation rates,and greater osteoblast numbers than the ectopic bone in WT *** WT mice,the ectopic bone consisted of a thin discontinuous outer cortical shell and scant inner trabecular *** contrast,in Smurf2−/−mice,the induced bone consisted of a thick,continuous outer cortical shell and abundant inner trabecular ***,rhBMP2-stimulated bone marrow stromal cells(BMSCs)from Smurf2−/−mice showed increased osteogenic ***2 induced the ubiquitination of Smad1/***/Smad signaling was enhanced in Smurf2−/−BMSCs stimulated with rhBMP2,and the inhibition of BMP/Smad signaling suppressed osteogenic differentiation of these *** findings demonstrate that Smurf2 negatively regulates BMP/Smad signaling,thereby identifying a new regulatory mechanism in bone metabolism.

Chiral Polyamides Having L-Glutamyl Residue as a Component

Polyamides with chiral environment were obtained from aromatic diamine, 1,3-phenylenediamine (1,3-PDA) or 1,4- phenylenediamine (1,4-PDA), and N-α-benzoyl-L-glutamic acid (Benzoyl-L-Glu). The optical rotation ([α]D) for 1,3- PDA-Benzoyl-L-Glu was determined to be 3.7 deg cm2 g–1, while that for 1,4-PDA-Benzoyl-L-Glu to be 9.7 deg cm2 g–1. 1,3-PDA-Benzoyl-L-Glu showed adsorption selectivity toward D-Glu and its adsorption selectivity was determined to be 1.68. Contrary to this, 1,4-PDA-Benzoyl-L-Glu showed adsorption selectivity toward L-Glu and the adsorption selectivity toward L-Glu was determined to be 1.33. From those results, those two types of chiral polyamide are expected to applicable to chiral separation or chiral recognition.

Support Operator’s Fault Diagnosis of Complex Plant Systemthrough Multilevel Flow Model and Ecological Interface Design

The paper describes a new human-interface system design method by combining the conception of Multilevel Flow Model and Ecological Interface Design to support operators’ fault diagnosis in the complex plant system. Modern man-made systems are always achieving many complex automatic and intelligent tasks so that they are becoming more and more complex and can be hardly understood by operators, who should be the primary role in system operating. This situation presents a big challenge to the operating support system that it should present the complex system in a direct and clear way to operators to and make operators understand the internal interaction of the system especially in the abnormal status to ensure the operating safety. The Multilevel Flow Model based on the idea of ”Abstraction Hierarchy”, aiming at decompressing a system by means-end and part-whole way, can be used to represent a complex system in a standard way and perform intelligent operating tasks such as fault diagnosis and process control. Ecological Interface Design, which based on the human cognitive properties, can present the internal interaction of the system in a direct way. This paper combines this two interface design conceptions to achieve two aspects, intelligent fault diagnosis and direct presentation of causal relationship of operating parameters, to support operators’ fault diagnosis in complex plant system. The design method is applied to a PWR power plant in this paper as an application example.
